猪瘟病毒反义基因在培养细胞上的效应研究——反义基因对猪瘟病毒的抑制作用

应用免疫荧光抗体技术检测了整合有猪瘟病毒反义基因的PK-15细胞克隆对猪瘟病毒的抑制效应。结果表明,五个不同的反义基因片段对猪瘟病毒的抑制效率存在很大差异,其中A片段的抑制效率最高(94％～98％),B片段次之(58％～76％),C片段再次(～64％),D片段和E片段未见明显的抑制效应。抑制效率的差异可能与反义基因片段的位置、长短以及反义RNA表达质粒载体的差异有关。     

Inhibition of rabbit sperm acrosomal enzymes by gossypol

The effect of gossypol on the activities of 10 acrosomal enzymes of the rabbit sperm was evaluated. Acrosin, Azocoll proteinase, neuraminidase, and arylsulfatase were significantly inhibited or completely inactivated by 12–76 μM gossypol. Hyaluronidase, β-glucuronidase, and acid phosphatase were inhibited only at a higher concentration of gossypol (380 μM). Phospholipase C, alkaline phosphatase, and β-N-Acetyl glucosaminidase were not inhibited even at 380 μM gossypol. Gossypol was found to be a noncompetitive inhibitor of arylsulfatase with a Ki of 120 μM. The inhibition was reversible and dose-dependent. As the acrosomal enzymes were more sensitive to the inhibition by gossypol compared to sperm enzymes involved in glycolysis or energy production, these assays may serve as a more reliable indicator for monitoring the occurence of gossypol-induced sterility. © 1995 Wiley-Liss, Inc.     

Use of paired plots and multivariate analysis for the determination of goat grazing preference

Abstract. Goat grazing is examined as a possible tool for grassland management. In this paper, goat grazing preferences are described. The proportion of biomass of particular species eaten by a goat was estimated on the basis of comparison of paired quadrats. The control quadrats were clipped before grazing and their biomass was compared with the biomass in grazed quadrats. Estimates of the proportion consumed were calculated for frequent species; they were significantly different among species; the most preferred species was Polygonum bistorta, a species with broad, soft, nutritious leaves. The least grazed species were Agrostis tenuis and Galium harcynicum. Various indices of food selectivity (e.g. forage ratio, electivity index) were calculated and their behaviour was compared. Canonical Correspondence Analysis and Redundancy Analysis with appropriate covariates reflecting the block design of the experiment were used to evaluate overall changes in species composition and yielded results similar to those from the direct estimation of the proportion consumed.     

Ovarian mesothelial and extramesothelial cells in interactive culture

Summary The ovarian mesothelium (OM) represents the tissue of origin of ovarian epithelial cancer. To gain insight into the regulation of this tissue, OM organoids and submesothelial ovarian stromal cells (SC) were isolated from New Zealand White rabbits by a stepwise tissue dispersal technique, while granulosa cells (GC) were aspirated from mature follicles (14±4 groups/animal). OM and SC dispersal were sequentially accomplished by: a) 1-h incubation in collagenase type I (300 U/ml), gentle scraping of the ovarian surface, and 1 g sedimentation of OM organoids (equivalent to 0.93±0.40 × 10⁶ cells/animal) on 5% bovine serum albumin (BSA); b) 2-h incubation in pronase-collagenase (0.5%–300 U/ml) under periodical resuspension and gentle scraping of SC (1.40±0.25 × 10⁶/animal) from OM-denuded ovaries. After a week-long in vitro expansion, OM cells (OMC) were cultured alone and with SC or GC within monocameral vessels or bicameral transfilter vessels in serumless, fibronectinrich (4μg/ml) HL-1 medium. After 7 d of contact cell-cell interaction, cytokeratin-positive OMC became surrounded by fibroblastoid, vimentin-positive SC or by cytokeratin and vimentin-weakly positive GC. Filter-bound OMC humorally interacting with underlying SC or GC displayed a biphasic, epithelioid and spindle, morphology with universal cytokeratin expression. Bromo-2′-deoxyuridine (BrdU) immunoperoxidase revealed mean cell proliferation indices of 14.88% for OMC cultured alone, 11.21% and 19.39% for OMC cultured with GC or SC in monocameral dishes, and 15.25% or 22.47% for OMC cultured in bicameral vessels over GC or SC, respectively. This model provides an experimental tool for investigating the unexplored role of stromal-mesothelial interaction in OM pathobiology.     

