Summary The apparent substrate constants of the amylolytic enzymes produced by the mould Trichoderma harzianum CBS 354.33 were measured. The value for -amylase was 64 mg starch·1-1 which is very low as compared with those of other -amylases. The substrate constant for glucoamylase was 78 mg starch·l-1. Both enzymes were sensitive to Acarbose; 50% inhibition was observed at 2.5 mg·l-1 (-amylase) and 0.10 mg·l-1 (glucoamylase).  相似文献

Lysophosphatidic acid (LPA) is a lipid mediator of a large number of biological processes, including wound healing, brain development, vascular remodeling, and tumor progression. Its role in tumor progression is probably linked to its ability to induce cell proliferation, migration, and survival. In particular, the ascites of ovarian cancers is rich in LPA and has been implicated in growth and invasion of ovarian tumor cells. LPA binds to specific G protein-coupled receptors and thereby activates multiple signal transduction pathways, including those initiated by the small GTPases Ras, Rho, and Rac. We report here a genetic screen with retroviral cDNA expression libraries to identify genes that allow bypass of the p53-dependent replicative senescence response in mouse neuronal cells, conditionally immortalized by a temperature-sensitive mutant of SV40 large T antigen. Using this approach, we identified the LPA receptor type 2 (LPA(2)) and the Rho-specific guanine nucleotide exchange factor Dbs as potent inducers of senescence bypass. Enhanced expression of LPA(2) or Dbs also results in senescence bypass in primary mouse embryo fibroblasts in the presence of wild-type p53, in a Rho GTPase-dependent manner. Our results reveal a novel and unexpected link between LPA signaling and the p53 tumor-suppressive pathway.  相似文献

Embryonic stem cells (ESCs) form descendants of all three germ layers when differentiated as aggregates, termed embryoid bodies. In vivo, differentiation of cells depends on signals and morphogen gradients that provide instructive and positional cues, but do such gradients exist in embryoid bodies? We report here the establishment of anteroposterior polarity and the formation of a primitive streak-like region in the embryoid body, dependent on local activation of the Wnt pathway. In this region, cells undergo an epithelial-to-mesenchymal transition and differentiate into mesendodermal progenitors. Exogenous Wnt3a protein posteriorizes the embryoid body, resulting in predominantly mesendodermal differentiation. Conversely, inhibiting Wnt signaling promotes anterior character and results in neurectodermal differentiation. The activation of Wnt signaling and primitive streak formation requires external signals but is self-reinforcing after initiation. Our findings show that the Wnt pathway mediates the local execution of a gastrulation-like process in the embryoid body, which displays an unexpected degree of self-organization.  相似文献

The total nitrogen mass balance was studied over a two-year period in two reservoirs located in south-eastern Poland. Theobserved nitrogen retention (Nret) inthe reservoirs ranged from 0.18 to 2.12g m-2d-1 (mean 0.83g m-2d-1) in case of theRzeszów Reservoir and from -0.02 to0.41 g m-2d-1 (mean 0.13g m-2d-1) in case of the SolinaReservoir, while the percentage retention(Nret) ranged from 7 to 42% (mean22%) and -15 to 73% (mean 35%),respectively. A good relationship betweenNret and nitrogen loads was observedfor the Solina Reservoir. The data obtainedby the author and collected from theresearches of 6 other reservoirs located insouthern Poland were used to develop anempirical model suitable to predict thenitrogen seasonal dynamics in a reservoir.The model allows for the determination ofboth the annual mean Nret value andits seasonal variation in a reservoir.  相似文献

Autotransporters (ATs) of Gram-negative bacteria are often produced with an unusual signal peptide that carries a conserved N-terminal extension. Using combined in vitro and in vivo approaches we show that the extension of the AT hemoglobin protease (Hbp) does not affect targeting of Hbp via the SRP-pathway, suggesting that the extension is not involved in targeting pathway selection.  相似文献

Abstract. By the approval of the European Bird Directive (79/409/EEC) in 1979 and the Habitat Directive (92/43/EEC) in 1992, nature protection has become an important part of European law with direct consequences for land use, industrial development and agriculture. If a member state reclaims an area that is protected by these directives, it has to take compensational measures. When such an area includes saline grasslands that require very specific abiotic conditions, a place with similar conditions must be available to re‐establish those grasslands at another location in a sustainable way. This will limit the number and extent of possible sites for compensation. In the conflict between economy and ecology in the Schelde polders, Flanders has chosen to expand the port of Antwerp in the polders at the left Schelde bank and destroy a protected saline grassland site. As a consequence, it has to compensate for this loss. Therefore, we made a detailed study of the hydrology, geology and soil conditions of the endangered saline grassland site. The same was done in four possible compensation sites, all situated in nearby polders. We found that the presence of upwelling groundwater flowing through a saline peat layer is necessary for the occurrence of saline grasslands. Only in one of the four possible compensation sites, called Muggenhoek, we found such conditions. So, we indicate this place as a possible site for the re‐creation of saline grasslands and give some recommendations for their development.  相似文献
