The complete primary structures of the major Asn-linked oligosaccharides from the type II variant surface glycoproteins (VSGs), MITat 1.2 and MITat 1.7, and the type III VSG, MITat 1.5, were determined using a combination of exo- and endoglycosidase digestions, methylation analysis, acetolysis, and 500 MHz 1H NMR spectroscopy. Each variant contained classical branched oligomannose-type and biantennary complex oligosaccharides, a proportion of the latter substituted with terminal alpha(1-3)-linked galactose residues, the first report of the presence of this epitope in Trypanosoma brucei. In addition both the type II variants contained relatively large amounts of the unusual small oligomannose-type oligosaccharides, Man4GlcNAc2 and Man3GlcNAc2, and a diverse array of novel branched poly-N-acetyllactosamine oligosaccharides, similar but not identical to those from mammalian glycoproteins. These latter structures were also partially substituted with terminal alpha(1-3)-linked galactose residues. Glycosylation in the type II variants showed site specificity in that the poly-N-acetyllactosamine and Man(9-5)GlcNAc2 oligosaccharides were located exclusively at Asn-glycosylation site 1 very close to the C terminus, whereas the Man(4-3)GlcNAc2 and biantennary complex oligosaccharides were located exclusively at site 2. Sagittaria papillosa Buch. is monoecious with unisexual flowers, pistillate below, staminate above, typically with an unbranched scape. A large population with unusual numbers of staminate and bisexual flowers on the lowest whorl of the inflorescence and many particles was quantitatively evaluated. First-formed inflorescences had more staminate and bisexual flowers than those produced later. Branched scapes were predominantly found to be the second inflorescence produced by a given plant. Genetic crosses between flowers on recemes and panicles produced no branched inflorescences. When grown under greenhouse conditions all tested plants had racemes with pistillate flowers in the lower whorls and staminate ones above. Data from soil parameters, daylengths and air temperatures are compared to reported information on modification of flower sexuality by these factors. 相似文献

Use of habitat is a critical component related to structure of small-mammal communities, with partitioning occurring primarily along dimensions of microhabitat, although use of microhabitat often does not explain fully use at a macrohabitat level. Through grid studies of small mammals in coastal Colima, Mexico (during January 2003–2005), we appraised influence of available habitat, species richness, abundance, and cumulative abundance of other small mammals on variation in habitat used by species. We evaluated 14 habitat variables (reflecting ground cover, slope, canopy, and vegetation density on vertical and horizontal axes) and developed a composite variable (principal component 1) reflecting general openness of habitat through which we addressed habitat use. For the four most common mammalian species (Sigmodon mascotensis, Heteromys pictus, Baiomys musculus, and Oryzomys couesi), two measures of variation in habitat used were employed to estimate niche breadth, one of which assessed variation in habitat use relative to variation present on a grid. Sigmodon mascotensis and B. musculus preferred areas that were more open, and H. pictus and O. couesi occupied less-open areas; breadth of habitat use did not differ interspecifically. Habitat use was more variable on grids with more variability in habitat, although not greater than chance expectations. Findings do not lend support to the resource-breadth hypothesis as an explanation for population densities of species at a local level or the habitat-heterogeneity hypothesis as a predictor of species richness. Variation in habitat used by S. mascotensis did not proportionally increase when diverse habitat was available but was greater when the species was more abundant. For H. pictus, when cumulative abundance of other small mammals was greater, breadth of habitat used was greater. Intraspecific density-dependent habitat selection may result in S. mascotensis selecting a greater variety of habitats, while greater interspecific abundance is related to a greater range in use of habitats by H. pictus. Baiomys musculus used a higher proportion of habitat relative to that available when more species were present on a grid. Variation in habitat used by O. couesi was unrelated to any factor examined. Overall, the four species responded in notably different ways with respect to availability of habitat, abundance, and presence of other species. 相似文献

A general formalism for calculating parameters describing physiological acid-base balance in single compartments is extended to multicompartment systems and demonstrated for the multicompartment example of human whole blood. Expressions for total titratable base, strong ion difference, change in total titratable base, change in strong ion difference, and change in Van Slyke standard bicarbonate are derived, giving calculated values in agreement with experimental data. The equations for multicompartment systems are found to have the same mathematical interrelationships as those for single compartments, and the relationship of the present formalism to the traditional form of the Van Slyke equation is also demonstrated. The multicompartment model brings the strong ion difference theory to the same quantitative level as the base excess method. 相似文献

Two thousand plants of the water hyacinth,Eichornia crassipes Solms., were introduced on April 11, 1971, into a series of five ponds, each 5000 sq. ft. in area and 2.6 ft. deep. Treated waste water effluent from the Ames sewage treatment plant filled the ponds and was added to pond 1 at 127 gallons per minute. By growth and vegetative reproduction, these plants increased to more than 500,000, and all five ponds were covered completely by July 26. On that date, the extrapolated estimate of total wet weight was 287 U.S. tons/ acre (645 metric tons/hectare; 64500g/m2). The estimate of oven dry weight was 13.2 U.S. tons/acre (29.7 metric tons/hectare; 2970g/m2). Ammonia and nitrate disappeared rapidly from the pond water, and phosphate concentrations were lowered appreciably. Evapotranspiration and seepage accounted for water losses of more than 0.5 inches per day. The potential economic values of this plant and its possible use in tertiary treatment to reduce N and P components in waste waters are discussed briefly. 相似文献

Rapid purification of protein kinase C from rat brain. A novel method employing protamine-agarose affinity column chromatography 总被引:3,自引:0,他引:3
We describe a rapid purification of protein kinase C from rat brain cytosol employing a specific substrate, protamine-coupled to agarose. Sequential chromatography on DEAE-Sephacel, phenyl-Sepharose CL-4B, and protamine-agarose columns resulted in a 1,500-fold purification of protein kinase C. SDS-PAGE analysis of the purified enzyme resolved a doublet protein of 77-80 kDa. This doublet was recognized by a polyclonal antiserum against protein kinase C. Proteolytic digestion of each protein band generated similar peptide fragments. The underlying principle of the protamine sulfate purification method was also clarified. Protamine can serve as a Ca2+/phospholipid-independent substrate. We demonstrate phosphorylation of protamine on the column; phosphorylated protamine did not bind the enzyme with the same affinity and this covalent modification was most probably responsible for releasing the bound enzyme from the column after addition of Mg2+ and ATP. The C kinase inhibitor, H7, inhibits protamine phosphorylation in a dose-dependent fashion but does not prevent binding of the enzyme to a protamine-agarose column. We therefore conclude that protamine interacts with the active center of the enzyme enabling it to be phosphorylated, upon which it loses its binding affinity for C kinase. 相似文献

Treatment of intact rat pancreatic acini with phorbol ester (12-O-tetradecanoyl-phorbol-13-acetate, TPA) resulted in a time- and concentration-dependent translocation of phospholipid/Ca2+-dependent protein kinase (PL/Ca-PK) from the soluble fraction. Redistribution of PL/Ca-PK was concurrent with stimulation of amylase secretion by TPA-treated acini. Polymyxin B, a potent and selective inhibitor of PL/Ca-PK completely inhibited TPA-induced amylase secretion. Cyclins are indispensable elements of the cell cycle and derangement of their function can lead to cancer formation. Recent studies have also revealed more mechanisms through which cyclins can express their oncogenic potential. This review focuses on the aberrant expression of G1/S cyclins and especially cyclin D and cyclin E; the pathways through which they lead to tumour formation and their involvement in different types of cancer.
