The mechanical stimuli resulting from weight loading play an important role in mature bone remodeling. However, the effect of weight loading on the developmental process in young bones is less well understood. In this work, chicks were loaded with bags weighing 10% of their body weight during their rapid growth phase. The increased load reduced the length and diameter of the long bones. The average width of the bag-loaded group's growth plates was 75 +/- 4% that of the controls, and the plates showed increased mineralization. Northern blot analysis, in situ hybridization, and longitudinal cell counting of mechanically loaded growth plates showed narrowed expression zones of collagen types II and X compared with controls, with no differences between the relative proportions of those areas. An increase in osteopontin (OPN) expression with loading was most pronounced at the bone-cartilage interface. This extended expression overlapped with tartarate-resistant acid phosphatase staining and with the front of the mineralized matrix in the chondro-osseous junction. Moreover, weight loading enhanced the penetration of blood vessels into the growth plates and enhanced the gene expression of the matrix metalloproteinases MMP9 and MMP13 in those growth plates. On the basis of these results, we speculate that the mechanical strain on the chondrocytes in the growth plate causes overexpression of OPN, MMP9, and MMP13. The MMPs enable penetration of the blood vessels, which carry osteoclasts and osteoblasts. OPN recruits the osteoclasts to the cartilage-bone border, thus accelerating cartilage resorption in this zone and subsequent ossification which, in turn, contributes to the observed phenotype of narrower growth plate and shorter bones. 相似文献

Decreased central arterial compliance is an emerging risk factor for cardiovascular disease. Resistance training is associated with reductions in the elastic properties of central arteries. Currently, it is not known whether this reduction is from one bout of resistance exercise or from an adaptation to multiple bouts of resistance training. Sixteen healthy sedentary or recreationally active adults (11 men and 5 women, age 27 +/- 1 yr) were studied under parallel experimental conditions on 2 separate days. The order of experiments was randomized between resistance exercise (9 resistance exercises at 75% of 1 repetition maximum) and sham control (seated rest in the exercise room). Baseline hemodynamic values were not different between the two experimental conditions. Carotid arterial compliance (via simultaneous B-mode ultrasound and applanation tonometry) decreased and beta-stiffness index increased (P < 0.01) immediately and 30 min after resistance exercise. Immediately after resistance exercise, carotid systolic blood pressure increased (P < 0.01), although no changes were observed in brachial systolic blood pressure at any time points. These measures returned to baseline values within 60 min after the completion of resistance exercise. No significant changes in these variables were observed during the sham control condition. These results indicate that one bout of resistance exercise acutely decreases central arterial compliance, but this effect is sustained for <60 min after the completion of resistance exercise. 相似文献

An accurate mathematical model of transmucosal gas exchange is prerequisite to understanding middle ear (ME) physiology. Current models require experimentally measured gas species time constants for all extant conditions as input parameters. However, studies on pulmonary gas exchange have shown that a morphometric model that incorporates more fundamental physiochemical and anatomic parameters accurately simulates transport from which the species time constants can be derived for all extant conditions. Here, we implemented a variant of that model for ME gas exchange that requires the measurement of diffusional length (tau) for the ME mucosa. That measure contributes to the mucosal diffusing capacity and reflects the resistance to gas flow between air space and capillary. Two methods for measuring tau have been proposed: linear distance between the air-mucosal boundary and capillary and the harmonic mean of all contributing pathway lengths. Oxygen diffusing capacity was calculated for different ME mucosal geometries by using the two tau measures, and the results were compared with those predicted by a detailed, two-dimensional finite element analysis. Predictive accuracy was improved by incorporating the harmonic tau measure, which captures important information regarding variations in capillary shape and distribution. However, compared with the oxygen diffusing capacity derived from the finite element analysis, both measures yielded nonlinear, positively biased estimates. The morphometric techniques underestimate diffusion length by failing to account for the curvilinear gas flow pathways predicted by the finite element model. 相似文献

The conditions for the formation and stability of quasi-crystal helical structures formed of charged dust grains of equal size in a complex plasma are investigated. A study is made of both the transverse confinement of such structures by means of an external parabolic potential well and the possibility of their self-confinement or confinement by the field of the smaller background grains. It is shown that, in the presence of dissipation, any initial random distribution of dust grains in a cylindrical external potential evolves into an equilibrium helical structure with a constant pitch angle. It is found that, when the external control parameter is smoothly varied, the pitch angle of the helix changes abruptly and the structure of the quasi-crystal undergoes bifurcations, during which the number of interwoven helices changes. The smaller the confinement parameter and the weaker the attractive forces between the grains, the larger the number of bifurcations. In addition, it is found that, because of the attraction (both noncollective and collective) between the dust grains, stable helical structures may exist not only at zero value of the external confinement parameter, but also at a negative one, and that the collective interaction between the grains not only increases the number of bifurcations but also changes their character. 相似文献

