Cytological observations were made on developing seeds of soybean (Glycine max (L.) Merr. “Amsoy 71”) using scanning and transmission electron microscopy and light microscopy. Attention was focused on the maternal tissues of the seed coat and embryo sac. The ex situ population of the Przewalski's horse (Equus ferus przewalskii) is not self-sustaining (20% foaling rate), and the demography is skewed toward aging individuals with low gene diversity. We designed the present study to gain a better understanding of the reproductive biology of the Przewalski's mare and to determine whether age and gene diversity influenced reproductive function. Urine samples were collected 3-7 days/wk from 19 mares from May to September, and ultrasound examinations of follicular structures were performed 3 days/wk for 5 wk from May through July in nine individuals. A high proportion of mares exhibited abnormal (endocrine, 5 [26.3%] of 19; follicular, 2 [22.2%] of 9) or acyclic (endocrine, 4 [21.1%] of 19; follicular, 3 [33.3%] of 9) reproductive patterns. In four cyclic mares, estrous cycle length was 25.1 ± 1.2 days, with 12.2 ± 0.9 days of diestrus. Follicles in cyclic mares grew 1.2 ± 0.6 mm per day and ovulated after reaching 40.4 ± 8.9 mm. Mares with a high coefficient of inbreeding excreted reduced levels of mean urinary estrogens (r(2) = 0.476, P < 0.05), but age had no significant impact on reproductive patterns in this population. Overall, these data suggest that long-term genetic management of this population is necessary to maintain reproductive fitness.  相似文献

DNA damage activates checkpoint controls which block progression of cells through the division cycle. Several different checkpoints exist that control transit at different positions in the cell cycle. A role for checkpoint activation in providing resistance of cells to genotoxic anticancer therapy, including chemotherapy and ionizing radiation, is widely recognized. Although the core molecular functions that execute different damage activated checkpoints are known, the signals that control checkpoint activation are far from understood. We used a kinome-spanning RNA interference screen to delineate signalling required for radiation-mediated retinoblastoma protein activation, the recognized executor of G(1) checkpoint control. Our results corroborate the involvement of the p53 tumour suppressor (TP53) and its downstream targets p21(CIP1/WAF1) but infer lack of involvement of canonical double strand break (DSB) recognition known for its role in activating TP53 in damaged cells. Instead our results predict signalling involving the known TP53 phosphorylating kinase PRPK/TP53RK and the JNK/p38MAPK activating kinase STK4/MST1, both hitherto unrecognised for their contribution to DNA damage G1 checkpoint signalling. Our results further predict a network topology whereby induction of p21(CIP1/WAF1) is required but not sufficient to elicit checkpoint activation. Our experiments document a role of the kinases identified in radiation protection proposing their pharmacological inhibition as a potential strategy to increase radiation sensitivity in proliferating cancer cells.  相似文献

Interannual variability of cyanobacterial blooms in lake erie   总被引:3,自引:0,他引:3  
After a 20-year absence, severe cyanobacterial blooms have returned to Lake Erie in the last decade, in spite of negligible change in the annual load of total phosphorus (TP). Medium-spectral Resolution Imaging Spectrometer (MERIS) imagery was used to quantify intensity of the cyanobacterial bloom for each year from 2002 to 2011. The blooms peaked in August or later, yet correlate to discharge (Q) and TP loads only for March through June. The influence of the spring TP load appears to have started in the late 1990 s, after Dreissenid mussels colonized the lake, as hindcasts prior to 1998 are inconsistent with the observed blooms. The total spring Q or TP load appears sufficient to predict bloom magnitude, permitting a seasonal forecast prior to the start of the bloom.  相似文献

