Summary A cDNA clone encoding the human T lymphocyte sheep erythrocyte receptor [the CD2 (T11) antigen] was used as a probe to define the chromosomal location of the gene. The signal, revealed by hybridisation to Southern blots of genomic DNA from somatic cell hybrids, showed a high degree of concordance for human chromosome 1. In particular, the hybrid F4Sc13C19 which contained the short arm only of human chromosome 1 was positive. The location of the CD2 gene to 1p13 was confirmed by in situ hybridisation. 相似文献

Summary Corner's rules for plant form relate the degree of branching to branch diameter, and branch diameter to leaf or inflorescence size. We report the first interspecific test of these rules for inflorescence size and branch diameter. We derived a simple corollary of Corner's rules; since leaf size and inflorescence size are both correlated to branch thickness, they may be correlated to each other. This corollary holds for Leucadendron and Protea (Proteaceae), and in certain other taxa in the Asteraceae, Bruniaceae and Pinaceae which also have leaves and reproductive structures on the same shoot. For such taxa this implies that selection for aspects of floral display (inflorescence size, pollination type) may also be expressed at the level of leaf size and vice versa. This has implications for many aspects of botany and also points to the importance of the co-ordinating role of plant architecture for aspects of plant form. 相似文献

Five cladoceran zooplankton species in a Venezuelan floodplainlake, Laguna la Orsinera, were analyzed to identify factorscontrolling their dynamics over a period encompassing isolation,inundation by the Orinoco River and drainage. The abundanceof each species increased rapidly to a maximum associated withflooding of the lake by the Orinoco River. We calculated birth,death and population growth rates, and developed a method forestimating uncertainty in these parameters, in order to interpretobserved cladoceran abundance patterns. Rapid increases in populationsizes resulted both from high birth rates associated with decreasingturbidity following inundation and from the hatching of restingeggs. Birth rates remained high throughout the study, but highdeath rates restricted each population to a brief maximum andthen maintained small population sizes throughout the remainderof the study. Our demographic analyses suggest that high mortalityresulted from intense predation by fish and by Chaoborus larvae,rather than from resource limitation, and that this mortalitywas the major factor regulating cladoceran abundance duringinundation and drainage in this tropical floodplain lake. 相似文献

Structures resembling Metallogenium spp. were observed in agar and in liquid cultures of a Mn-oxidizing basidiomycetous fungus only when Mn2+ was oxidized. Fungal viability was necessary for formation of the structures; Mn2+ concentration and the presence or absence of agar in the medium were important factors determining their morphology. Slide cultures revealed no identifiable cells in any stage of development. Fluorescent dyes that stained nucleic acids and polysaccharides in the fungal hyphae did not stain the Metallogenium-like structures. Likewise, Rhodamine 123, a fluorescent probe for membrane potential, stained fungal mitochondria, but did not stain the structures. Thin sections through the structures showed no biological membranes or other cellular features. Only the characteristic ultrastructure of biological Mn oxides were observed in serial thin sections. In agar, unfixed structures disappeared permanently during reduction of Mn oxides with hydroxylamine. Glutaraldehyde fixation stabilized these structures. Fixed structures lost most of their original phase density during reduction with hydroxylamine, but continuous microscopic observations showed that their phase density could be restored by staining with Coomassie blue. Structures that formed in liquid medium did not require stabilization with glutaraldehyde during reduction of Mn oxides. They, too, lost their original phase density during reduction with hydroxylamine; phase density could be restored by staining with cationic colloidal iron or Coomassie blue. The results suggest that the Metallogenium-like structures were formed as a result of Mn oxidation associated with exopolymers produced by the fungus.Non-standard abbreviations HEPES

