将来源于银白杨的异戊二烯合成酶基因按照大肠杆菌密码子偏爱性进行优化,克隆到表达载体pACYCDu-et-1上,在大肠杆菌BL21(DE3)中异源表达,采用镍柱亲和层析纯化重组蛋白并测定其异戊二烯合成酶活性,通过摇瓶发酵实验对重组菌产异戊二烯进行进一步研究。结果显示:银白杨异戊二烯合成酶在大肠杆菌中能够高效表达,经过镍柱纯化后,电泳检测到特异性表达条带;该重组异戊二烯合成酶能够催化异戊二烯的合成,重组菌的异戊二烯产量可达到60μg/L。  相似文献

甘蓝粉虱是一种危险性入侵害虫,中国大陆于2012年7月在北京市朝阳区首次发现,寄主植物为抱茎苦买菜;翌年8月份又在新疆乌鲁木齐和吐鲁番发现,寄主植物为开花生菜和抱茎苦买菜。文章描述了甘蓝粉虱的危害方式、寄主植物、分布范围、识别特征、天敌种类以及传播扩散方式,并分析了在我国大陆的发展趋势和需要密切关注的区域。  相似文献

为了明确叉角厉蝽 Eocanthecona furcellate (Wolff)对草地贪夜蛾 Spodoptera frugiperda (J.E.Smith)幼虫的控制作用,在实验室条件下测定了叉角厉蝽不同虫态对草地贪夜蛾不同龄期幼虫的捕食能力,研究了该蝽捕食功能反应、搜寻效应以及种内干扰作用。结果表明,叉角厉蝽3龄若虫、5龄若虫、成虫均可捕食草地贪夜蛾幼虫,总体呈现叉角厉蝽低龄若虫捕食草地贪夜蛾低龄幼虫的数量较多,高龄若虫和成虫捕食中间龄期幼虫数量较多的规律;叉角厉蝽3个虫态对草地贪夜蛾2~3龄幼虫的捕食功能反应均符合Holling II方程和HollingⅢ型功能反应新模型;5龄若虫对草地贪夜蛾2~3龄幼虫的捕食效能(a/Th)最强(214.0)、日最大捕食量(1/Th)最大(256.4头)。不同虫态叉角厉蝽的搜寻效应与草地贪夜蛾密度均呈负相关。建立了叉角厉蝽成虫密度、草地贪夜蛾4龄幼虫密度对捕食作用的干扰反应方程,干扰作用发生后该蝽的平均捕食量和捕食作用率均逐渐下降。本研究结果可为田间释放叉角厉蝽防治草地贪夜蛾提供依据。  相似文献

1. Maternal and offspring diet effects on life‐history traits of the bird cherry‐oat aphid Rhopalosiphum padi were tested on three wheat varieties. Using nine reciprocal combinations of wheat varieties, the effects of previous experience (maternal diet effect) on the aphid's response to resistant and susceptible varieties (offspring diet effect) were tested. Batis was susceptible, and Xiaoyan22 and Ww2730 were both resistant, but with different mechanisms. 2. Aphids produced the most alatae in the treatments with the most resistant maternal diet variety Xiaoyan22. The fecundity (F) and intrinsic rate of increase (rm) of these alatae were at their greatest in the most resistant offspring diet variety, but these traits were not influenced in the apterae. 3. There were significant interactions in the alatae production and apterae life‐history traits, such as rm, development time, weight gain, and mean relative growth rate, between the maternal and offspring diet varieties. The interactions in apterae responses between varieties, some of which were reciprocal, indicated phenotypic plasticity in these parthenogenetic aphids. 4. Rhopalosiphum padi produced more alatae on the most resistant variety; the alatae would disperse and were more fecund. The growth responses of the apterae showed phenotypic plasticity to the different combinations of maternal and offspring diet varieties. The phenotypic plasticity would allow R. padi to better utilise the variable environments represented by the small wheat plots of different varieties in China.  相似文献

Though the plasmonic property for a passive nanoparticle dimer has been studied widely, the performance of a nanoparticle dimer with gain material is still inexplicit to our knowledge. Therefore, in this paper, we focus on the plasmonic effect of a nanoshell dimer, with its core filled with different gain materials, under a polarized plane wave excitation using a three-dimensional finite difference time domain method. It is shown that the gain materials in the core of the nanoshell can compensate the intrinsic absorption of the metal shell, resulting in a local energy enhancement in the junction of the active nanoshell dimer. The physics is supported by the detailed energy distribution of the active nanoshell dimer in each geometry region. It is found that the plasmonic coupling between two active nanoshell particles is more compact than the case of passive ones. The influence of shell thickness on the interaction between two adjacent active nanoshells is also analyzed.  相似文献

