Aqueous extract prepared from garlic bulbs markedly suppressed the mutagenesis in both E. coli WP2 trp- and E. coli WP2 trp- uvrA- induced by 4-nitroquinoline 1-oxide (4NQO), but not that induced by UV. Cellular toxicity, inhibition of the expression of the Trp+ phenotype and delay of the first cell division after 4NQO treatment were not observed in the presence of the extract. Since the extract showed identical antimutagenic effects against 4NQO in both test strains but no effect on the mutagenesis of UV, it seems that the extract might act by inactivating the electrophilic group(s) of 4NQO or inhibiting its metabolic activation.  相似文献

1. The present study was designed to explore the mechanisms by which insulin stimulates system A of amino acid transport in extensor digitorum longus (EDL) muscles, by using a system A analogue, alpha-(methyl)aminoisobutyric acid (MeAIB). 2. Insulin stimulation of MeAIB uptake was noted after only 30 min of incubation and was maximal at 60 min. Kinetics of the insulin effect on MeAIB uptake were characterized by an increased Vmax. without modification of Km for MeAIB. 3. Incubation of EDL muscles with cycloheximide for 90 min did not modify MeAIB uptake in either the presence or the absence of insulin, indicating the independence of insulin action from protein synthesis de novo. Incubations for 180 min with cycloheximide caused a decrease in basal MeAIB uptake; however, the percentage stimulation of amino acid transport by insulin was unaltered. Basal MeAIB uptake was increased by incubation for 180 min, but under these conditions no change in the percentage effect of insulin was found. 4. Ouabain, gramicidin D, or both, markedly decreased basal MeAIB uptake by EDL muscle, but the percentage effect of insulin was unaltered. 5. We conclude that insulin action on amino acid transport through system A in muscle is rapid, is characterized by an increased Vmax., and is independent of protein synthesis de novo and the Na+ electrochemical gradient. Our data are compatible with insulin acting directly on the system A transporter.  相似文献

Summary In the present study, the topographical distribution of carbohydrate binding sites on the plasma membrane of bovine epididymal spermatozoa was investigated using 15 different fluorescent neoglycoproteins and asialoglycoproteins. With mannose-bovine serum albumin (BSA)-fluoresceinthiocarbamyl (FTC), mannose-6-phosphate-BSA-FTC, lactose-BSA-FTC, maltose-BSA-FTC, asialolactoferrin-FTC and asialotransferrin-FTC a marked fluorescence was observed in the postacrosomal area. These results further substantiate the concept that carbohydrate binding sites of the spermatozoan plasma membrane and corresponding carbohydrates of the zona pellucida play a significant role in gamete interactions.Dedicated to Professor Dr. T.H. Membrane vesicles which constitute the sarcotubular system were separated and the fraction enriched in T-tubules purified by a calcium loading procedure. The preparations of unfractioned microsomes and T-tubules have been analyzed for their relative content of enzyme markers and acetylcholinesterase. The amount of this enzyme in the T-tubule fraction was higher than in mixed microsomes but less than two-fold the value of vesicles derived from sarcoplasmic reticulum. Arrhenius plots of membrane-bound and soluble acetylcholinesterase from either mixed microsomes or fractions enriched in T-tubules show an anomalous behaviour as two break points were obtained. The first discontinuity was found at about 17 degrees C for membrane-bound, and 12-14 degrees C for soluble acetylcholinesterase. The second one being at about 25 degrees C for both particulate and detergent-solubilized enzyme. The changes in activity with temperature suggest that lipid-protein, detergent-protein and protein-protein interactions might be involved in the stabilization of the enzyme both in the natural membrane and in the soluble state.  相似文献