Direct measurements of net production rates and pore water profiles of solutes in the fine-grained sediments of Saginaw Bay, imply corresponding steady-state fluxes to the overlying water of 1.1–1.3 (I), 450–1010 (NH4
+), 1250–2650 (Si(OH)4), 3000–3400 (Ca2+), 440–1330 (Mg2+), 1.5–728 (Fe2+), and 179–281 (Mn2+) moles/m2/day and 11.0–11.8 (alkalinity) meq/m2/day at 17.5 °C. Silica production rates in sediments apparently follow first order kinetics with a rate coefficient of 0.09/day and a steady-state silica concentration of 1.2 mM at 23.5°C. The remaining solutes follow kinetics approximately independent of solute concentration over the range of concentrations observed. Measured solute production rates are consistent with observed solute profiles only if lateral diffusion gradients are maintained in the sediments by the burrowing and irrigation activity of benthic organisms such asChironomous, the dominant burrower in Saginaw Bay. Assuming that solute fluxes from Saginaw Bay are representative of all of the post-glacial sediments of Lake Huron, the iodine flux from sediments is comparable to the total fluvial input of iodine. Both the psychrophile Aquaspirillum arcticum and the psychrotroph Bacillus psychrophilus were found to acquire thermotolerance when either heat shocked or treated with nalidixic acid; two conditions which also resulted in the induction of heat shock proteins and/or stress proteins and also cell filamentation. The possible relatedness of acquisition of thermotolerance and cell filamentation was examined by inhibiting cell filamentation with 1.5% KCl. A. arcticum cells which were heat shocked in the presence of KCl did not become filamentous nor acquire thermotolerance suggesting that these two responses may be related. On the other hand, when cells of B. psychrophilus were treated in a similar fashion, they also were prevented from cell filamentation but their ability to become thermotolerant was unaffected. When A. arcticum cells were heat shocked in the presence of chloramphenicol, heat shock protein synthesis was inhibited but not the acquistion of thermotolerance. Similar experiments with B. psychrophilus revealed that partial induction of heat shock proteins still occurred; however, no thermotolerance was exhibited.Abbreviations hsp(s)

Two litters of rattlesnakes (Crotalus enyo; n = 6 per litter) were raised in large cages and small plastic boxes, respectively. No significant differences in exploration of a novel environment were observed between the litters, or between either of them and a group of wild-caught C. enyo, suggesting that the common practice of rearing baby snakes in small cages has no debilitating consequences on these measures. In the absence of differences between the litters, the two samples were pooled and additional analyses revealed the existence of reliable individual variation. Experiment 2 compared the two litters and the wild-caught snakes on measures of prey-directed behavior. Fatty acid hydroperoxides in the plasma of 18 patients who were undergoing normal postoperative periods following major thoracic or abdominal operations were measured by using a sensitive assay based upon the activation of the cyclooxygenase activity of prostaglandin H synthase. Following major thoracic operations of nine patients, the mean difference between the arterial (0.49 ± 0.13 μM, mean ± S.E.M.) and mixed venous (−0.09 ± 0.12 μM) level of hydroperoxide was 0.58 ± 0.13 μM (p < 0.01). In marked contrast to this result, major abdominal operations of nine patients led to a mean difference between the arterial (−0.19 ± 0.16 μM) and mixed venous (0.46 ± 0.08 μM) hydroperoxide levels of −0.65 ± 0.17 μM (p < 0.01). Both pulmonary and intraabdominal tissues appear capable of generating significant amounts of fatty acid hydroperoxide in response to standard surgical procedures. The A-MV differences suggest that the blood-borne hydroperoxides were rapidly cleared from the circulation by tissue capillary beds. 相似文献

New aspects of the distribution and developmental appearance of the 44-kDa bone phosphoprotein (44K BPP, also called sialoprotein I or osteopontin) and bone gamma-carboxyglutamic acid (Gla)-containing protein (BGP, also called osteocalcin) during osteogenesis and dentinogenesis were investigated with immunocytochemical techniques using monospecific, affinity-purified polyclonal antibodies. Sections from newborn rat incisors and from various bone anlagen of newborn animals and fetuses were processed for detection of 44K BPP or BGP antigenicity. In addition, histochemical reactions for detection of alkaline phosphatase or calcium salts were performed on a number of the sections. The 44K BPP appears to be synthesized and secreted by chondrocytes only in the areas of cartilage-to-bone transition; these cells could participate indirectly in the process of bone formation by providing a suitable scaffold onto which primary marrow osteoblasts attach and spread. During osteogenesis, 44K BPP is found in bone-forming cells almost concomitantly with the appearance of alkaline phosphatase and before osteoid deposition, whereas BGP is still absent during early stages of mineralization. We hypothesize that this dramatic difference between the developmental appearance of 44K BPP and BGP reflects the delayed expression of the BGP gene relative to that of 44K BPP. In long-term cultures of bone marrow from adult mice, some fibroblastic cells expressed the 44K BPP phenotype; these cells could represent early osteogenic progenitor cells. Some experiments also suggested that, as with BGP, 44K BPP or an immunologically related protein is synthesized by some odontoblasts and secreted into predentin, prior to the onset of mineralization. 相似文献
