c-Jun is essential for organization of the epidermal leading edge   总被引:9,自引:0,他引:9  
The migration of epithelial layers requires specific and coordinated organization of the cells at the leading edge of the sheet. Mice that are conditionally deleted for the c-jun protooncogene in epidermis are born at expected frequencies, but with open eyes and with defects in epidermal wound healing. Keratinocytes lacking c-Jun are unable to migrate or elongate properly in culture at the border of scratch assays. Histological analyses in vitro and in vivo demonstrate an inability to activate EGF receptor at the leading edge of wounds, and we demonstrate that this can be rescued by supplementation with conditioned medium or the EGF receptor ligand HB-EGF. Lack of c-Jun prevents EGF-induced expression of HB-EGF, indicating that c-jun controls formation of the epidermal leading edge through its control of an EGF receptor autocrine loop.  相似文献

Summary Unfermented cassava flour, also called high quality cassava flour (HQCF) is used as a partial substitute for wheat flour in the baking industry. This work was carried out to produce an indigenous fermented cassava dough, agbelima, from high quality cassava flour in order to diversify uses for the flour. An isolate of each of the dominant species of lactic acid bacteria isolated from agbelima, was used to ferment reconstituted HQCF dough into agbelima, whilst pH changes, population on MRS, and the organoleptic quality were assessed. The antimicrobial properties of the fermenting samples against three enteric pathogens were also investigated. Lactobacillus plantarum, Lactobacillus brevis, Lactobacillus fermentum and Leuconostoc mesenteroides were isolated at levels of 1010, 109, 109 and 108 c.f.u. g−1 respectively in agbelima. All isolates and a control sample, produced agbelima with pH of between 4.11 and 3.82 in 48 h, and ANOVA showed no significant difference between the pH of the samples at P ≤ 0.05. Spontaneous fermentation of the reconstituted dough was possible, because the flour was found to contain Lactobacillus plantarum at a level of 106 c.f.u. g−1 as well as the other species isolated from agbelima. Counts of Gram-positive catalase-negative rods and cocci on MRS in all the fermented samples were at levels of 108 to 109 c.f.u. g−1, with ANOVA showing no significant difference between the populations at P ≤ 0.05. Three enteric pathogens, Salmonella typhimurium 9, Echerichia coli D2188 and Vibrio cholerae C-230, inoculated into the samples at 106 to 107 c.f.u. g−1 at the start of fermentation, could not be detected in 10 g of any of the samples at the end of 48 h fermentation. A taste panel preferred banku - a stiff porridge made from a combination of fermented maize dough and fermented cassava dough – prepared with fermented HQCF dough to banku prepared with a market sample of agbelima or reconstituted agbelima flour. Processing the highly perishable cassava roots into high quality cassava flour, therefore, offers a means for preserving cassava, which can subsequently be used for both industrial and traditional purposes.  相似文献

This study investigates the effect of magnesium (Mg2+) on the secretory responses and the mobilization of calcium (Ca2+) and Mg2+ evoked by cholecystokinin-octapeptide (CCK-8) in the exocrine rat pancreas. In the isolated intact perfused pancreas CCK-8 (10–10 M) produced marked increases in juice flow and total protein output in zero and normal (1.1 mM) extracellular Mg2+ [Mg2+]o compared to a much reduced secretory response in elevated (5 mM and 10 mM) [Mg2+]o Similar effects of perturbation of [Mg2+]o on amylase secretion and 45Ca2+ uptake (influx) were obtained in isolated pancreatic segments. In pancreatic acinar cells loaded with the fluorescent bioprobe fura-2 acetomethylester (AM), CCK-8 evoked marked increases in cytosolic free Ca2+ concentration [Ca2+]i in zero and normal [Mg2+]o compared to a much reduced response in elevated [Mg2+]o Pretreatment of acinar cells with either dibutyryl cyclic AMP (DB2 cAMP) or forskolin had no effect on the CCK-8 induced changes in [Ca2+]i. In magfura-2-loaded acinar cells CCK-8 (10–8 M) stimulated an initial transient rise in intracellular free Mg2+ concentration [Mg2+]i followed by a more prolonged and sustained decrease. This response was abolished when sodium Na+ was replaced with N-methyl-D-glucamine (NMDG). Incubation of acinar cells with 10 mM Mg2+ resulted in an elevation in [Mg2+]i. Upon stimulation with CCK-8, [Mg2+]i. decreased only slightly compared with the response obtained in normal [Mg2+]o. CCK-8 caused a net efflux of Mg2+ in pancreatic segments; this effect was abolished when extracellular sodium [Na+]o was replaced with either NMDG or choline. The results indicate that Mg2+ can regulate CCK-8-evoked secretory responses in the exocrine pancreas possibly via Ca2+ mobilization. Moreover, the movement of Mg2+ in pancreatic acinar cells is dependent upon extracellular Na+.  相似文献

Phenotypic revertants of Finkel-Biskis-Riley (FBR)-murine sarcoma virus-transformed rat fibroblasts were isolated on the basis of their adherence to plastic tissue culture dishes in the absence of divalent cations. Some revertants had sustained deletions or inactivating mutations of the v-fos gene. However, two revertants expressed a functional v-fos gene at levels equal to that in the transformed parental cells, and therefore phenotypic reversion was due to mutations in nonviral genes. These revertants were considered nontransformed according to four criteria: (i) they were flat and had a nontransformed morphology, (ii) they were contact inhibited when grown to confluence, (iii) they did not display anchorage-independent growth in soft agar, and (iv) they did not form tumors in nude mice. Somatic-cell hybrids between the revertants and the transformed parental cells were nontransformed, suggesting that the revertants had sustained an activating mutation of a gene capable of suppressing transformation. The expression of c-jun, junB, and junD was not altered in the revertants, and they could not be transformed by transfection with a c-jun expression vector. The revertants were resistant to transformation by an activated c-Ha-ras gene but were susceptible to transformation by simian virus 40. Our results demonstrate the existence of a class of revertants that harbor genes capable of suppressing transformation by v-fos and some other oncogenes. This contrasts with previously described revertants of transformation by v-fos that contain recessive mutations.  相似文献

An immobilized lipase (triacylglycerol acylhydrolase, EC 3.1.1.3) suitable for fat interesterification has been prepared by precipitation onto diatomaceous earth (Celite) with acetone of a crude lipase preparation from an Aspergillus. Non-lipase material present in the preparation which precipitated at high acetone concentrations or ovalbumin added prior to the immobilization reduced the measured interesterification activity without affecting lipolytic activity. The non-lipase material reduced the interesterification activity by as much as 50%. The interesterification activity of immobilized preparations was enhanced by the use of higher concentrations of the crude lipase or, more substantially, by admixture of purified lipase.  相似文献