Distributions of the dusty plasma parameters (electron, ion, and dust densities; dust grain charge; and ion drift velocity) in quasineutral dust structures whose dimensions are much greater than the mean free path of the ions in their interactions with neutral particles are calculated numerically under conditions such that ionization sources are located outside the structures. Planar, cylindrical, and spherical structures are investigated. It is shown that static equilibrium structures are governed by a single (basic) parameter: the electrostatic potential drop between the center of the structure and its boundary. It is found that the maximum value of the basic parameter (in energy units) does not exceed the electron temperature. The basic parameter also determines the total number of dust grains in the structure and the power of external ionization sources that are necessary to sustain this structure. Significant improvements of plasma performance after ICRF boronization have been achieved in the full range of HT-7 operation parameters. Electron power balance is analyzed in the steady state ohmic discharges of the HT-7 tokamak. The ratio of the total radiation power to ohmic input power increases with increasing the central line-averaged electron density, but decreases with plasma current. It is obviously decreased after wall conditioning. Electron heat diffusivity χe deduced from the power balance analysis is reduced throughout the main plasma after boronization. χe decreases with increasing central line-averaged electron density in the parameter range of our study. After boronization, the plasma current profile is broadened and a higher current can be easily obtained on the HT-7 tokamak experiment. It is expected that the fact that the bootstrap current increases after boronization will explain these phenomena. After boronization, the plasma pressure gradient and the electron temperature near the boundary are larger than before, these factors influencing that the ratio of bootstrap current to total plasma current increases from several percent to above 10%. 相似文献

The effects of CO2 concentration (C
a) on growth, photosynthesis, and the activity of enzymes associated with the translocation and assimilation of CO2 were studied in sugar beet (Beta vulgaris L. subsp. saccharifera, cv. Ramonskaya) plants. The plants were grown in controlled-climate chamber to the stage of 3–4 leaves and then used in experiments. Experimental plants were exposed in boxes to doubled C
a (700 µl/l, 2C plants), whereas control plants were kept in a chamber with ambient atmosphere (350 µl/l, 1C plants). As compared with 1C plants, in 3 and 8 days, the leaf area of 2C plants increased by 14 and 26%, respectively. The rate of their photosynthesis (P
n) measured in 3, 6, and 8 days increased by 85, 47, and 52%, respectively, whereas in normal air, the values of P
n in 2C plants were by 12, 19, and 15% lower than in 1C plants. After 8-day growth, the content of soluble carbohydrates in the leaves of 2C plants attained 7.2%, being by 80% greater than in 1C plants; the content of starch did not exceed 3%. The total content of chlorophylls a and b in the leaves of 2C plants was by 14% greater than in 1C plants, but their ratio was essentially the same. The level of protein in 2C plants was by 13.4% lower than in 1C plants. The activity and content of Rubisco in 1C and 2C plants were similar. As compared with 1C plants, in 2C plants the activity of soluble carbonic anhydrase (sCA) was lower by 34% in 3 days and by 18% in 8 days; the activity of carbonic anhydrase of membrane preparations (mCA), was lower by 24 and 77%, respectively. Catalase activity in 2C plants became by 8% lower than in 1C plants only after 8 days. The kinetic curves of dark reduction of P700+ (oxidized primary donor of PSI) after far-red light irradiation were studied on broad bean (Vicia faba L.) leaves treated with antimycin A, methyl viologen, or diuron. Four components of P700+ reduction were found in untreated leaves, namely, an ultrafast component with a half-time of 25 ms, and fast (210 ms), middle (790 ms), and slow (6100 ms) components. The fast component disappeared in leaves treated with antimycin A or methyl viologen. At the same time, these substances did not affect other components of P700+ reduction. Treatment of leaves with diuron abolished both the ultrafast and fast components of P700+ reduction. As the length of far-red light exposure was increased, a lag phase appeared in the development of middle component in leaves treated with diuron, antimycin A, or methyl viologen. In thus treated leaves, an exponential pattern of the middle component was displayed with a certain delay after darkening. A conclusion was drawn that the minor ultrafast component of P700+ dark reduction in broad bean leaves was caused by electron donation to PSI from PSII, whereas the fast component of this process was determined by the operation of ferredoxin-dependent electron transport around PSI.
