The mammalian in vivo micronucleus assay is widely used as part of the genotoxicity testing battery required during the development of new drugs. As such, the in vivo micronucleus assay has been used in a battery of assays for the assessment of cigarette ingredients or design modifications to help ensure that there is no increase in risk or any new risk introduced by these additions or modifications. The present series of studies was conducted to optimize and evaluate this assay for the assessment of the effects of mainstream smoke on the micronucleus frequency in the bone marrow and peripheral blood of rats. In a first experiment, the optimal conditions for performing the micronucleus assay in these tissues were determined. This was done by use of two compounds known for their micronucleus-inducing activity, i.e., the clastogen cyclophosphamide and the aneugen colchicine. In a second experiment, the effects of tube restraint on untreated control rats were investigated. In a third experiment, the optimal conditions were used to assess the clastogenic/aneugenic activity of cigarette smoke in Sprague-Dawley rats. The rat micronucleus assay in both bone marrow and peripheral blood is able to detect clastogenic and aneugenic activity. The flow cytometric determination of micronucleated cells in rat blood is at least as sensitive as determinations in bone marrow. No statistically significant differences were observed in micronucleus frequencies between rats with and without the additional stress of tube restraint; however, the cautious approach would be to use a fresh-air-exposed group (with tube restraint) as the negative control in inhalation experiments. Using the conditions identified as optimal in the above-mentioned experiments, the micronucleus assay was not able to detect effects induced by smoke from conventional cigarettes. Nevertheless, the micronucleus assay will remain a valuable tool as part of a testing battery used to investigate possible adverse effects related to product modifications.  相似文献

This paper explores the use of the rank set sampling (RSS) protocol as it pertains to the estimation of a population proportion. The maximum likelihood estimator (MLE) and the sample proportion, both based on the RSS data, are discussed and their corresponding asymptotic distributions are derived. Based on these results the MLE is found to be uniformly more efficient than the sample proportion. Nevertheless, both estimators are more efficient than the simple random sample proportion. The greatest gains in efficiency are obtained at the center of the parameter space. Finally, these results remain valid in the presence of judgment error. (© 2004 WILEY‐VCH Verlag GmbH & Co. Coffee beans contain the diterpene cafestol, which raises plasma cholesterol concentrations in humans. Daily consumption of 2 g coffee oil, which provides approximately 60 mg cafestol (equivalent to 5.7 mg cafestol/MJ), increases plasma cholesterol concentrations by 28%. We studied the effect of cafestol in coffee oil on gerbils and rats to determine whether the pathways that lead to cafestol-induced hypercholesterolemia in humans are also present in other species. We fed coffee oil from the same batch used in humans to female gerbils and rats. Gerbils were fed a semipurified diet containing 0.5% or 5% (w/w) coffee oil (equivalent to 8.7 and 86.8 mg cafestol/MJ, respectively) in the presence or absence of 0.05% (w/w) cholesterol for a period of 10 weeks. When compared with the gerbils fed no coffee oil, the addition of 0.5% coffee oil to the diets did not affect plasma cholesterol. Plasma cholesterol was significantly higher only when 5% coffee oil was fed, both in the absence (1.01 mmol/L, 33% higher) and presence (1.87 mmol/L, 70% higher) of dietary cholesterol. Liver weight was also significantly higher when 5% coffee oil was fed. Rats were also fed diets containing 0.5% or 5% coffee oil (equivalent to 8.7 and 86.8 mg cafestol/MJ) with and without 0.05% cholesterol for 8 weeks. Feeding 0.5% coffee oil compared with no coffee oil resulted in significantly higher plasma cholesterol levels throughout the study both in the absence (0.46 mmol/L, 27% higher) and presence (0.28 mmol/L, 15% higher) of dietary cholesterol. Diets containing 5% coffee oil appeared to be toxic. Thus, coffee oil diterpenes can result in higher plasma cholesterol in gerbils and rats. The failure to observe these effects in previous studies may be due to doses that were too low.  相似文献

