该文以青钱柳幼嫩茎段为外植体,研究其种苗快速繁殖技术。结果表明:青钱柳最佳采样时间为4月—6月,最佳外植体为轻微木质化茎段;最佳的外植体表面消毒方法为用0.1%HgCl2浸泡5~7 min,消毒成功率54.1%,无菌外植体存活率88.7%;初代芽诱导培养基为MS+6-BA 2.0 mg·L-1+IBA 0.2 mg·L-1+蔗糖30.0 g·L-1,芽诱导率80.5%,培养21 d初代芽苗平均高度3.0 cm;最佳继代增殖培养基为MS+6-BA 0.5mg·L-1+IBA 0.05 mg·L-1+TIBA 0.02 mg·L-1+蔗糖30.0 g·L-1,培养35 d,增殖系数7.0/35 d,平均苗高4.5cm,芽苗健壮且无玻璃化;生根前的壮苗培养基为MS+6-BA 0.5 mg·L-1+IBA 0.05 mg·L-1+蔗糖30.0 g·L-1,...  相似文献

Phylogenetic relationships within the palaeotropical tribe Lepisoroideae (Polypodiaceae) were investigated by studying sequence variation of four plastid DNA regions: rbcL, rps4 plus rps4‐trnS IGS, trnL intron plus trnL‐F IGS, rbcL‐atpB IGS plus part of atpB. In total, over 4000 nucleotides were sequenced for 39 species. Seven well‐supported clades were found in the analyses of the combined data set. We provide a new classification of Lepisoroideae by integrating phylogenetic results and known variation of morphological characters. The two small genera Neocheiropteris and Tricholepidium are supported as monophyletic, the genus Paragramma is resurrected and the genera Lepisorus, Neolepisorus, Lemmaphyllum and Lepidomicrosorium are re‐circumscribed. We proposed 14 new combinations, among which Caobangia is treated as a synonym of Lemmaphyllum. 水生植物的化感作用对淡水生态系统中水草的可持续管理和湖泊富营养化的生态控制具有非常重要的意义。本文综述了淡水水生植物化感作用的发展历史和研究现状,讨论了水生植物化感作用研究过程中所涉及的实验方法、生物测试方法、化感物质分离方法以及影响水生植物化感研究的环境因素。抑藻圈试验、藻类生长试验和浮萍生长试验是常用的生物测试方法;采用共生培养或单独培养的方法,从种植水中分离、鉴定化感物质,对证实复杂水环境中水生植物化感作用的存在,研究它们化感物质的释放途径和作用机理都具有十分重要的作用。  相似文献

用DME:Ham's F12(1∶1)培养液,添加3个水平的表皮生长因子和2个水平的胰岛素,组合成6种 培养体系(CS)分别培养大熊猫皮肤成纤维细胞。通过对细胞生长速度和染色体数目变异率进行测定,测得在 添加10μg/mL的胰岛素和40 ng/mL的表皮生长因子的培养体系(CS-5)中:以(1.673±0.185)×105/mL密 度接种细胞,经3.5 d,密度达到6.890×105/mL,其生长速度最快;染色体数目为二倍体细胞的比率75.77%; 核型分析显示,培养的细胞是大熊猫体细胞。综合衡量,CS-5更适合大熊猫皮肤成纤维细胞的培养。  相似文献

Anthropogenic soil erosion severely affects land ecosystems by reducing plant productivity and stimulating horizontal carbon and nitrogen movement at the surface. Climate warming may accelerate soil erosion by altering soil temperature, moisture, and vegetation coverage. However, no experiments have been carried out to quantify soil erosion with warming. In a long‐term field experiment, we explored how annual clipping for biofuel feedstock production and warming caused soil erosion and accompanying carbon and nitrogen losses in tallgrass prairie in Oklahoma, USA. We measured relative changes in soil surface elevation between clipped and unclipped plots with or without experimental warming. Our results show that average relative erosion depth caused by clipping was 1.65±0.09 and 0.54±0.08 mm yr?1, respectively, in warmed and control plots from November 21, 1999 to April 21, 2009. The soil erosion rate was 2148±121 g m?2 yr?1 in the warmed plots and 693±113 g m?2 yr?1 in the control plots. Soil organic carbon was lost at a rate of 69.6±5.6 g m?2 yr?1 in the warmed plots and 22.5±2.7 g m?2 yr?1 in the control plots. Total nitrogen was lost at a rate of 4.6±0.4 g m?2 yr?1 in the warmed plots and 1.4±0.1 g m?2 yr?2 in the control plots. The amount of carbon and nitrogen loss caused by clipping is equivalent to or even larger than changes caused by global change factors such as warming and rising atmospheric CO2 concentration. In addition, soil erosion rates were significantly correlated with clipping‐induced changes in soil moisture. Our results suggest that clipping for biofuel harvest results in significant soil erosion and accompanying losses of soil carbon and nitrogen, which is aggravated by warming.  相似文献

