Co-C force constants from resonance Raman spectra of alkylcobalamins: insensitivity to dimethylbenzylimidazole coordination

The Co-C stretching vibration has been identified in resonance Raman spectra of alkyl-cobalamins, via isotope substitution, permitting estimation of the Co-C force constants, f = 1.85, 1.77 and 1.50 mdyn Å⁻¹ for methyl-, ethyl- and deoxyadenosyl-cobalamin, respectively (ν_Co-C = 506, 471 and 442/429 cm⁻¹). These values scale with the reported bond dissociation energies, and support the view that the Co-C bond weakens with increasing bulk of the alkyl group due to steric interaction with the corrin ring. However, the force constants are unaffected by dissociation of the dimethylbenzimidazole ligand at low pH, even though the bond dissociation energy rises significantly upon DMB dissociation in AdoCbl. This increase must therefore reflect destabilization of the Co^II product, rather than Co-C bond strengthening in the AdoCbl ground state. The insensitivity of the force constants to dimethylbenzimidazole dissociation implies that the steric effect of DMB coordination is not transmitted to the Co-C bond by the corrin ring. Consistent with this interpretation, the RR frequencies of the corrin ring modes are minimally perturbed by DMB dissociation, supporting earlier NMR results that indicated little change in the corrin conformation.     

Cloning and Bacterial Expression of Sesquiterpene Cyclase, a Key Branch Point Enzyme for the Synthesis of Sesquiterpenoid Phytoalexin Capsidiol in UV-Challenged Leaves of Capsicum annuum

Sesquiterpene cyclase, a branch point enzyme in the generalisoprenoid pathway for the synthesis of phytoalexin capsidiol,was induced in detached leaves of Capsicum annuum (pepper) byUV treatment. The inducibility of cyclase enzyme activitiesparalleled the absolute amount of cyclase protein(s) of pepperimmunodetected by monoclonal antibodies raised against tobaccosesquiterpene cyclase. A cDNA library was constructed with poly(A)⁺RNA isolated from 24 h UV-challenged leaves of pepper. A cDNAclone for sesquiterpene cyclase in pepper was isolated by usinga tobacco 5-epi aristolochene synthase gene as a hetero-logousprobe. The predicted protein encoded by this cDNA was comprisedof 559 amino acids and had a relative molecular mass of 65,095.The primary structural information from the cDNA clone revealedthat it shared 77%, 72% and 49% identity with 5-epi aristolochene,vetispiradiene, and cadinene synthase, respectively. The enzymaticproduct catalyzed by the cDNA clone in bacteria was identifiedas 5-epi aristolochene, as judged by argentation TLC. RNA blothybridization demonstrated the induction of an mRNA consistentwith the induction of cyclase enzyme activity in UV-treatedpepper. (Received March 2, 1998; Accepted June 15, 1998)     

Wastewater Treatment in a Packed-Bed Reactor with Immobilized Cells onto a New Ceramic Carrier

A new porous ceramic carrier based on casting sand, sawdust and zeolite was used to immobilize microorganisms for continuous wastewater treatment in a packed-bed bioreactor. Scanning electron microscopy showed that the ceramic carrier has a high porosity with diameters of around 5~300µm. Immobilization capacity of the carrier was 62 mg/ g dry carrier, in terms of mixed liquor volatile suspended solids. When synthetic wastewater (chemical oxygen demand 250 mg/l) as a model feed solution and 30 mg kaolin/l as a suspended solid (SS) were used, the removal efficiency as measured by the total organic carbon and SS were 91% and 71%, respectively, at a hydraulic retention time of 3 h.     

Inhibition stereochemistry of hydroxamate inhibitors for thermolysin

N-Acyl-N-hydroxy-β-amino acid derivatives were prepared and tested as inhibitors for thermolysin to find that these inhibitors show the -stereospecificity in contrast to the corresponding hydroxamates prepared from -amino acid, which exhibit the -stereochemistry. N-Formyl-N-hydroxy-β- -Phe-NHMe is the most potent inhibitor having the K_i value of 1.66 μM.     

Synthesis and biological activity of [Tic] didemnin B

A didemnin B analog containing a Tic (1,2,3,4-tetrahydroisoquinoline-3-carboxylic acid) as a conformationally restrained replacement for tyrosine has been synthesized and shown to have comparable potency as a protein biosynthesis inhibitor. Synthetic highlights include an oxidation of an alcohol to an acid in the presence of the sensitive Tic heterocycle and a modified Schmidt-type one-pot macrocyclization.     