Song in oscine birds is a culturally inherited mating signal and sexually dimorphic. From differences in song production learning, sex differences in song recognition learning have been inferred but rarely put to a stringent test. In zebra finches, Taeniopygia guttata, females never sing and the species has one of the greatest neuroanatomical differences in song-related brain nuclei reported for songbirds. Preference tests with sibling groups for which exposure to song had been identical during the sensitive phase for song learning in males, revealed equally strong influence of the tutor's song (here the father) on males' and females' adult song preferences. Both sexes significantly preferred the father's over unfamiliar song when having free control over exposure to playbacks via an operant task. The sibling comparisons suggest that this preference developed independently of the song's absolute quality: variation between siblings was as great as between nests. The results show that early exposure has an equally strong influence on males' and females' song preferences despite the sexual asymmetry in song production learning. This suggests that the trajectory for song recognition learning is independent of the one for song production learning.  相似文献

A marker-assisted selection (MAS) study was conducted on two recombinant inbred line (RIL) populations of common bean (Phaseolus vulgaris) to test the effectiveness of MAS for resistance to white mold (Sclerotinia sclerotiorum). Markers for quantitative trait loci (QTL) on linkage groups B2 and B7 that were previously associated with resistance and plant architectural avoidance traits in the resistant parent Bunsi were chosen. In the Bunsi/Midland population 10 RILs included in the MAS selected group developed significantly less disease than the control group based on two years of field evaluation under white mold pressure. Growth habit had no significant effect on disease severity or incidence. In the Bunsi/Raven RIL population, disease scores in the MAS selected group were significantly lower than scores in the control group over two years. Additional progress in enhancing resistance to white mold was detected when yield and plant architecture were included in the selection process. Lower disease scores among RILs were observed when comparisons were made to RILs selected using MAS alone. Yield is an important trait that should be considered when selecting for resistance to white mold. Finally the potential of Bunsi as a genetic donor of QTL for white mold resistance was confirmed in both populations studied. This study supported the effectiveness of MAS to enhance selection for a complexly inherited trait such as resistance to white mold in common bean.  相似文献

When studying histological characteristics of porcine pancreata in relation to islet isolation, a remarkably high number of hyperemic islets (HIs) was encountered. The abnormalities observed in these HIs ranged from a single dilated vessel to hemorrhages extending into the surrounding exocrine tissue. The aim of the present study was to compare pancreata with and without HI on islet isolation outcomes. This study involved a histological examination of 143 purebred (74 juvenile and 69 adult) and 47 crossbred (only juvenile) porcine pancreata. Islet isolation was performed in 48 purebred adult pigs and in 25 crossbred pigs. Tissue samples were stained with Aldehyde Fuchsine. The presence of HIs was scored semi-quantitatively (HI-, HI+). We observed HIs in 48% of the purebred and in 68% of the crossbred pigs. However, only 3.3±3.1% and 3.1±4.7% of all assessed islets was hyperemic in HI+ pancreata in purebred and crossbred pigs, respectively. In both groups, significantly higher endocrine cell mass was found in the HI+ pancreata (p<0.01). When the higher endocrine cell mass was taken into account, we found significantly lower yields in the HI+ pancreata in both purebred and crossbred pigs (p=0.03 in both groups). The presence of HIs occurs frequently in porcine donor-pancreata and is associated with reduced isolation outcomes.  相似文献

An amino acid sequence "fingerprint" has been derived that can be used to test if a particular sequence will fold into a beta alpha beta-unit with ADP-binding properties. It was deduced from a careful analysis of the known three-dimensional structures of ADP-binding beta alpha beta-folds. This fingerprint is in fact a set of 11 rules describing the type of amino acid that should occur at a specific position in a peptide fragment. The total length of this fingerprint varies between 29 and 31 residues. By checking against all possible sequences in a database, it appeared that every peptide, which exactly follows this fingerprint, does indeed fold into an ADP-binding beta alpha beta-unit.  相似文献
