Two methods are described for locating the O-(carboxymethyl) groups in O-(carboxymethyl)guar. In Method I, O-(carboxymethyl)guar was depolymerized by methanolysis, the O-(carboxymethyl) groups were reduced, and the mixture of methyl glycosides and O-(2-hydroxyethyl)-substituted methyl glycosides was converted into a mixture of per-O-acetylated alditols and partially O-(2-acetoxyethyl)ated, partially O-acetylated alditols. Analysis of these alditols by gas-liquid chromatography-mass spectrometry allowed the positions of substitution of the O-(carboxymethyl) groups on the galactosyl groups and mannosyl residues to be determined. However, this method did not distinguish between O-(carboxymethyl) substitution on 4-linked and 4,6-linked mannosyl residues. This limitation was overcome by the more-detailed analysis provided by Method II, in which O-(carboxymethyl)guar was carboxyl-reduced, the product methylated, the glycosyl residues hydrolyzed, the sugars reduced, and the alditols acetylated to yield a mixture of partially O-acetylated, partially O-methylated alditols and partially O-acetylated, partially O-(2-methoxyethyl)ated, partially O-methylated alditols. These derivatives, when separated and quantitated by g.l.c., and identified by g.l.c.-m.s., gave a quantitative measure of every type of carboxymethyl substitution in guar.  相似文献

Unexposed blanks prepared from Kodak AR 10 stripping film were dried by 3 methods: (1) fast, open drying with a fan for 25 min, (2) slow drying in a desiccator for 6 hr, and (3) very slow drying in a desiccator for 24 hr. The number of background grains depended on the mode of drying. Fast drying (method 1) gave 0.7 grain per 100 μ2, slow drying (method 2) gave 0.33 grain; very slow drying (method 3), only 0.17 grain. The increase of background after fast drying is assumed to be caused by the rapid shrinkage of wet emulsion. This causes an increase in the intraemulsion pressure which, in turn, sensitizes the silver bromide crystals to cause an increase in the number of developable grains.  相似文献

A total of 132 Fusarium isolates belonging to 19 species sensu Nelson et al (1983) originating from Poland, Italy, and international cultures collections were examined for their ability to produce mycotoxin moniliformin. Moniliformin was produced by the following isolates:

  • F solani (Mart) Appel & Wollenw: 1 out of 14,670 mg/kg
  • F subglutinans (Wollenw & Reinking) Nelson et al: 8 out of 20,70 – 1660 mg/kg
  • F tricinctum (Corda) Sacc: 2 out of 9,130 – 1330 mg/kg

    Summary In shaken cultures, a strain of Pseudomonas cepacia isolated from apple leaves produced pyrrolnitrin and four other phenylpyrrole antibiotics. The concentrations of these metabolites were determined at intervals for 7 days in three different media at two initial pH levels. Optical density measurements revealed maximum cell concentrations after 24 h in nutrient broth, after 48 h in King's B medium, and after 96 h in minimum salts solution. The effects caused by initiating fermentations at pH 5.8 rather than 7.0 were in most cases not dramatic, although in some instances, especially in minimum salts broth, higher concentrations of metabolites were produced with the lower initial pH. Concentrations of the phenylpyrrole antibiotics were greatly affected by choice of culture medium and incubation time. Concentrations of the two nitrophenyl metabolites, pyrrolnitrin and 2-chloropyrrolnitrin, rose throughout the 7-day incubation and were more than 20 times greater in minimum salts medium than in either King's B medium or nutrient broth. The maximum concentrations of each of the three aminophenyl metabolites (dichloroamino, trichloroamino and monochloroamino) occurred in different media, the monochloro compound in nutrient broth, the dichloro compound in Kings B medium and the trichloro compound in minimum salts medium. The time dependence of the concentrations of the five metabolites supports the proposed biosynthesis of these pyrroles from tryptophan by successive chlorinations followed by oxidation of the amino group at the end of the pathway.  相似文献

    We have surveyed naturally occurring plasmids in strains of Bacillus subtilis and the closely related species B. mojavensis and B. licheniformis. Previous studies have failed to find host-benefitting functions for plasmids of these species, suggesting that these plasmids are nonmutualistic. Only one type of plasmid was found in each plasmid-bearing strain, suggesting that most of the plasmids infecting these Bacillus species are in the same incompatibility group. A sample of 18 plasmids from these species ranged in size from 6.9 to 16 kb, with all but 6 plasmids falling into three size groups. These groups differed in the sizes of their host ranges and geographical ranges. All but 1 of the 18 plasmids from these three host species are homologous with one another. The cryptic plasmids from these three species are far less diverse than are plasmids (from other species) that are known to benefit their bacterial hosts. The low-level diversity among these cryptic plasmids is consistent with the hypothesis that host-benefitting adaptations play an important role in fostering the coexistence of plasmid populations, but other explanations for the low-level plasmid diversity are possible. Comparison of the phylogenies of the plasmids with those of their hosts suggests that Bacillus plasmids are horizontally transferred in nature at a low rate similar to that found for the colicin plasmids of Escherichia coli.  相似文献

