The large N-linked oligosaccharides released from Schizosaccharomyces pombe by endo-beta-N-acetylglucosaminidase H were examined to determine how the negatively chargedpyruvylated galactoses present (Gemmill,T.R., and Trimble,R.B., 1996, J. Biol. Chem ., 271, 25945-25949) were attached to the oligosaccharide chains. Binding of biotinylated human serum amyloid P and peanut agglutinin to native and depyruvylated S.pombe glycoproteins, respectively, indicated that the pyruvylated epitope was likely to be in the beta configuration. Examination by high- field 1H NMR of whole glycans and a disaccharide fragment released from them on partial acid hydrolysis showed that the pyruvylated galactose species was in fact beta1,3-linked to a second galactose, and this occurred an average of five to six times on nominal Gal57Man64GlcNAc N- glycans. The pyruvate-2,(4,6)Gal-beta1,3Gal epitope is chemically similar to acetaldehyde-Galbeta1,3Gal groups found on the glycoproteins from Paramyxovirus-infected bovine kidney cells (Prehm, P., Scheid,A. and Choppin,P.W. ,1979, J. Biol. Chem ., 254, 9669-9677). The 1:1 stoichiometry between pyruvate and beta-linked galactose in these S.pombe glycans indicates that either pyruvate addition to terminal beta1,3Gal is highly efficient or that pyruvylated Gal is transferred en bloc to alpha1,2-linked Gal residues in theN-linked chains. In contradiction to many galactomannan-producing fungi, which add substantial amounts of Gal in the furanose form to their glycoproteins, all detectable Gal in the large S.pombe galactomannans is in the pyranose form, as found in higher eukaryotes. The current work shows that the S.pombe outer chain structure is a poly-alpha1,6Man backbone 2- O-substituted with either Gal or the pyruvylated galactobiose and contains little alpha1,2-linked or 2-O-substituted Man. This is in contrast to the S. cerevisiae outer chain, which is poly-alpha1,6Man substituted with alpha1,2-linked Man sidechains (Ballou,C.E. ,1990, Methods Enzymol , 185, 440-470).   相似文献

The abundant chromatin-associated human factor HCF-1 is a heterodimeric complex of HCF-1N and HCF-1C subunits that are essential for two stages of the cell cycle. The HCF-1N subunit promotes G1 phase progression, whereas the HCF-1C subunit ensures proper cytokinesis at completion of M phase. How the HCF-1C subunit functions is unknown. Here, we show that HCF-1C subunit depletion causes extensive mitotic defects, including a switch from monomethyl to dimethyl lysine 20 of histone H4 (H4-K20) and defective chromosome alignment and segregation. Consistent with these activities, the HCF-1C subunit can associate with chromatin independently of the HCF-1N subunit and regulates the expression of the H4-K20 methyltransferase PR-Set7. Indeed, upregulation of PR-Set7 expression upon loss of HCF-1 leads to improper mitotic H4-K20 methylation and cytokinesis defects. These results establish the HCF-1C subunit as an important M phase regulator and suggest that H4-K20 methylation status contributes to chromosome behavior during mitosis and proper cytokinesis.  相似文献

A method was developed for the simultaneous measurement of acetylene reduction, carbon dioxide evolution and oxygen uptake by individual root nodules of intact nitrogen-fixing plants (Alnus rubra Bong.). The nodules were enclosed in a temperature-controlled leak-tight cuvette. Assay gas mixtures were passed through the cuvette at a constant, known flow rate and gas exchange was measured by the difference between inlet and outlet gas compositions. Gas concentrations were assayed by a combination of an automated gas chromatograph and a programmable electronic integrator. Carbon dioxide and ethylene evolution were determined with a coefficient of variation which was less than 2%, whereas the coefficient of variation for oxygen uptake measurements was less than 5%. Nodules subjected to repeated removal from and reinsertion into the cuvette and to long exposures of 10% v/v acetylene showed no irreversible decline in respiration or acetylene reduction. This system offers long-term stability and freedom from disturbance artifacts plus the ability to monitor continuously, rapidly and specifically the changes in root nodule activity caused by environmental perturbation.  相似文献

