A suspension culture of sycamore (Acer pseudoplatanus L.) was synchronised in division by release from nitrogen starvation. Cell samples, taken during the lag phase and synchronous growth, were examined cytologically and subjected to a freeze-preservation protocol. A high positive correlation was found between mitotic index (percentage of cells showing mitotic figures) and cell survival, as measured by fluorescein diacetate staining and reduction of 2,3,5-triphenyl tetrazolium chloride. Specific staining of lethally damaged cells and subsequent examination of the surviving cells demonstrated that the latter had a lower mitotic index and more consistent, low value of nuclear DNA than the total population. This indicated that it is cells, newly entered into G1 phase of the cell cycle, which are particularly resistant to the stresses imposed by the freeze preservation protocol.  相似文献

Laminin (LN)- or fibronectin (FN)-dependent adhesion in Krieger's ldlD 14 (D14) cells is enhanced significantly in the presence vs absence, of galactose (Gal), whereas LN- or FN-induced haptotactic cell motility is barely affected unless cells express CD82 by its gene transfection (cells termed D14/CD82). The effect of CD82 on LN- or FN-induced motility is based on its ability to associate with alpha3 or alpha5 integrin to form a complex associated with a low-density lipid membrane domain (termed GEM or GSD). Complex formation is greatly affected by N-glycosylation of both integrin and CD82, as well as by concurrent GM3 ganglioside synthesis. The effect of glycosylation on alpha5-CD82 complex was also studied in D14 cells expressing mutant CD82, defective in all three N-glycosylation sites. LN-induced motility was greatly inhibited, whereas FN-induced motility was enhanced, with complete N-glycosylation in D14/CD82 cells in Gal-added medium, whereby alpha5-CD82 complex formation did not occur or occurred at a minimal level. Both LN- and FN-induced motility were inhibited when N-glycosylation was impaired, or N-glycosylation of CD82 was deleted, whereby alpha5-CD82 complex formation occurred strongly. Thus, glycosylation profoundly affects interaction of integrin with CD82, leading to significant inhibition or promotion of cell motility.  相似文献

The therapeutic potential of glycosides has made them an attractive target for drug development. The biological extraction and chemical synthesis of these molecules is often challenging and low yielding, thus alternative methods for the synthesis of polysaccharides are being pursued. A new class of enzymes, glycosynthases, which are nucleophile mutants of glycosidases, can perform the transglycosylation reaction without hydrolyzing the product, and thus provide a valuable resource for polysaccharide and glycan synthesis. Directed evolution of glycosynthases has expanded the repertoire of glycosidic linkages formed and the donors and acceptors (both sugar and nonsugar) that can be used by the glycosynthase. The application of new screening methods, such as FACS, to the directed evolution of glycosynthases will aid in the development of enzymes that are able to efficiently synthesize new, and therapeutically relevant glycosidic linkages.  相似文献

The stereochemical courses of the hydrolyses catalysed by three glycosidases have been determined directly by 1H nmr. The anomeric configuration of the initially formed product was ascertained in each case by observation of the chemical shift and coupling constant of the anomeric proton at the new hemiacetal centre. Two of the enzymes investigated, an endo-glucanase and an exo-glucanase are components of the cellulase complex of Cellulomonas fimi. The third enzyme is the beta-glucosidase from almond emulsin. Two of these enzymes, the exo-glucanase and the almond beta-glucosidase catalysed hydrolysis with retention of anomeric configuration, in agreement with previous observations on the almond enzyme. The endo-glucanase catalysed hydrolysis with inversion of configuration, this result being confirmed by optical rotation measurements. This 1H nmr approach has several advantages over other techniques in that it is applicable to a wide variety of glycosidases and substrates and it is non-destructive, allowing recovery of the enzyme.  相似文献

选取上海环城林带7种植物群落,采用美景度评判法,从林内景观和林外景观2个空间层次和春、夏、秋、冬4个季节,应用数量化理论Ⅰ建立了美景度和各景观因子类目之间的景观评价与预测的多元回归模型,分析了群落的结构特征和季相特征对林内景观以及外貌特征对林外景观的影响,并提出相应的优化对策。结果表明:(1)群落结构特征对林内景观的影响主要因子为胸径(平均胸径和胸径变异系数)、郁闭度和疏透度。在春季,林内美景度随着树木胸径增大而增加;在夏季,郁闭度增大会提升林内美景度;在秋季,胸径变异小的群落具有更高的林内观赏性;在冬季,疏透度对林内景观美景度影响最大。(2)群落季相特征对林内景观的影响,在各季节表现亦不同。在春季,黄色、紫色等明度较高的色相和开花量适中的群落美景度最佳;在夏季,生长势好、林冠层变化小以及树干清晰度高的群落具较高的美景度,且观花可显著提高夏季林内美景度;在秋季,色彩越纯美景度越高;而在冬季,树皮颜色深的群落美景度高。(3)群落外貌特征对林外景观有显著影响,其中林冠线对林外景观美景度影响最大,其次为林缘线。具有起伏不大林冠线和自然流畅林缘线的植物群落美景度高。旨在通过对典型植被群落不同季相的美景度评价,对上海环城林带的群落景观进行定量的评价,进而为不同情景下的群落结构优化提出相应的对策,为城市森林的群落建构与管理提供科学依据。  相似文献
