Clathrin-mediated endocytosis mediates the internalization of signaling and nutrient receptors, ion channels and regulates the endocytic recycling of pre- and postsynaptic membrane proteins. During early stages endocytic adaptors recognize sorting signals within this diverse array of cargo proteins destined for internalization. Cargo sequestration is mechanistically coupled to membrane deformation, a process involving BAR domain proteins, resulting in the generation of endocytic intermediates that finally undergo dynamin-mediated fission. Here we summarize recent insights gathered from a combination of structural, biochemical, and cell biological studies that have revealed a remarkable complexity of the machinery for endocytic sorting and membrane deformation. 相似文献

ATP-binding cassette (ABC) transport systems mediate the translocation of solutes across biological membranes at the expense of ATP. They share a common modular architecture comprising two pore-forming transmembrane domains and two nucleotide binding domains. In prokaryotes, ABC transporters are involved in the uptake of a large variety of chemicals, including nutrients, osmoprotectants and signal molecules. In pathogenic bacteria, some ABC importers are virulence factors. Canonical ABC import systems require an additional component, a substrate-specific receptor or binding protein for function. Interaction of the liganded receptor with extracytoplasmic loop regions of the transmembrane domains initiate the transport cycle. In this review we summarize the current knowledge on receptor-transporter interplay provided by crystal structures as well as by biochemical and biophysical means. In particular, we focus on the maltose/maltodextrin transporter of enterobacteria and the transporters for positively charged amino acids from the thermophile Geobacillus stearothermophilus and Salmonella enterica serovar Typhimurium. 相似文献

It is currently unclear whether tissue changes surrounding multifocal epithelial tumors are a cause or consequence of cancer. Here, we provide evidence that loss of mesenchymal Notch/CSL signaling causes tissue alterations, including stromal atrophy and inflammation, which precede and are potent triggers for epithelial tumors. Mice carrying a mesenchymal-specific deletion of CSL/RBP-Jκ, a key Notch effector, exhibit spontaneous multifocal keratinocyte tumors that develop after dermal atrophy and inflammation. CSL-deficient dermal fibroblasts promote increased tumor cell proliferation through upregulation of c-Jun and c-Fos expression and consequently higher levels of diffusible growth factors, inflammatory cytokines, and matrix-remodeling enzymes. In human skin samples, stromal fields adjacent to multifocal premalignant actinic keratosis lesions exhibit decreased Notch/CSL signaling and associated molecular changes. Importantly, these changes in gene expression are also induced by UVA, a known environmental cause of cutaneous field cancerization and skin cancer. 相似文献

To study the impact of nutritional factors on protein expression of intestinal bacteria, gnotobiotic mice monoassociated with Escherichia coli K-12 were fed three different diets: a diet rich in starch, a diet rich in nondigestible lactose, and a diet rich in casein. Two-dimensional gel electrophoresis and electrospray-tandem mass spectrometry were used to identify differentially expressed proteins of bacteria recovered from small intestine and cecum. Oxidative stress response proteins such as AhpF, Dps, and Fur, all of which belong to the oxyR regulon, were upregulated in E. coli isolates from mice fed the lactose-rich diet. Luciferase reporter gene assays demonstrated that osmotic stress caused by carbohydrates led to the expression of ahpCF and dps, which was not observed in an E. coli ΔoxyR mutant. Growth of ahpCF and oxyR deletion mutants was strongly impaired when nondigestible sucrose was present in the medium. The wild-type phenotype could be restored by complementation of the deletions with plasmids containing the corresponding genes and promoters. The results indicate that some OxyR-dependent proteins play a major role in the adaptation of E. coli to osmotic stress. We conclude that there is an overlap of osmotic and oxidative stress responses. Mice fed the lactose-rich diet possibly had a higher intestinal osmolality, leading to the upregulation of OxyR-dependent proteins, which enable intestinal E. coli to better cope with diet-induced osmotic stress. 相似文献

Carnivorous plants trap, digest and absorb animals in order to supplement their mineral nutrition. Nutrients absorbed by the plant include different nitrogen species, phosphate, potassium, trace elements and small organic compounds. Uptake is usually thought to be performed via specific channels, but this study provides evidence that endocytosis is involved as well. Traps of the carnivorous plants Nepenthes coccinea, Nepenthes ventrata, Cephalotus follicularis, Drosophyllum lusitanicum, Drosera capensis, Dionaea muscipula, Aldrovanda vesiculosa, Genlisea violacea × lobata, Sarracenia psittacina and Sarracenia purpurea were stained with methylene blue in order to identify possible sites of uptake. The permeable parts of the traps were incubated with fluorescein isothiocyanate labelled bovine serum albumin (FITC-BSA) and other fluorescent endocytosis markers, combined with the soluble protein BSA or respiratory inhibitors. Uptake was studied by confocal microscopy. In Nepenthes, small fluorescent vesicles became visible 1 h after incubation with FITC-BSA. These vesicles fused to larger compartments within 30 h. A similar behaviour was found in the related genera Drosera, Dionaea, Aldrovanda and Drosophyllum but also in Cephalotus with glands of different evolutionary origin. In Genlisea and Sarracenia, no evidence for endocytosis was found. We propose that in many carnivorous plants, nutrient uptake by carriers is supplemented by endocytosis, which enables absorption and intracellular digestion of whole proteins. The advantage for the plant of reducing secretion of enzymes for extracellular digestion is evident. 相似文献

Interventions to conserve carbon stored in forests are central to the emerging global climate change regime. Widely referred to as REDD+, these interventions engage local resource holders in contracts to restrict their use of land and forests in exchange for conditional benefits, effectively creating a market for forest carbon—a new and intangible commodity. Delving into the social and material implications of this, three case studies (Papua New Guinea, Philippines, Cambodia) examine property relations in the early stages of forest carbon production in different tenure contexts. The case studies reveal that: (a) the risk of local exclusion from forest and lands under REDD+ is real, but is mediated by dynamic negotiations over knowledge and property; (b) the relationship between forest carbon and underlying property relations around land and forests is recursive and mutually constitutive; and (c) due to ongoing and entrenched property contests in REDD+ locations, there remains an unstable foundation for forest carbon markets. 相似文献

Global conservation discourses and practices increasingly rely on market-based solutions to fulfill the dual objective of
forest conservation and economic development. Although varied, these interventions are premised on the assumption that natural
resources are most effectively managed and preserved while benefiting livelihoods if the market-incentives of a liberalised
economy are correctly in place. By examining three nationally supported payment for ecosystem service (PES) schemes in Vietnam
we show how insecure land tenure, high transaction costs and high opportunity costs can undermine the long-term benefits of
PES programmes for local households and, hence, potentially threaten their livelihood viability. In many cases, the income
from PES programmes does not reach the poor because of political and economic constraints. Local elite capture of PES benefits
through the monopolization of access to forestland and existing state forestry management are identified as key problems.
We argue that as PES schemes create a market for ecosystem services, such markets must be understood not simply as bald economic
exchanges between ‘rational actors’ but rather as exchanges embedded in particular socio-political and historical contexts
to support the sustainable use of forest resources and local livelihoods in Vietnam. 相似文献