The biogeographic affinities of the Cretaceous and early Tertiary angiosperm floras of the North American area (which includes Meso-America, and the Greater Antilles) have been the subject of considerable interest. Although recent treatments of isolated taxa have shown affinities between North American, European, east Asian and Neotropic floras, the relationships have not been quantified. This study compiles the records of fossils whose familial relationships seem secure. This provides a carefully culled, and uniformly presented review of the Cretaceous and Paleogene record from 1950 to 1989 and supplements LaMotte (1950). A subset of these records, which showed compelling evidence of subfamilial relationships, was analyzed to quantify the relationships of the Cretaceous, Paleocene, Eocene and Oligocene floras to other regions. The analysis suggests that for the entire period 24% of the fossil species had affinities with extant taxa from the Northern Hemisphere; 10% with taxa from the Northern Hemisphere that have a few species in South America; 17% with taxa from Eurasia; 3% with taxa with a disjunct Eurasian-South American pattern; 19% with taxa from South America and/or Africa; 8% with taxa from South America and/or Africa that have an important sister group in southeast Asia; 5% with taxa from the Old World; and 13% with taxa having other distribution patterns. Those fossils with affinities to Laurasian taxa are mostly found in the northern and western portions of the North American area. The fossils with affinities to South American and/or African taxa are found in the southern portions of North America, Meso-America, and the Greater Antilles. The taxa with disjunct distributions show both patterns. These patterns suggest that during this time there were wide-spread temperate elements, found throughout Laurasia; Boreotropical flora elements, distributed in North America, Europe and along the Tethys seaway to southeast Asia; and West Gondwana elements which show dispersion from South America across the proto-Caribbean. The paleobotanical data are compatible with current geological, paleontological and biogeographical studies.  相似文献

Summary Using a root nodule cuvette and a continuous flow gas exchange system, we simultaneously measured the rates of carbon dioxide evolution, oxygen uptake and acetylene reduction by nodules ofAlnus rubra. This system allowed us to measure the respiration rates of single nodules and to determine the effects of oxygen concentration and temperature on the energy cost of nitrogen fixation. Energy cost was virtually unchanged (2.8–3.5 moles of carbon dioxide or oxygen per mole of ethylene) from 16 to 26°C (pO2=20 kPa) while respiration and nitrogenase activity were highly temperature dependent. At temperatures below 16°C, nitrogenase activity decreased more than did respiration and as a result, energy cost rose sharply. Acetylene reduction ceased below 8°C. Inhibition of nitrogenase activity at low temperatures was rapidly reversed upon return to higher temperatures. At high temperatures (above 30°C) nitrogenase activity declined irreversibly, while respiration and energy cost increased.Energy cost was nearly unchanged at oxygen partial pressures of 5 to 20 kPa (temperature of 20°C). Respiration and nitrogenase activity were strongly correlated with oxygen tension. Below 5 kPa, acetylene reduction and oxygen uptake decreased sharply while production of carbon dioxide increased, indicating fermentation. Fermentation alone was unable to support nitrogenase activity. Acetylene reduction was independent of oxygen concentration from 15 to 30 kPa. Nitrogenase activity decreased and energy cost rose above 30 kPa until nearly complete inactivation of nitrogenase at 70–80 kPa. Activity declined gradually, such that acetylene reduction at a constant oxygen concentration was stable, but showed further inactivation when oxygen concentration was once again increased. While ecologists have long recognized the influence of spatial resolution on species distribution models (SDMs), they have given relatively little attention to the influence of temporal resolution. Considering temporal resolutions is critical in distribution modelling of highly mobile marine animals, as they interact with dynamic oceanographic processes that vary at time‐scales from seconds to decades. We guide ecologists in selecting temporal resolutions that best match ecological questions and ecosystems, and managers in applying these models. We group the temporal resolutions of environmental variables used in SDMs into three classes: instantaneous, contemporaneous and climatological. We posit that animal associations with fine‐scale and ephemeral features are best modelled with instantaneous covariates. Associations with large scale and persistent oceanographic features are best modelled with climatological covariates. Associations with mesoscale features are best modelled with instantaneous or contemporaneous covariates if ephemeral processes are present or interannual variability occurs, and climatological covariates if seasonal processes dominate and interannual variability is weak.  相似文献
