Floating and nodularin-producing strains of Nodularia spumigena from the Baltic Sea are regarded as belonging to one species. However, intraspecific variation in the response of N. spumigena to environmental factors has been commonly overlooked. As blooms of N. spumigena occur in late summer, a period with strong light and stable water-column stratification, the cells can be expected to also be exposed to ultraviolet-B radiation (UV-B, 280–320 nm). The UV-B tolerance of four different strains of N. spumigena, isolated from the Baltic Sea, was investigated in the laboratory for 8 days, by measuring photosynthesis, growth and pigment composition. Variables included maximum quantum yield of photosynthesis (Fv/Fm, PAM fluorometry), growth rate (cell counts) and photosynthetic pigments, as well as mycosporine-like amino acids (HPLC). Intraspecific differences regardless of treatment were found for cell dimension, growth rate, Fv/Fm and pigment concentrations. UV-B related effects differed between strains. By Day 8 one of the four strains showed a lower Fv/Fm when treated with UV-B; in another strain the growth rate and cell numbers were lower. In three strains, UV-B exposure resulted in higher cell concentrations of carotenoids and chlorophyll a. In all strains, the concentrations of total mycosporine-like amino acids were 60–130% higher in the UV-B treated samples compared with samples shielded from UV-B. Although strain-specific differences in UV-B tolerance were observed, it is concluded that N. spumigena is a species that is not generally negatively affected by moderate levels of UV-B radiation.  相似文献

Abstract The ecosystems of coastal and enclosed seas are under increasing anthropogenic pressure worldwide, with Chesapeake Bay, the Gulf of Mexico and the Black and Baltic Seas as well known examples. We use an ecosystem model (Ecopath with Ecosim, EwE) to show that reduced top-down control (seal predation) and increased bottom-up forcing (eutrophication) can largely explain the historical dynamics of the main fish stocks (cod, herring and sprat) in the Baltic Sea between 1900 and 1980. Based on these results and the historical fish stock development we identify two major ecological transitions. A shift from seal to cod domination was caused by a virtual elimination of marine mammals followed by a shift from an oligotrophic to a eutrophic state. A third shift from cod to clupeid domination in the late 1980s has previously been explained by overfishing of cod and climatic changes. We propose that the shift from an oligotrophic to a eutrophic state represents a true regime shift with a stabilizing mechanism for a hysteresis phenomenon. There are also mechanisms that could stabilize the shift from a cod to clupeid dominated ecosystem, but there are no indications that the ecosystem has been pushed that far yet. We argue that the shifts in the Baltic Sea are a consequence of human impacts, although variations in climate may have influenced their timing, magnitude and persistence. Fumonisins are a group of fungal toxins, occurring worldwide in maize infected mainly by Fusarium verticillioides. This paper describes the level of fumonisins in maize seed samples and the ability of F. verticillioides strains isolated from maize seeds grown in India to produce fumonisins. Forty-three seed samples intended to be used for consumption were collected from different regions of Karnataka and Andhra Pradesh. The samples were subjected to the agar plate method for the detection of F. verticillioides. Identification of F. verticillioides was done based on morphological characters and further confirmed by polymerase chain reaction. The majority of the samples were infected by F. verticillioides and infection percentage in the individual samples ranged from 5 to 51%. Twenty-three out of 35 (65%) strains were positive for fumonisin production in high performance liquid chromatography (HPLC) and competitive direct-enzyme linked immuno sorbent assay (CD-ELISA). Fumonisin level in seed samples ranged from 200 to 1,722 μg/g using CD-ELISA. HPLC could differentiate FB1 and FB2 toxins; out of 35 strains, 14 (40%) showed both FB1 and FB2 production. These findings indicate that there may be a risk of human exposure to fumonisins through the consumption of F. verticillioides infected corn-based foods in India.  相似文献

African and Asian populations of Fusarium spp. (Gibberella fujikuroi species complex) associated with Bakanae of rice (Oryzae sativa L.) were isolated from seeds and characterized with respect to ecology, phylogenetics, pathogenicity and mycotoxin production. Independent of the origin, Fusarium spp. were detected in the different rice seed samples with infection rate ranges that varied from 0.25% to 9%. Four Fusaria (F. andiyazi, F. fujikuroi, F. proliferatum and F. verticillioides) were found associated with Bakanae of rice. While three of the Fusaria were found in both African and Asian seed samples, F. fujikuroi was only detected in seed samples from Asia. Phylogenetic studies showed a broad genetic variation among the strains that were distributed into four different genetic clades. Pathogenicity tests showed that all strains reduced seed germination and possessed varying ability to cause symptoms of Bakanae on rice, some species (i.e. F. fujikuroi) being more pathogenic than others. The ability to produce fumonisins (FB1 and FB2) and gibberellin A3 in vitro also differed according to the Fusarium species. While fumonisins were produced by most of the strains of F. verticillioides and F. proliferatum, gibberellin A3 was only produced by F. fujikuroi. Neither fumonisin nor gibberellin was synthesized by most of the strains of F. andiyazi. These findings provide new information on the variation within the G. fujikuroi species complex associated with rice seed and Bakanae disease.  相似文献