具液晶性的天然高分子—N—邻苯二甲酰化壳聚糖的合成与表征

本文在不同条件下制备了一种液晶性天然高分子——N-邻苯二甲酰化壳聚糖,定性讨论了反应条件对壳聚糖的N-邻苯二甲酰化反应的影响。研究了N一邻苯二甲酰化壳聚糖的溶解性、成膜性、结晶性,并重点研究了其液晶性,测定出室温下它在二氯乙酸和二甲亚砜溶液中的临界浓度都为24wt％,在三氟乙酸溶液中的临界浓度为14wt％。     

米非司酮对大白鼠子宫内膜抗着床作用的组织化学观察

３０只雌性ＳＤ大鼠分为五组,即Ａ组（阴性对照组）,Ｂ组（孕三烯酮阳性对照组１ｍｇ／ｋｇ）,Ｃ组（米非司酮实验组,Ｃｌ１２ｍｇ／ｋｇ,Ｃ２６ｍｇ／ｋｇ,Ｃ３３ｍｇ／ｋｇ）。用组织化学方法观察米非司酮对子宫内膜一氧化氮合成酶（ＮＯＳ）、琥珀酸脱氢酶（ＳＤＨ）、乳酸脱氢酶（ＬＤＨ）、酸性磷酸酶（ＡＣＰ）、硷性磷酸酶（ＡＬＰ）活性和糖原含量的影响。实验结果显示：ＮＯＳ,ＳＤＨ和ＡＬＰ活性较阴性对照组弱,ＬＤＨ和ＡＣＰ活性较阴性对照组强,糖原含量略低于阴性对照组。     

天花粉蛋白Y14F/R22L定点突变及其活性研究

利用多聚酶链式反应（ＰＣＲ）技术,对天然天花粉蛋白（ｎＴＣＳ）基因在Ｔｙｒ１４和Ａｒｇ２２两个保守残基处同时进行定点突变,即Ｔｙｒ１４变成Ｐｈｅ,Ａｒｇ２２变成Ｌｅｕ,然后克隆到ｐＥＴ－８ｃ高效表达载体上,构建成重组质粒ｐＥＴＹ１４Ｆ／Ｒ２２Ｌ．经序列分析,定点突变的结果与预先设计的完全一致,突变后的天花粉蛋白命名为Ｙ１４Ｆ／Ｒ２２ＬＴＣＳ．将ｐＥＴＹ１４Ｆ／Ｒ２２Ｌ转化到Ｅ．ｃｏｌｉＢＬ２１（ＤＥ３,ｐＬｙｓＳ）中,进行表达．经ＣＭ－ＳｅｐｈａｒｏｓｅＣＬ－６Ｂ柱纯化,ＳＤＳ－ＰＡＧＥ鉴定,纯度可达９０％．ＲＩＰ活性测定显示,Ｙ１４Ｆ／Ｒ２２ＬＴＣＳ的活性比ｎＴＣＳ降低了７．５倍,活性变化不显著,因此,ＴＣＳ的Ｔｒｙ１４和Ａｒｇ２２对维持其活性部位构象并不是必需的．但由于Ｙ１４Ｆ／Ｒ２２ＬＴＣＳ在Ｅ．ｃｏｌｉ中的表达量与ｎＴＣＳ相比明显下降,因此,Ｔｙｒ１４和Ａｒｇ２２可能与ＴＣＳ翻译后的折叠有关．     

大鼠骨骼肌多催化功能蛋白酶的提取、鉴定及其抗血清的制备

为了研究多催化功能蛋白酶（ｍｕｌｔｉｃａｔａｌｙｔｉｃａｌｐｒｏｔｅｉｎａｓｅ,ＭＣＰ）在负氮平衡形成中的作用,以大鼠骨骼肌为原料,提取此酶并制备其抗血清．将大鼠骨骼肌粗提物经４５％～６５％饱和度硫酸铵分级盐析、阴离子交换层析和凝胶过滤,最后从Ｓｅｐｈａｒｏｓｅ４Ｂ层析柱上获得单一活性洗脱峰．酶活性用Ｃａｒｂｏｂｅｎｚｏｘｙ－Ｖａｌ－Ｇｌｙ－Ａｒｇ－４－ｎｉｔｒｉｎｉｌｉｄｅａｃｅｔａｔｅ作底物检测．非变性ＰＡＧＥ银盐染色显示单一区带的骨骼肌多催化功能蛋白酶,ＳＤＳ－ＰＡＧＥ银盐染色显示１０条亚基电泳区带,分子量在２５～３２ｋＤ之间．纯化的酶免疫兔８周后,抗血清效价达１３２,用分级盐析和离子交换层析纯化抗血清,显示单一电泳区带的ＩｇＧ．Ｗｅｓｔｅｒｎ－ｂｌｏｔ分析显示只在２５～３２ｋＤ之间出现多条亚基区带．这些结果提示已获得电泳纯ＭＣＰ及其较高特异性的多克隆抗体．
