We describe a new species of small-eared shrew, genus Cryptotis Pomel, 1848 (Eulipotyphla: Soricidae), from near the community of Monteverde in the Tilarán highlands of northwestern Costa Rica. The new species is immediately distinguished from all other Costa Rican shrews its large size and long tail. Morphologically, it belongs to the Cryptotis thomasi group of small-eared shrews, a clade that is more typically distributed in the Andes Cordillera and other highland regions of northern South America. The new Costa Rican species and the Panamanian endemic Cryptotis endersi Setzer, 1950 are the only two members of this species group known to occur in Central America. Like most other members of the C. thomasi group for which the postcranial skeleton has been studied, the new species tends be more ambulatory (rather than semi-fossorial) when compared with other members of the genus. Our survey efforts over several decades failed to locate a population of the new species, and we discuss its conservation status in light of its limited potential distribution in the Tilarán highlands and the significant climatic change that has been documented in the Monteverde region during the past four decades.  相似文献

To obtain reliable classical and integrated interspecies maps of the long arm of chromosome 7 of tomato, detailed mapping work was undertaken and several phenotypic and molecular markers were assigned loci on both maps to provide reliable cross-reference points. To maximise the value of the new maps, pair-wise segregation data for classical genetic markers from the literature were included, based on large segregating populations with readily scorable phenotypes. In addition, to increase confidence in these maps, introgression lines were used to confirm important map locations. The revised classical map is based on two- and three-point test-cross data from a number of F2 and BC1 mapping populations. The integrated interspecies map is based on F2 mapping populations derived from crosses of Lycopersicon esculentum with Lycopersicon pennellii (LA716). The genetic analyses for both maps were performed using the computer package JoinMap. The revised composite classical map indicates that some of the map positions reported in the literature are incorrect. The linear order of the classical markers common to both the revised classical and integrated interspecies maps are in complete agreement. Production of the integrated interspecies map resulted in re-orientation of the existing molecular map. Received: 13 September 2000 / Accepted: 20 December 2000  相似文献

Myosoricinae is a small clade of shrews (Mammalia, Eulipotyphla, Soricidae) that is currently restricted to the African continent. Individual species have limited distributions that are often associated with higher elevations. Although the majority of species in the subfamily are considered ambulatory in their locomotory behavior, species of the myosoricine genus Surdisorex are known to be semifossorial. To better characterize variation in locomotory behaviors among myosoricines, we calculated 32 morphological indices from skeletal measurements from nine species representing all three genera that comprise the subfamily (i.e., Congosorex, Myosorex, Surdisorex) and compared them to indices calculated for two species with well‐documented locomotory behaviors: the ambulatory talpid Uropsilus soricipes and the semifossorial talpid Neurotrichus gibbsii. We summarized the 22 most complete morphological variables by 1) calculating a mean percentile rank for each species and 2) using the first principal component from principal component analysis of the indices. The two methods yielded similar results and indicate grades of adaptations reflecting a range of potential locomotory behaviors from ambulatory to semifossorial that exceeds the range represented by the two talpids. Morphological variation reflecting grades of increased semifossoriality among myosoricine shrews is similar in many respects to that seen for soricines, but some features are unique to the Myosoricinae. Recombination in enteroviruses provides an evolutionary mechanism for acquiring extensive regions of novel sequence, is suggested to have a role in genotype diversity and is known to have been key to the emergence of novel neuropathogenic variants of poliovirus. Despite the importance of this evolutionary mechanism, the recombination process remains relatively poorly understood. We investigated heterologous recombination using a novel reverse genetic approach that resulted in the isolation of intermediate chimeric intertypic polioviruses bearing genomes with extensive duplicated sequences at the recombination junction. Serial passage of viruses exhibiting such imprecise junctions yielded progeny with increased fitness which had lost the duplicated sequences. Mutations or inhibitors that changed polymerase fidelity or the coalescence of replication complexes markedly altered the yield of recombinants (but did not influence non-replicative recombination) indicating both that the process is replicative and that it may be possible to enhance or reduce recombination-mediated viral evolution if required. We propose that extant recombinants result from a biphasic process in which an initial recombination event is followed by a process of resolution, deleting extraneous sequences and optimizing viral fitness. This process has implications for our wider understanding of ‘evolution by duplication’ in the positive-strand RNA viruses.  相似文献