We examined the molecular basis of factor IX deficiency in 53 unrelated Polish patients with hemophilia B. Heteroduplex analysis and direct sequencing of polymerase chain reaction (PCR) products were applied to identify the gene defect. Forty-three different point mutations were detected in the factor IX gene of 47 patients. There were 29 missense mutations, 9 nonsense mutations, 4 splice site mutations and 1 point mutation in the promoter region. Twelve mutations were novel. The results of this study emphasize a very high degree of heterogeneity of hemophilia B.  相似文献

Various solid dispersions of alpha-, beta- and gamma-cyclodextrin (CD) in PEG 6000 with and without the addition of 5% w/w indomethacin were prepared by the melting method using the original components. The samples were investigated by solid-state (13)C NMR, and the interactions between the drug and the cyclodextrins were evaluated. The indomethacin-gamma-CD phase with tetragonal symmetry found in a previous X-ray study gave chemical shifts which suggested that this phase is a complex between indomethacin and gamma-CD. Evidence of an indomethacin-beta-CD complex were found. A distribution of the chemical shifts for beta-CD was attributed to the possible formation of different types of complexes between indomethacin and beta-CD. No complex formation was found in the alpha-CD system. The degree of relative crystallinity of the samples in the gamma-CD system was measured by (1)H NMR, X-ray powder diffraction (XRD), differential scanning calorimetry (DSC), and modulated-temperature DSC (MTDSC). The results obtained by the NMR, XRD, and DSC techniques showed that the dispersions were less crystalline than the pure polymer carrier, and the dispersion containing the indomethacin-gamma-CD complex had the lowest degree of crystallinity. By the MTDSC method a deviation was found for the PEG 6000/indomethacin dispersion. This emphasizes that the different techniques give specific information on the crystallinity.  相似文献

Forty-five adult outpatients with endoscopically confirmed gastric ulceration completed a double-blind trial of either cimetidine (1 g/day) or placebo. After six weeks 18 of the 23 patients receiving cimetidine showed complete ulcer healing compared with only six of the 22 patients receiving placebo. The cimetidine group also had fewer days with pain than the placebo group but the difference was not statistically significant. Cimetidine therefore seems to promote healing of gastric ulcers without severe side effects, although its effect on pain is less pronounced than in patients with duodenal ulcers.  相似文献

The coordination sphere of the two metal-binding sites/subunit of the homotetrameric D-xylose isomerase from Streptomyces rubiginosus has been probed by the investigation of the Co2(+)-substituted enzyme using electronic absorption, CD and magnetic circular dichroic spectroscopies in the visible region. The spectrum of the high-affinity site (B site) has an absorption coefficient, epsilon 545, of 18 M-1 cm-1, indicating a distorted octahedral complex geometry. The spectrum of the low-affinity site (A site) shows two absorption maxima at 505 nm and 586 nm with epsilon values of 170 M-1 cm-1 and 240 M-1 cm-1, respectively, which indicates a distorted tetrahedral or pentacoordinated complex structure as also observed for the enzyme from Streptomyces violaceoruber [Callens et al. (1988) Biochem. J. 250, 285-290] having the same feature but lower epsilon values. The first 4 mol Co2+ added/mol apoenzyme occupy both sites nearly equally. Subsequently the Co2+ located in the A site slowly moves into the B site. After equilibrium is reached, the next 4 mol Co2+/mol again occupy the A site with its typical spectrum, restoring full activity. Addition of 4 mol Cd2+ or Pb2+/mol Co4-loaded derivative displaces the Co2+ from the B site to form the Pb4/Co4 derivative containing Co2+ in the A site, reducing activity fourfold while the Pb4/Pb4 species is completely inactive. In contrast, Eu3+ displaces Co2+ preferentially from the A site. Thus, the high- and low-affinity sites may be different for different cations. After addition of the substrates D-xylose, D-glucose and D-fructose and the inhibitor xylitol the intense Co2+ A-site spectrum of both the active Co4/Co4 derivative and the less active Pb4/PCo4 derivative decreases, indicating that these compounds are bound to the A site, changing the distorted tetrahedral or pentacoordinated symmetry there to a distorted octahedral complex geometry.  相似文献

In the mammalian retina, glutamate re-uptake is mediated by the sodium dependent cotransport systems EAAT1-5 thus terminating neuronal excitation and preventing neuroexcitotoxicity. In retinal amacrine and ganglion cells, EAAT5 is colocalized with the serum and glucocorticoid inducible kinase SGK1, a serine/threonine kinase known to regulate transport. The study explored the possible regulation of EAAT5 by SGK1, its isoform SGK3, and the closely related protein kinase B. EAAT5 was coexpressed in Xenopus laevis oocytes with or without the respective kinases. Transport activity was quantified by electrophysiology and cell surface expression was determined by chemiluminescence. Both EAAT5 mediated currents and EAAT5 protein abundance at the cell surface were increased by a factor of 1.5-2 upon coexpression of SGK1 or SGK3 but not following coexpression of PKB. In conclusion, the kinases SGK1 and SGK3 increase EAAT5 activity by increasing cell surface abundance of the carrier.  相似文献