Piroplasmosis is a serious debilitating and sometimes fatal disease. Phylogenetic relationships within piroplasmida are complex and remain unclear. We compared the intron–exon structure and DNA sequences of the RPS8 gene from Babesia and Theileria spp. isolates in China. Similar to 18S rDNA, the 40S ribosomal protein S8 gene, RPS8, including both coding and non-coding regions is a useful and novel genetic marker for defining species boundaries and for inferring phylogenies because it tends to have little intra-specific variation but considerable inter-specific difference. However, more samples are needed to verify the usefulness of the RPS8 (coding and non-coding regions) gene as a marker for the phylogenetic position and detection of most Babesia and Theileria species, particularly for some closely related species.  相似文献

目的:实现棘孢木霉(Trichoderma asperellum)几丁质酶基因tachi2的原核高效表达,研究几丁质酶Tachi2的酶学性质.方法:利用PCR技术扩增得到几丁质酶基因tachi2,将其克隆到原核表达载体pEHISTEV中,测序后,转化大肠杆菌BL21感受态细胞,经异丙基硫代-β-D-半乳糖苷(IPTG)诱导后进行Tachi2蛋白的纯化和复性.用纯化的目的蛋白Tachi2进行几丁质酶酶学性质的研究.结果:tachi2基因在重组大肠杆菌中正确表达,其主要以包涵体形式存在;重组蛋白Tachi2分子量约为44kDa,经过纯化和复性后得到的Tachi2有较高的几丁质酶活性.该酶的最适温度为40℃,最适pH值为7.0,几丁质酶在40℃以下比较稳定、pH 6~9时酶有较高活性,受Cu2和Zn2+的强烈抑制.结论:成功实现了棘孢木霉几丁质酶基因tachi2的原核高效表达,表达纯化了重组蛋白,明确了几丁质酶Tachi2的酶学性质,为该几丁质酶的进一步开发利用和深入研究奠定了基础.  相似文献

【目的】外来物种在入侵地的扩张蔓延是其造成危害的关键。南水北调工程可为已入侵外来物种的进一步扩张蔓延提供通道和驱动力。风险评估可为预防入侵植物通过该途径的扩散蔓延提供依据。【方法】以国家重点管理外来入侵物种名录中的入侵植物为研究对象,根据生物入侵的发生过程(传入、定殖、扩散、危害)构建了包括4个层次29个指标的外来物种入侵综合风险评估的指标体系,规范了风险指数的计算方法,系统评估了24种入侵植物沿南水北调输水通道向北扩散而入侵京津冀受水区的风险。【结果】紫茎泽兰和互花米草的入侵风险值(R)相对较低(R0.6),其余22种入侵植物的风险值均较高(R0.6),其中,喜旱莲子草、凤眼莲和大薸3种外来水生植物的入侵风险等级最高(R0.8)。【结论】南水北调工程可能会促进入侵植物向北扩张蔓延并最终入侵京津冀受水区。建议尽早开展监测预警工作,以控制入侵物种随南水北调工程的扩张蔓延,进而阻止或降低其对京津冀尤其是华北最大水源地——白洋淀和雄安新区生态环境的威胁。  相似文献

In Arabidopsis thaliana, the expression of two genes encoding acyl‐CoA‐binding proteins (ACBPs) AtACBP1 and AtACBP4, were observed to be induced by lead [Pb(II)] in shoots and roots in qRT‐PCR analyses. Quantitative GUS (β‐glucuronidase) activity assays confirmed induction of AtACBP1pro::GUS by Pb(II). Electrophoretic mobility shift assays (EMSAs) revealed that Pas elements in the 5′‐flanking region of AtACBP1 were responsive to Pb(II) treatment. AtACBP1 and AtACBP4 were further compared in Pb(II) uptake using Brassica juncea, a potential candidate for phytoremediation given its rapid growth, large roots, high biomass and good capacity to accumulate heavy metals. Results from atomic absorption analyses on transgenic B. juncea expressing AtACBP1 or AtACBP4 indicated Pb(II) accumulation in roots. Subsequent Pb(II)‐tracing assays demonstrated Pb(II) accumulation in the cytosol of root tips and vascular tissues of transgenic B. juncea AtACBP1‐overexpressors (OXs) and AtACBP4‐OXs and transgenic Arabidopsis AtACBP1‐OXs. Transgenic Arabidopsis AtACBP1‐OXs sequestered Pb(II) in the trichomes and displayed tolerance to hydrogen peroxide (H2O2) treatment. In addition, AtACBP1 and AtACBP4 were H2O2‐induced in the roots of wild‐type Arabidopsis, while lipid hydroperoxide (LOOH) measurements of B. juncea AtACBP1‐OX and AtACBP4‐OX roots suggested that AtACBP1 and AtACBP4 can protect lipids against Pb(II)‐induced lipid peroxidation.  相似文献
