Synthesis of enzymes involved in methylamine oxidation via methylamine dehydrogenase (MADH) is encoded by genes present in the mau cluster. Here we describe the sequence of the mauE and mauD genes from Paracoccus denitrificans as well as some properties of mauE and mauD mutants of this organism. The amino acid sequences derived from the mauE and mauD genes showed high similarity with their counterparts in related methylotrophs. Secondary structure analyses of the amino acid sequences predicted that MauE is a membrane protein with five transmembrane-spanning helices and that MauD is a soluble protein with an N-terminal hydrophobic tail. Sequence comparison of MauD proteins from different organisms showed that these proteins have a conserved motif, Cys-Pro-Xaa-Cys, which is similar to a conserved motif found in periplasmic proteins that are involved in the biosynthesis of bacterial periplasmic enzymes containing haem c and/or disulphide bonds. The mauE and mauD mutant strains were unable to grow on methylamine but they grew well on other C1-compounds. These mutants grown under MADH-inducing conditions contained normal levels of the natural electron acceptor amicyanin, but undetectable levels of the -subunit and low levels of the -subunit of MADH. It is proposed, therefore, that MauE and MauD are specifically involved in the processing, transport, and/or maturation of the -subunit and that the absence of each of these proteins leads to production of a non-functional -subunit which becomes rapidly degraded. 相似文献

An overview of the development of anti-tumor organotin derivatives, sometimes as active in vitro as doxorubicin, is presented and discussed. Solubility in water is an important issue, dominating the in vivo testing of compounds with promising in vitro properties. Several water-soluble organotin compounds gave the best in vitro activities. In a pot trial growth and transpiration of 3-year-old Douglas-fir seedlings on an acid, sandy soil was examined at a deficient

1) and an excessive level (120 kg N ha –

1) of NH4 application. Dissolved ammonium sulphate was applied to the pots weekly for two growing seasons. In half of the pots a complete
set of other nutrients was applied in optimal proportions to the applied nitrogen. Water supply was optimal and transpiration
was recorded. At the end of the second treatment season irrigation was stopped for 2 weeks during dry and sunny weather. Both
high application of NH4 and additional nutrients increased shoot growth and transpiration demand in the first treatment year. The root system was
smaller at higher N level and this reduced water uptake accordingly. In the second year the combination of high NH4
+ and additional nutrients affected root functioning predominantly due to salinity effects and this seriously decreased water
uptake capacity and shoot water potentials, finally resulting in tree death. Without addition of other nutrients the high
NH4
+ application resulted in a high degree of soil acidification, which damaged the roots, that showed a decrease in water uptake
capacity. At the low NH4 supply level soil acidification was lower, and root functioning was not affected, and the trees recovered quickly from the
imposed drought. Higher needle K and P status depressed transpiration rates at the low NH4 application rate.

Intracellular localization studies of various potyvirus proteins have been made in hope of finding clues to their function(s). Immunocytological studies localized many of the tobacco etch virus (TEV)-encoded proteins in infected cells. We used antiserum against the nonstructural P3 protein of TEV to determine the subcellular location of the P3 protein in ultrathin sections of virus-infected cells. Immunogold labeling with the antiserum showed labels associated with nucleoli, nuclei, or NIs. Absorption of antiserum with purified NIs or P3 protein resulted in no labeling. TEV NIs are known to contain a bifunctional genome-linked protein–viral proteinase (NIa–VPg) and RNA-dependent RNA polymerase (NIb). It appeared that the TEV P3 protein was a third nonstructural viral protein of NIs of TEV if the NIa–VPg is considered one protein. The presence of P3 in NIs was also supported by Western blot assays. P3 protein in the nucleolus and nucleus could indicate that it, too, is involved in early stages of viral replication. 相似文献

The effects of natural nematode communities on bacterial activity and abundance were investigated in a microcosm study. Nematodes
were added at different densities to a freshwater sediment and bacterial parameters were measured after 1, 5, 9, and 17 days.
Significant effects of nematode density on bacterial activity were noted on day 5. No long-term changes in bacterial activity
were recorded. Bacterial abundance displayed an overall decrease in both treatments and controls. In a second experiment,
the effect of nematode feeding-type on bacterial activity was studied. Microcosms were incubated with 100 individuals of a
fungus-feeding (Aphelenchus avenae) or a bacteria-feeding nematode species (Caenorhabditis elegans) respectively, and bacterial activity was determined after 0, 1, 2, 4, and 7 days. Significant time and feeding-type effects
were found, with consistently higher bacterial activity estimates in treatments with bacteria-feeding nematodes. These results
suggest that grazing affects bacterial activity, and indicate that grazing by nematodes may be more important in stimulating
bacterial activity than bioturbation or excretion. Combining these results, we conclude that natural nematode communities
may have an impact on bacterial activity, and that the magnitude of this impact depends on the proportion of actively feeding
bactivores within the community.

Alicycliphilus denitrificans strain BC and A. denitrificans strain K601(T) degrade cyclic hydrocarbons. These strains have been isolated from a mixture of wastewater treatment plant material and benzene-polluted soil and from a wastewater treatment plant, respectively, suggesting their role in bioremediation of soil and water. Although the strains are phylogenetically closely related, there are some clear physiological differences. The hydrocarbon cyclohexanol, for example, can be degraded by strain K601(T) but not by strain BC. Furthermore, both strains can use nitrate and oxygen as an electron acceptor, but only strain BC can use chlorate as electron acceptor. To better understand the nitrate and chlorate reduction mechanisms coupled to the oxidation of cyclic compounds, the genomes of A. denitrificans strains BC and K601(T) were sequenced. Barth syndrome is an X-linked genetic disorder caused by mutations in the tafazzin (taz) gene and characterized by dilated cardiomyopathy, exercise intolerance, chronic fatigue, delayed growth, and neutropenia. Tafazzin is a mitochondrial transacylase required for cardiolipin remodeling. Although tafazzin function has been studied in non-mammalian model organisms, mammalian genetic loss of function approaches have not been used. We examined the consequences of tafazzin knockdown on sarcomeric mitochondria and cardiac function in mice. Tafazzin knockdown resulted in a dramatic decrease of tetralinoleoyl cardiolipin in cardiac and skeletal muscles and accumulation of monolysocardiolipins and cardiolipin molecular species with aberrant acyl groups. Electron microscopy revealed pathological changes in mitochondria, myofibrils, and mitochondrion-associated membranes in skeletal and cardiac muscles. Echocardiography and magnetic resonance imaging revealed severe cardiac abnormalities, including left ventricular dilation, left ventricular mass reduction, and depression of fractional shortening and ejection fraction in tafazzin-deficient mice. Tafazzin knockdown mice provide the first mammalian model system for Barth syndrome in which the pathophysiological relationships between altered content of mitochondrial phospholipids, ultrastructural abnormalities, myocardial and mitochondrial dysfunction, and clinical outcome can be completely investigated. 相似文献