The inconsistency problem in systematics refers in part to the fact that disparate taxa of identical Linnean rank are not necessarily similar or even readily comparable in any other specifiable biological feature. This shortcoming led to a ‘temporal banding’ proposal in which extant clades associated with particular taxonomic ranks would be standardized according to a universal metric: the absolute time of evolutionary origin. However, one underexplored possibility is that same‐level taxa in disparate organismal groups already might be similar (fortuitously so) in evolutionary age. In the present study, we explicitly address this possibility by reviewing published molecular inferences about the known or suspected origination dates of taxonomic genera, families, and orders in diverse organismal groups. Our findings empirically confirm that currently recognized taxa are far from temporally standardized, thereby adding support for the contention that this kind of taxonomic inconsistency should ultimately be rectified in our biological classifications. © 2011 The Linnean Society of London, Biological Journal of the Linnean Society, 2011, 102 , 707–714.  相似文献

Crop architecture parameters such as tiller number, angle and plant height are important agronomic traits that have been considered for breeding programmes. Auxin distribution within the plant has long been recognized to alter architecture. The rice (Oryza sativa L.) genome contains 12 putative PIN genes encoding auxin efflux transporters, including four PIN1 and one PIN2 genes. Here, we report that over-expression of OsPIN2 through a transgenic approach in rice (Japonica cv. Nipponbare) led to a shorter plant height, more tillers and a larger tiller angle when compared with wild type (WT). The expression patterns of the auxin reporter DR5::GUS and quantification of auxin distribution showed that OsPIN2 over-expression increased auxin transport from the shoot to the root-shoot junction, resulting in a non-tissue-specific accumulation of more free auxin at the root-shoot junction relative to WT. Over-expression of OsPIN2 enhanced auxin transport from shoots to roots, but did not alter the polar auxin pattern in the roots. Transgenic plants were less sensitive to N-1-naphthylphthalamic acid, an auxin transport inhibitor, than WT in their root growth. OsPIN2-over-expressing plants had suppressed the expression of a gravitropism-related gene OsLazy1 in the shoots, but unaltered expression of OsPIN1b and OsTAC1, which were reported as tiller angle controllers in rice. The data suggest that OsPIN2 has a distinct auxin-dependent regulation pathway together with OsPIN1b and OsTAC1 controlling rice shoot architecture. Altering OsPIN2 expression by genetic transformation can be directly used for modifying rice architecture.  相似文献

目的:以重组大肠杆菌YJM23为试材菌株,进行培养基优化提高异戊二烯产量。方法:利用Design expert软件中Placket-Burman设计法对影响供试菌株生产异戊二烯的8个因素进行了筛选;用最陡爬坡实验及基于中心组合设计的响应面分析法对三因素最佳水平及交互作用进行了研究。结果:确定了牛肉膏(P,0.0313)、微量元素混合液(P,0.0360)和K2HPO4.3H2O(P,0.0340)为影响摇瓶生产异戊二烯的主要因素;并确定这三种成分的最佳使用浓度:牛肉膏7.7 g/L,微量元素混合液0.1 ml/L,K2HPO4.3H2O 10.09 g/L,该三种成分的两两交互作用不显著。结论:优化条件下异戊二烯产量从140.16 mg/L提升至935.02 mg/L,提高6.67倍。  相似文献

目的:实现棘孢木霉菌T4的遗传转化并优化其转化体系.方法:以潮霉素抗性为选择标记,利用农杆菌转化法介导转化棘孢木霉菌.结果:潮霉素基因成功整合到受体菌基因组中,转化子抗性基因可稳定遗传.结论:最优的转化体系和条件为:IM和CM培养基中AS浓度为200 μg/mL,棘孢木霉T4孢子浓度为106/mL,农杆菌浓度为200 μL( OD600约0.8),共培养时间为48 h,转化效率约为50个转化子/106个孢子.  相似文献

几丁质酶作为木霉菌防治植物病虫害的主要因子,在生物防治和环境保护等领域发挥着重要的作用.为了研究棘孢木霉(Trichoderma asperellum)的生防机制并获得与其相关的功能基因,本研究通过RT-PCR、3′-RACE及5′-TAIL- PCR技术克隆了T. asperellum 1个几丁质酶基因Tachi1,对该基因进行了生物信息学分析,并利用毕赤酵母表达系统进行表达验证. Tachi1的DNA序列长1 635 bp,含有3个内含子,包含1 275 bp的开放阅读框,编码424个氨基酸;Tachi1属于糖基水解酶18家族内切几丁质酶,包含SIGGW底物结合位点和FDGIDXDWE活性中心位点,信号肽长度为22个氨基酸,成熟肽分子量为44 kD,二级结构以α-螺旋 、β-折叠和无规则卷曲为结构元件,三级结构为(α/β)8的圆桶形结构. 转Tachi1基因酵母工程菌可高效分泌表达几丁质酶Tachi1,甲醇诱导培养8 d几丁质酶酶活可达9.25 U/mL.  相似文献
