Matrix mineralization is a terminal process in osteoblast differentiation, and several approaches have been introduced to characterize the process in tissues or cultured cells. However, an analytical technique that quantitates in vitro matrix mineralization of live cells without any labeling or complex treatments is still lacking. In this study, we investigate a simple and enhanced optical method based on surface plasmon resonance (SPR) detection that can monitor the surface-limited refractive index change in real-time. During monitoring MC3T3-E1 cells in vitro culture every 2 days for over 4 weeks, the SPR angle is shifted with a greater resonance change in cells cultured with osteogenic reagents than those without the reagents. In addition, the SPR results obtained have a close relevance with the tendency of conventional mineralization staining and an inductively coupled plasma-based calcium content measure. These results suggest a new approach of a real-time SPR monitoring in vitro matrix mineralization of cultured cells. 相似文献

Background
This study aimed to generate prioritised goals for oral health services for people with disabilities as a first step in meeting the need for evidence based oral health services for people with disabilities in Ireland.Methods
The study used a three round modified e-Delphi method, involving dental service professionals and people with disabilities or their representatives, in Ireland. Three rounds were completed online using SurveyMonkey. Round 1 asked: “List what you think dental services for people with disabilities in Ireland should be like.” Items for subsequent rounds were generated from responses to Round 1. Round 2 and Round 3 used 5 point Likert scales to rank these items by priority: from No Priority (1) to Top Priority (5). Consensus was achieved on each item where at least 80% of respondents considered an item either High or Top Priority. A consensus meeting concluded the process.Results
Sixty-one panelists started and 48 completed the survey. The Delphi panel agreed on level of priority for 69 items and generated 16 consensus statements. These statements covered a range of topics such as access to care, availability of information and training, quality of care, dental treatment and cost. A recurrent theme relating to the appropriateness of care to individual need arose across topics suggesting a need to match service delivery according to the individual''s needs, wants and expectations rather than the disability type/diagnosis based service which predominates today.Conclusions
This process produced a list of prioritised goals for dental services for people with disabilities. This creates a foundation for building evidence-based service models for people with disabilities in Ireland. 相似文献997.

The oxygen transport system in mammals is extensively remodelled in response to repeated bouts of activity, but many reptiles
appear to be ‘metabolically inflexible’ in response to exercise training. A recent report showed that estuarine crocodiles
(Crocodylus porosus) increase their maximum metabolic rate in response to exhaustive treadmill training, and in the present study, we confirm
this response in another crocodilian, American alligator (Alligator mississippiensis). We further specify the nature of the crocodilian training response by analysing effects of training on aerobic [citrate
synthase (CS)] and anaerobic [lactate dehydrogenase (LDH)] enzyme activities in selected skeletal muscles, ventricular and
skeletal muscle masses and haematocrit. Compared to sedentary control animals, alligators regularly trained for 15 months
on a treadmill (run group) or in a flume (swim group) exhibited peak oxygen consumption rates higher by 27 and 16%, respectively.
Run and swim exercise training significantly increased ventricular mass (~11%) and haematocrit (~11%), but not the mass of
skeletal muscles. However, exercise training did not alter CS or LDH activities of skeletal muscles. Similar to mammals, alligators
respond to exercise training by increasing convective oxygen transport mechanisms, specifically heart size (potentially greater
stroke volume) and haematocrit (increased oxygen carrying-capacity of the blood). Unlike mammals, but similar to squamate

We used immunohistochemical procedures to investigate embryonic erythropoiesis in serial sections of chicken embryos after 2-13 days of incubation. Antibodies specific for the erythrocyte-specific histone H5, for embryonic hemoglobin, and for adult hemoglobin were used as markers for general, primitive, and definitive erythropoiesis, respectively. Histone H5 was present in erythrocytes at all of the stages studied, i.e., in both the primitive and definitive cells. Cell of the definitive lineage were first detected, at about 5-6 days of incubation, in erythroid foci in the mesenchyme around the vitelline stalk. At 7-9 days of incubation, a massive mesenchymal conglomeration of erythropoietic cells developed, extending from the cervical to the abdominal region and ventrally to the vertebral body, with its largest extensions being around the arteries in the mediastinum. Immunostaining revealed that these erythroid cells belonged to the definitive erythropoietic lineage. These cells had disappeared completely after 12 days of incubation, i.e., before erythropoiesis is visible in the bone marrow. These observations are consistent with the notion that the yolk sac is essential for the formation of the definitive erythroid lineage. 相似文献