Single-channel conductance fluctuations are analysed for gramicidin A incorporated into binary-mixed black lipid membranes of charged phosphatidic acid and neutral lecithin in different molar ratios. At very low Ca++ concentrations in the electrolyte (i.e. in the presence of EDTA) homogeneous lipid mixtures are identified through their conductance and life time probability distributions for integral gramicidin pores. As for the pure lipid components, the conductance histograms each show a single maximum with regular width and for all channels a single mean lifetime is found.For Ca++-levels (10-6–10-5 M) that are close to the critical demixing concentration (10-4 M) unusually broad conductance distributions and reduced lifetimes are found provided the PC content, x, of the membrane is close to the critical mixture (x crit0.5). We interpret this as a first example of the coupling of a membrane function (the transport of ions) to a lipid matrix with locally fluctuating composition close to a critical demixing point.For the conductance histogram of gramicidin A in an equimolar mixture of PA and PC shows two well-separated maxima. A correlation analysis between conductance and lifetime of the single pores shows that the two channel populations also differ significantly in their mean channel lifetime, *. Zusammenfassung Einleitend wird auf die vielfältigen, z.T. sich widersprechenden Resultate über die gefelderten und fibrillären Muskelfasern der Wirbeltiere hingewiesen.In den Befunden wird gezeigt, daß in den dünnen Muskelfasern des Zwerchfells der Ratte in I gelegene Mitochondrienroste durch longitudinale Verbindungen zu einem Gerüst verbunden sind. Die Mitochondrien der dicken Fasern beschränken sich auf unverbundene Netze in den isotropen Schichten der Sarkomere. Der prozentuale Anteil der Mitochondrien am Gesamtfaservolumen ist bei den dicken Fasern bis zur Hälfte geringer als bei den dünnen. Außerdem erhalten sie weniger Glykogen und kein Fett in Tropfenform.Anhand der Abstände der Myosinfäden wird der Mindestabstand der in Z quadratisch angeordneten Aktinfilamente berechnet.Die unterschiedlichen Formen des T-Systems in kontrahiertem und gedehntem Zustand der Faser sind Anlaß, die an Membranmodellen gewonnenen Vorstellungen über die Membraneigenschaften auf die Triaden zu übertragen.Die verschiedenen Anteile des interfibrillären Raumes werden zu den sie umgebenden Strukturen in funktionelle Beziehung gesetzt.
Summary Existing information on mammalian muscle fibers with Fibrillenstruktur and of those with Felderstruktur (Krüger) is in part contradictory. The present study reveals in the thin fibers (Felderstruktur) of the rat diaphragm, mitochondrial grids at the level of each I-disc. These are joined by means of longitudinal connections to form a continuous three-dimensional mitochondrial framework. In the thick fibers (Fibrillenstruktur) the mitochondria form only discontinuous nets or sieves in each isotropic layer of the sarcomere. In thick fibers the amount of mitochondrial material relative to total fiber volume is only about 1/2 of that in thin fibers. Furthermore, the thick fibers contain less glycogen and no lipid in the form of droplets. The T-systems differ in appearance in the contracted and in the expanded state of the fiber. This suggests the extension of the concepts on properties of membranes derived from membrane models to include the triads. The various components of the interfibrillar space are brought into functional relationship to the acjacent structures.

Zusammenfassung Zellen des aus unreifen Plasmoblasten bestehenden, transplantablen KG 13-Plasmocytoms des Goldhamsters zeigen bei phasenoptischer Lebendbeobachtung in vitro ein besonders großes, stark strukturiertes Golgifeld. Dieses wird von einem Saum heller Pinocytosetröpfchen gegen die Speichergranula abgesetzt. Die Form des Golgifeldes, die Gesetzmäßigkeit seiner Veränderungen, die Spezifität seiner Strukturen und seine Beziehung zum Zellkern werden beschrieben. Pinocytosebläschen wandern bei dieser Zellart im gesamten Verlauf sichtbar von der Zellmembran zwischen den Speichergranula hindurch zur Randzone des Golgifeldes und lösen sich dort auf. Die Granula des Cytoplasmas entstehen offenbar im Golgifeld und werden in seiner Randzone erstmalig als solche erkennbar, um ins Cytoplasma zu wandern.
Summary The KG 13 plasmocytoma from the Syrian golden hamster consists of immature Plasmoblasts. Living cells of this plasmocytoma observed by phase contrast microscopy in the hanging drop in vitro have a particularly large Golgi field surrounded by a border of light pinocytose bubbles. The cytoplasma is filled with storage granules. The form and the modifications of the Golgi field are described as well as his structure and his relations to the nucleus. Pinocytose bubbles reach the border of the Golgi field after crossing granules and disappeare. The granules are evidently formed in the Golgi field and become visible in its periphery.

Retinoic acid (RA) inhibited the in vitro growth of the mouse mast cell tumor line P815 in a dose- and time-dependent manner. The inhibition was accompanied by an increase in the amount of neutral intracellular mucopolysaccharides. Study of cell cycle kinetics showed that exposure to retinoic acid led to a slowing-down of the cell-cycle progression possibly related to a more differentiated cell population disclosed by microscopy with a lower proliferative capacity. In vivo, delays in both tumor appearance and mouse mortality were observed after injecting RA into mice bearing mastocytomas. These results suggest that RA could be of interest in the treatment of human malignant systemic mastocytosis with proliferation of immature mast cells.  相似文献