Expression of soybean nodulin 26 in transgenic tobacco. Targeting to the vacuolar membrane and effects on floral and seed development.

Nodulin 26 is an integral membrane protein of the symbiosome membrane of nitrogen-fixing soybean nodules. We expressed a nodulin 26 cDNA in transgenic tobacco (TN26 tobacco) under the control of the cauliflower mosaic virus 35S promoter to study subcellular targeting and the physiological effect(s) of its expression. Based on Northern and Western blots, the expression of nodulin 26 mRNA and protein in transgenic plants is high in apical shoot sections, flowers, and stems, low in mature leaves, and absent in roots. Western blot analysis revealed high levels of transgenic nodulin 26 protein in tonoplast membranes. In contrast, nodulin 26 protein was not found in isolated plasma membranes, the soluble fraction, nor in chloroplast and mitochondria-enriched membrane fractions. About 50-60% of the flowers and pods from TN26 tobacco plants abscised prematurely. Seed capsule size and seed fill per capsule from the remainder of surviving flowers were about 50% of that of control plants. Pollen viability was found to be normal, but flowers from TN26 tobacco plants showed shorter anther filaments compared with control plants. Normal seed production and capsule size was restored by manually crossing the stigmas from TN26 plants with isolated pollen from either transgenic or control plants. Thus, the aberrant filament growth could have resulted in the reproductive defects associated with the plants.     

Escherichia coli protein StpA stimulates self-splicing by promoting RNA assembly in vitro.

An Escherichia coli gene, stpA, has been identified and cloned based on its ability to suppress the Td- phenotype of a resident, splicing-defective phage T4 td (thymidylate synthase) gene. The stpA gene, which was localized to 60.24 min on the E. coli chromosome, encodes a 15.3-kDa protein. Overproduction of StpA in vivo led to an increase in td pre-mRNA levels and modest enhancement of td mRNA:pre-mRNA ratios. Consistent with its in vivo effect, purified StpA promoted RNA splicing in vitro, and facilitated RNA annealing and strand exchange with model substrates. These results suggest that StpA promotes splicing of the intron by binding RNA nonspecifically, resolving misfolded precursor molecules and facilitating association of critical base pair elements. Furthermore, proteinase K treatment of StpA-assembled precursors prior to the initiation of the splicing reaction still resulted in splicing enhancement, indicating that StpA is not required for the catalytic step, unlike the Neurospora splicing effector CYT-18, whose presence was necessary for catalysis to proceed. Together these results suggest that StpA has chaperone activity in vitro, with the property of promoting assembly of the precursors into an active conformation, in contrast to splicing effectors that stabilize the catalytically active intron structure.     

新疆六个民族指纹白线的调查研究The hand Print of Six Nationalities in Xinjiang

本文首次报道了新家6各民族6134人（南3181人、女2953人）的指纹白线出现频率。结果表明,白线频率女高于男,民族间亦有差异,其中维吾尔、哈萨克、鸟孜别克族频率接近。塔吉克族居住高寒高原地区,属白色人种,频率明显低于以上3个民族,锡伯族和汉族生活习惯近似,且高于以上4个民族,作者认为,白线出现频率的高低与种族、性别、年龄和群体生长环境及生活习惯有关。We’ve studied 6134 individuals (male 3181, fimale 2953) of six nationalities in Xinjiang.The result is that the white line of hand print is associated with rae,sex,age,grow environments of groups and life habbits.The ifference of races is:The appearant rates of white line of Weivuerite,Hasakese and Wuzibese are near,Tajike’s rate is lower than xiboman’s and han are higher than others.