    We have examined whether low temperature, as a pervasive thermodynamicstimulus, is sensed independently in different parts of thecell by studying low temperature responses of phosphoproteinsin isolated nuclei. The isolated alfalfa (Medicago saliva) nucleirespond to cold by rapid and reversible changes in phosphorylationlevel of their proteins. The population of such cold-regulatedphosphoproteins and the cold-stimulation of their phosphorylationare greater in a freezing-tolerant cultivar Apica than in asensitive cultivar Trek. With a 4-day cold treatment of theseedlings, additional proteins showing cold-stimulated phosphorylationappear in the nucleus of Apica while there is little changein the case of Trek. Furthermore, nuclei from cold-treated seedlingsof Apica, but not of Trek, show a large accumulation of heat-stableproteins. These results support the view that the low temperaturesensing and acclimation occur in all vital parts of the celland that accumulation of heat-stable nuclear proteins may berelated to freezing tolerance.     Summary Two experiments were performed to test needle architecture of Scots pine (Pinus sylvestris L.) seedlings when affected by non-shading neighbouring vegetation. Both experiments showed that seedlings reacted to the presence of such adjacent vegetation by changes in needle distribution around the main stem: more needles were found on the opposite side of the main stem to where the vegetation was located. However, this reaction was confined to needles from the main stem. Responses of seedlings to dead neighbours differed from the response to live neighbours and also from the response to a black curtain substituted for plant neighbours. It is suggested that reactions are mediated by the red/far red ratio of incident light.  相似文献

    Abdominal aortic aneurysms are a degenerative disease of the aorta associated with high mortality. To date, in vivo information to characterize the individual elastic properties of the aneurysm wall in terms of rupture risk is lacking. We have used time-resolved 3D ultrasound strain imaging to calculate spatially resolved in-plane strain distributions characterized by mean and local maximum strains, as well as indices of local variations in strains. Likewise, we here present a method to generate averaged models from multiple segmentations. Strains were then calculated for single segmentations and averaged models. After registration with aneurysm geometries based on CT-A imaging, local strains were divided into two groups with and without calcifications and compared. Geometry comparison from both imaging modalities showed good agreement with a root mean squared error of 1.22 ± 0.15 mm and Hausdorff Distance of 5.45 ± 1.56 mm (mean ± sd, respectively). Using averaged models, circumferential strains in areas with calcifications were 23.2 ± 11.7% (mean ± sd) smaller and significantly distinguishable at the 5% level from areas without calcifications. For single segmentations, this was possible only in 50% of cases. The areas without calcifications showed greater heterogeneity, larger maximum strains, and smaller strain ratios when computed by use of the averaged models. Using these averaged models, reliable conclusions can be made about the local elastic properties of individual aneurysm (and long-term observations of their change), rather than just group comparisons. This is an important prerequisite for clinical application and provides qualitatively new information about the change of an abdominal aortic aneurysm in the course of disease progression compared to the diameter criterion.

    Abstract A hybridoma cell line producing a human anti-lipid A monoclonal antibody (mAb), FKF-1F3 (IgM (κ)) was obtained by cell fusion of Epstein-Barr virus-transformed cells and mouse myeloma. The mAb bound to not only Gram-negative bacterial lipid A, but also to polysaccharide portions of Pseudomonas aeruginosa lipopolysaccharides (LPS). The mAb seemed to recognize two distinct regions of P. aeruginosa LPS other than lipid A, namely the outer core regions of some serotype strains and the O -polysaccharide region of serotype A strains. The mAb cross-reacted with N -acetyl-β-glucosamine-conjugated bovine serum albumin, N -acetyl-β-galactosamine-conjugated bovine serum albumin, myosin and actin, but not with other autoantigens such as ss- and ds-DNA, cardiolipin and glycosaminoglycans. The mAb conferred protective activity against a mouse pseudomonal infection model. The evidence suggested that the mAb was a naturally occurring polyspecific antibody that participated in defense against pseudomonal infections.  相似文献