Small‐eared shrews (Mammalia: Soricidae: Cryptotis), exhibit modifications of the forelimb skeleton that have been interpreted as adaptations for semifossoriality. Most species inhabit remote regions, however, and their locomotory and foraging behaviors remain mostly speculative. To better understand the morphological modifications in the absence of direct observations, we quantified variation in these species by measuring 151 individuals representing 18 species and populations of Cryptotis and two species of moles (Talpidae) for comparison. From our measurements, we calculated 22 indices, most of which have been used previously to characterize substrate use among rodents and other taxa. We analyzed the indices using 1) average percentile ranks, 2) principal components analysis, and 3) cluster analysis. From these analyses, we determined that three basic modes of substrate adaptation are present within Cryptotis: 1) a primarily terrestrial mode, with species that are capable of burrowing, but lack adaptations to increase digging efficiency, 2) a semifossorial mode, with species whose forelimbs bones show strong muscle attachment areas and increased mechanical advantage, and 3) an intermediate mode. In addition to identifying new morphological characters and contributing to our understanding of the functional morphology of soricids, these analyses provide additional insight into the ecology of the species of interest. The early endosome is organised into domains to ensure the separation of cargo. Activated mitogenic receptors, such as epidermal growth factor (EGF) receptor, are concentrated into vacuoles enriched for the small GTPase Rab5, which progressively exclude nutrient receptors, such as transferrin receptor, into neighbouring tubules. These vacuoles become enlarged, increase their content of intralumenal vesicles as EGF receptor is sorted from the limiting membrane, and eventually mature to late endosomes. Maturation is governed by the loss of Rab5 and is accompanied by the movement of endosomes along microtubules towards the cell centre. Here, we show that EGF relocates to the cell centre in a dynein-dependent fashion, concomitant with the sorting away of transferrin receptor, although it remains in Rab5-positive early endosomes. When dynein function is acutely disrupted, efficient recycling of transferrin from EGF-containing endosomes is retarded, loss of Rab5 is slowed and endosome enlargement is reduced.  相似文献

The scorpion, Urodacus manicatus (Scorpionida: Urodacidae), inhabits temperate sclerophyll woodland in south-eastern Australia and excavates a shallow burrow into soil beneath a rock. Mean minimum and maximum temperatures within a burrow were respectively higher and lower than outside measurements, and relative humidity within a burrow remained higher than outside the burrow at all times. Using flow-through respirometry, VCO2 and water loss were measured at three ecologically relevant temperatures (10, 20 and 30 °C) to assess patterns of gas exchange and the temperature dependence of VCO2 and water loss rates. Daytime VCO2 corresponds to a lower metabolic rate than reported for the closely related, but more arid region inhabiting species Urodacus armatus and Urodacus yaschenkoi. CO2 output is continuous at 10 °C; however, at 20 °C scattered sharp depressions are observed and these become more frequent and periodic at 30 °C. Total water loss in U. manicatus is nearly double that in U. armatus, indicative of lessened selective pressure for water conservation within a mesic rather than xeric environment.  相似文献

The purpose of this study was to test the hypothesis that interval sprint training (IST) selectively increases endothelium-dependent dilation (EDD) and endothelial nitric oxide synthase and/or superoxide dismutase-1 protein content in arteries and/or arterioles that perfuse the white portion of rat gastrocnemius muscle (WG). Male Sprague-Dawley rats completed 10 wk of IST (n = 62) or remained sedentary (Sed) (n = 63). IST rats performed six 2.5-min exercise bouts, with 4.5 min of rest between bouts (60 m/min, 15% incline), 5 days/wk. EDD was assessed from acetylcholine (ACh)-induced increases in muscle blood flow measured in situ and by ACh-induced dilation of arteries and arterioles [first to third order (1A-3A)] that perfuse red gastrocnemius muscle (RG) and WG. Artery protein content was determined with immunoblot analysis. ACh-induced increases in blood flow were enhanced in WG of IST rats. eNOS content was increased in conduit arteries, gastrocnemius feed artery, and fourth-order arterioles from WG and fifth-order arterioles of RG but not in 2As from RG. EDD was examined in 2As and 3As from a subset of IST and Sed rats. Arterioles were canulated with micropipettes, and intraluminal pressure was set at 60 cmH2O. Results indicate that passive diameter (measured in 0 calcium PSS) of WG 2As was similar in IST and Sed, whereas diameter of WG 3As was greater in IST (96 +/- 8 microm) than Sed (73 +/- 4 microm). WG 2As and 3As of IST rats exhibited greater spontaneous tone, but sensitivity to stretch, phenylephrine, and sodium nitroprusside was similar to Sed arterioles. ACh-induced dilation was enhanced by IST in WG 2As but not in RG 2As or WG 3As. We conclude that IST induces vascular adaptations nonuniformly among arteries that perfuse WG muscle.  相似文献

Allozyme balances serve as markers of quantitative behavior of electrophoretically distinguishable alleles. By the use of ADH Set I allozyme balances, it is demonstrated that all Adh1-S/Adh1-F individuals from more than 20 diverse S/F families exhibit a reciprocal correlation between Adh1 quantitative behavior in two maize organs: the scutellum and primary root. Within an electrophoretic mobility class, the Adh1 allele that is relatively underexpressed in the scutellum is relatively overexpressed in the primary root, and vice versa. Segregation tests prove that this "reciprocal effect" is the property of a cis-acting site that is closely linked to or within the Adh1 structural gene, and it is not affected by diverse genetic backgrounds. Immunological and [(3)H]-leucine incorporation experiments establish that Adh1 quantitative variants differ in ADH1.ADH1 synthetic rates in the anaerobic primary root. The reciprocal-effect phenomenon suggests that the cis-acting loci controlling Adh1 quantitative expression in each respective organ are at least in close proximity, or may share common DNA sequences. We discuss the possibility that the reciprocal-effect locus is a regulatory component of the Adh1 cistron.  相似文献