There is increasing evidence that hyperoxia, particularly at the time of birth, may result in neurological injury, in particular to the susceptible vasculature of these tissues. This study was aimed at determining whether overexpression of extracellular superoxide dismutase (EC-SOD) is protective against brain injury induced by hyperoxia. Transgenic (TG) mice (with an extra copy of the human extracellular superoxide dismutase gene) and wild-type (WT) neonate mice were exposed to hyperoxia (95% of F(i) o(2) ) for 7 days after birth versus the control group in room air. Brain positron emission tomography (PET) scanning with fludeoxyglucose (FDG) isotope uptake was performed after exposure. To assess apoptosis induced by hyperoxia exposure, caspase 3 ELISA and terminal deoxynucleotidyl transferase dUTP nick end labeling (TUNEL) staining were performed. Quantitative western blot for the following inflammatory markers was performed: glial fibrillary acidic protein, ionized calcium-binding adaptor molecule 1, macrophage-inhibiting factor, and phospho-AMP-activated protein kinase. PET scanning with FDG isotope uptake showed significantly higher uptake in the WT hyperoxia neonate brain group (0.14 ± 0.03) than in both the TG group (0.09 ± 0.01) and the control group (0.08 ± 0.02) (P< 0.05). Histopathological investigation showed more apoptosis and dead neurons in hippocampus and cerebellum brain sections of WT neonate mice after exposure to hyperoxia than in TG mice; this finding was also confirmed by TUNEL staining. The caspase 3 assay confirmed the finding of more apoptosis in WT hyperoxia neonates (0.814 ± 0.112) than in the TG hyperoxic group (0.579 ± 0.144) (P < 0.05); this finding was also confirmed by TUNEL staining. Quantitative western blotting for the inflammatory and metabolic markers showed significantly higher expression in the WT group than in the TG and control groups. Thus, overexpression of EC-SOD in the neonate brain offers significant protection against hyperoxia-induced brain damage.  相似文献

Minisatellites are highly variable tandem repeats used for over 20 years in humans for DNA fingerprinting. In prokaryotes fingerprinting techniques exploiting VNTR (variable number of tandem repeats) polymorphisms have become widely used recently in bacterial typing. However although many investigations into the mechanisms underlying minisatellite variation in humans have been performed, relatively little is known about the processes that mediate bacterial minisatellite polymorphism. An understanding of this is important since it will influence how the results from VNTR experiments are interpreted. The minisatellites of Mycobacterium tuberculosis are well characterized since they are some of the few polymorphic loci in what is otherwise a very homogeneous organism. Using VNTR results from a well-defined and characterized set of M. tuberculosis strains we show that the repeats at a locus are likely to evolve by stepwise contraction or expansion in the number of repeats. A stochastic continuous-time population mathematical model was developed to simulate the evolution of the repeats. This allowed estimation of the tendency of the repeats to increase or decrease and the rate at which they change. The majority of loci tend to lose rather than gain repeats. All of the loci mutate extremely slowly, with an average rate of 2.3 x 10(-8), which is 350 times slower than that of a set of VNTR repeats with similar diversity observed experimentally in Escherichia coli. This suggests that the VNTR profile of a strain of M. tuberculosis will be indicative of its clonal lineage and will be unlikely to vary in epidemiologically-related strains.  相似文献

Experimental work was performed on drill cuttings sampled from off-shore drilling operations. The cuttings contained remnants of two different types of drilling muds: a water-ilmenite based mud and an olefin-barite based mud. In a pilot experiment, a gradient of up to 65 mm thick layers of water-ilmenite based cuttings were added to 78 cm2 core samples. In another set-up, 3 mm layers of water-ilmenite and olefin-barite based cuttings were added to replicate 1000 cm2 box-core samples with natural benthic communities transferred from the Oslofjord, S.E. Norway. Boxes with no addition and addition of 3 mm “clean” sediment were used for control. Increased consumption of oxygen and nitrate indicated the presence of degradable organic phases in both mud systems. The release of silicate showed a general decrease with increasing thickness of the cuttings layer, but maximum rates in the 3.1 mm treatment indicated increased bioturbation at low and moderate doses. The initial (field) composition of the macrobenthic communities was maintained throughout the 3 months experimental period, and at community level, no significant difference was observed between the four treatments at the end of the exposure period. However, after grouping the treatments into “clean” (untreated control and 3 mm clean sediment) and “cuttings” (3 mm water-ilmenite and 3 mm olefin-barite based cuttings), multivariate statistical analyses revealed a significant difference in community composition between clean and cuttings treatments, and three taxa showed significantly reduced abundances in the cuttings treatments. Chemical toxicity of mud components is assumed to be small, and the observed effect was more likely a result of physical properties such as the size or shape of cuttings particles.  相似文献
