It is well known that calcium-induced calcium-release in cardiac myocytes takes place in spatially restricted regions known as dyads, where discrete patches of junctional sarcoplasmic reticulum tightly associate with the t-tubule membrane. The dimensions of a dyad are so small that it contains only a few Ca2+ ions at any given time. Ca2+ signaling in the dyad is therefore noisy, and dominated by the Brownian motion of Ca2+ ions in a potential field. Remarkably, from this complexity emerges the integrated behavior of the myocyte in which, under normal conditions, precise control of Ca2+ release and muscle contraction is maintained over the life of the cell. This is but one example of how signal processing within the cardiac myocyte and other cells often occurs in small “nano-domains” where proteins and protein complexes interact at spatial dimensions on the order of ∼1-10 nm and at time-scales on the order of nanoseconds to perform the functions of the cell. In this article, we will review several examples of local signaling in nano-domains, how it contributes to the integrative behavior of the cardiac myocyte, and present computational methods for modeling signal processing within these domains across differing spatio-temporal scales. 相似文献

CD4 T cells are essential for immune control of γ-herpesvirus latency. We previously identified a murine MHC class II-restricted epitope in γ-herpesvirus-68 gp150 (gp150(67-83)I-A(b)) that elicits CD4 T cells that are maintained throughout long-term infection. However, it is unknown whether naive cells can be recruited into the antiviral CD4 T cell pool during latency. In this study, we generate a mouse transgenic for a gp150-specific TCR and show epitope-specific activation of transgenic CD4 T cells during acute and latent infections. Furthermore, although only dendritic cells can stimulate virus-specific CD8 T cells during latency, we show that both dendritic cells and B cells stimulate transgenic CD4 T cells. These studies demonstrate that naive CD4 T cells specific for a viral glycoprotein can be stimulated throughout infection, even during quiescent latency, suggesting that CD4 T cell memory is maintained in part by the continual recruitment of naive cells. 相似文献

Molecular cloning of the genes encoding the muscarinic acetylcholine receptors has shown that receptor subtypes classified on the basis of pharmacological properties are related polypeptides encoded by distinct genes. These studies have also revealed the existence of novel muscarinic receptor subtypes. Functional analysis of each of the subtypes expressed in mammalian cells indicates that the different subtypes activate distinct biochemical pathways, a finding that explains the tissue-specific physiological response elicited by the neurotransmitter, acetylcholine. 相似文献

Functionally distinct G proteins selectively couple different receptors to PI hydrolysis in the same cell 总被引:32,自引:0,他引:32
The number of G proteins identified by molecular cloning exceeds the number of known G protein functions. Here we show that a cell can possess multiple G proteins that carry out a similar function, the activation of phospholipase C, but couple selectively to different receptors, which are endogenous to the cell or introduced by DNA transfection. These G proteins (termed Gp) can be distinguished by their sensitivity to pertussis toxin. The assignment of a given Gp pathway to specific receptors is confirmed by the additivity relationships of the PI hydrolysis response mediated by the different receptors. Significantly different amounts of PI hydrolysis are activated through each Gp pathway, suggesting that Gp proteins also differ in their coupling to phospholipase C. These results indicate that distinct Gp pathways in a given cell exist to couple different receptors to PI hydrolysis selectively, and may specify the nature of the cellular response to different receptors by determining the magnitude of PI hydrolysis. 相似文献

Red light-induced changes in the gravitropism of roots of Zea mays variety Merit is a very low fluence response with a threshold of 10−9 moles per square meter and is not reversible by far red light. Blue light also affects root gravitropism but the sensitivity of roots to blue is 50 to 100 times less than to an equal fluence of red. A native of the Peruvian Andes (4,250 m) was studied before and after isovolemic hemodilution of the hematocrit from 62 to 42%. O2 transport was studied with newly developed catheters in the radial and pulmonary arteries. These catheters allowed continuous measurement of arteriovenous O2 content and intermittent cardiac output by thermodilution. During exercise tests, breath-by-breath gas exchange measurements also allowed cardiac output to be calculated by the O2-Fick technique. A complex series of interrelated physiological changes occurred in response to hemodilution. These included increased ventilation, increased arterial and mixed venous PO2, increased cardiac output (both heart rate and stroke volume), and improved ventilation-flow match. The general improvement in symptoms that followed hemodilution correlated well with increased anaerobic threshold and mixed venous PO2 during exercise. 相似文献
