Rapid self-assembly of alpha-synuclein observed by in situ atomic force microscopy

Self-assembly of alpha-synuclein resulting in protein aggregates of diverse morphology has been implicated in the pathogenesis of Parkinson's disease and other neurodegenerative disorders known as synucleinopathies. Apart from its biomedical relevance, this aggregation process is representative of the interconversion of an unfolded protein into nanostructures with typical amyloid features. We have used in situ tapping mode atomic force microscopy to continuously monitor the self-assembly of wild-type alpha-synuclein, its disease-related mutants A30P and A53T, and the C-terminally truncated variant alpha-synuclein(1-108). Different aggregation modes were observed depending on experimental conditions, i.e. pH, protein concentration, polyamine concentration, temperature and the supporting substrate. At pH 7.5, in the absence of the biogenic polyamines spermidine or spermine, elongated sheets 1.1(+/-0.2)nm in height and presumably representing individual beta-sheet structures, were formed on mica substrates within a few minutes. Their orientation was directed by the crystalline substructure of the substrate. In contrast, sheet formation was not observed with hydrophobic highly oriented pyrolytic graphite substrates, suggesting that negatively charged surfaces promote alpha-synuclein self-assembly. In the presence of spermidine or spermine 5.9(+/-1.0)nm high spheroidal structures were preferentially formed, sharing characteristics with similar structures previously reported for several amyloidogenic proteins and linked to neurotoxicity. alpha-Synuclein spheroid formation depended critically on polyamine binding to the C terminus, revealing a promoting effect of the C terminus on alpha-synuclein assembly in the bound state. In rare cases, fibril growth from spheroids or preformed aggregates was observed. At pH 5.0, fibrils were formed initially and incorporated into amorphous aggregates in the course of the aggregation process, providing evidence for the potential of amyloid fibril surfaces to act as nucleation sites in amorphous aggregation. This study provides a direct insight into different modes of alpha-synuclein self-assembly and identifies key factors modulating the aggregation process.     

A contribution to the rust flora (Uredinales) on Aizoaceae in southern Africa

Rust fungi on Aizoaceae in southern Africa have been examined and reported based on 27 specimens collected during a biodiversity study and previously collected herbarium specimens. Eight species including five new species have been recognized, and they are described in detail and illustrated. Together with two additional species in literature, ten species of rust fungi are now recognized on Aizoaceae in southern Africa. Part 223 in the series “Studies in Heterobasidiomycetes” from the Botanical Institute, University of Tübingen.     

Gruppen-Gründung und Reproduktion bei Nacktmullen (Heterocephalus glaber) im Zoo Dresden

When establishing a new breeding group of Naked mole-rats three significant hurdles are to overcome: 1. Determination of sex. 2. Get over the group specific smell (between individuals of different groups). 3. Get over the reproductive blockade. This report describes experiences achieving these goals as well as establishes requirements for a successful breeding. The sex determination of nonreproductive Naked mole rats is tainted with great uncertainty, because the anogenital region of both sexes appears quite similar. For a lack of births and a failure of rearing offspring the following reasons seem to be responsible: 1. Incorrect sex determination at the time of group founding. 2. The reproductive blockade could not be overcome because of disadvantageous hierarchical relations within the group. 3. Pregnancy is interrupted due to disturbances from outside the enclosure or social stress within the group. 4. Due to these disturbances the alpha-female is not often enough willing to suckle her youngsters.     

Pluralizing the Costeños—Reflections on Michael Schroeder’s cultural geographies of grievance and war: Nicaragua’s Atlantic Coast region in the first Sandinista revolution, 1927–1934

In this response article to Michael Schroeder's contribution "Cultural Geographies of Grievance and War: Nicaragua's Atlantic Coast Region in the First Sandinista Revolution, 1927–1934", the author argues that Sandino's limited success in the region was not mainly the result of an erroneous view of the local situation. His main difficulty was the absence of a coherent Atlantic Coast society and culture. In view of the mosaic of heterogeneous groups involved in long-standing rivalries or recent conflicts immersed in the racist environment of the enclave economy, a political project that could have integrated all major population groups was impossible. Sandinista propaganda did not merely consist of lofty nationalist and spiritualist jargon. There were also efforts to address the concrete problems of the people on the Atlantic Coast, such as economic depression and unemployment, and the Moravian missionaries’ attempt to enforce strict sexual morals in indigenous and Creole settlements.     

Synthesis and analytics of 2,2,3,4,4-d5-19-nor-5alpha-androsterone--an internal standard in doping analysis

A short and efficient synthesis of pentadeuterated 2,2,3,4,4-d5-19-nor-5alpha-androsterone 7 starting from 19-norandrost-4-ene-3,17-dione 1 by a d1-L-Selectride mediated stereo- and regioselective reduction of the 3-keto group is presented. The use of compound 7 as internal standard for the detection of anabolic steroids via mass spectrometric techniques such as gas chromatography-mass spectrometry (GC-MS) is discussed.
