mtDNA genotypes of six domestic horses (three adult short horses whose heights are under 1 m and three common domestic horses) from a small region of 15 km2 in Malipo county of Yunnan province of China were investigated by the technique of restriction fragment length polymorphism (RFLP) with 16 restriction endonucleases which recognize 6-bp sequences. An average of 56 fragments for an individual was obtained. Unlike other domestic animals, this population of horses exhibits high mtDNA genetic diversity. Each of the six horses has a specific mtDNA genotype showing a pattern of multiple maternal origins, as suggested by fossil and literature records. We think the population of horses is an amazing seed-resource pool of horses and hence deserves to be paid more attention from the view of conservation genetics. However, it is also remarkable that we did not find any typical mtDNA genetic markers which would discriminate between short horses and common domestic horses.  相似文献

在发现利用钙离子螯合剂EGTA螯合细胞外钙离子(Ca 2 )后,可以显著抑制促滤泡(激)素(FSH)刺激体外培养的颗粒细胞合成和分泌雌激素,并且该抑制作用呈剂量依赖性.假设该特异性反应是通过Ca2 影响细胞内腺苷酸环化酶(AC)发挥作用的,因为Ca 2 具有激活ACVIII的作用.通过RT-PCR和Northern印迹检测大鼠不同阶段卵巢组织中ACVIII的表达.结果表明,虽然Ca2 可以调控颗粒细胞类固醇激素的合成,但不同阶段的卵巢组织中均检测不到ACVIII的mRNA.实验间接提示了Ca2 促进颗粒细胞成熟的作用不是通过ACVIII发挥作用的,而可能是通过其他AC同工酶或其他Ca2 信号通路发挥作用.  相似文献

In this study, the feasibility of biological urea nitrogen removal in anaerobic Anammox co-culture was investigated. After 100 days of operation, complete urea nitrogen removal of 0.35 g (NH(2))(2)CO-N L(-1) d(-1) was achieved. The pure Anammox bacteria were obtained by percoll density-gradient centrifugation and found to be of incapable to hydrolyze urea. The ureolytic bacteria were isolated from the Anammox co-culture by the spread plate and streak. Comparative analysis of partial 16S rDNA sequence presented it belongs to Bacillus sp., and so named as Bacillus sp. LST-1. Fluorescence in situ hybridization was applied to identify the ratio of Bacillus sp. and Anammox in the reactor and the value was approximately 1:4. Urea nitrogen removal was realized in this autotrophic, anoxic reactor via the combined process of urea hydrolysis by Bacillus sp. LST-1 and ammonium oxidizing by Anammox. The investigation of this combined process might have an actual significance in engineering application for its low operational cost.  相似文献

Polycomb repressive complexes (PRCs) play critical roles in cell fate decisions during normal development as well as disease progression through mediating histone modifications such as H3K27me3 and H2AK119ub. How exactly PRCs recruited to chromatin remains to be fully illuminated. Here, we report that YTHDF1, the N6-methyladenine (m6A) RNA reader that was previously known to be mainly cytoplasmic, associates with RNF2, a PRC1 protein that mediates H2AK119ub in human embryonic stem cells (hESCs). A portion of YTHDF1 localizes in the nuclei and associates with RNF2/H2AK119ub on a subset of gene loci related to neural development functions. Knock-down YTHDF1 attenuates H2AK119ub modification on these genes and promotes neural differentiation in hESCs. Our findings provide a noncanonical mechanism that YTHDF1 participates in PRC1 functions in hESCs.  相似文献

Angiogenesis is essential for transplantation of mesenchymal stem cells (MSCs). Vascular endothelial growth factor (VEGF) is one of the most potent angiogenic factors identified to date. Elevated VEGF levels in MSCs correlate with the potential of MSCs transplantation. As an indirect angiogenic agent, transforming growth factor-β1 (TGF-β1) plays a pivotal role in the regulation of vasculogenesis and angiogenesis. However, the effect of TGF-β1 on VEGF synthesis in MSCs is still unknown. Besides, the intracellular signaling mechanism by which TGF-β1 stimulates this process remains poorly understood. In this article, we demonstrated that exposure of MSCs to TGF-β1 stimulated the synthesis of VEGF. Meanwhile, TGF-β1 stimulated the phosphorylation of Akt and extracellular signal-regulated kinase 1/2 (ERK1/2). Moreover, Ly 294002, a specific inhibitor of phosphatidylinositol-3-kinase (PI3K)/Akt significantly attenuated the VEGF synthesis stimulated by TGF-β1. Additionally, U0126, a specific inhibitor of ERK1/2, also significantly attenuated the TGF-β1-stimulated VEGF synthesis. These results indicated that TGF-β1 enhanced VEGF synthesis in MSCs, and the Akt and ERK1/2 activation were involved in this process.  相似文献

Circulating soluble adhesion molecules have been suggested as useful markers to predict several clinical conditions such as atherosclerosis, type 2 diabetes, obesity, and hypertension. To determine genetic factors influencing plasma levels of soluble vascular cell adhesion molecule-1 (VCAM-1) and P-selectin, quantitative trait locus (QTL) analysis was performed on an intercross between C57BL/6J (B6) and C3H/HeJ (C3H) mouse strains deficient in apolipoprotein E-deficient (apoE−/−). Female F2 mice were fed a western diet for 12 weeks. One significant QTL, named sVcam1 (71 cM, LOD 3.9), on chromosome 9 and three suggestive QTLs on chromosomes 5, 13 and 15 were identified to affect soluble VCAM-1 levels. Soluble P-selectin levels were controlled by one significant QTL, named sSelp1 (8.5 cM, LOD 3.4), on chromosome 16 and two suggestive QTLs on chromosomes 10 and 13. Both adhesion molecules showed significant or an apparent trend of correlations with body weight, total cholesterol, and LDL/VLDL cholesterol levels in the F2 population. These results indicate that plasma VCAM-1 and P-selectin levels are complex traits regulated by multiple genes, and this regulation is conferred, at least partially, by acting on body weight and lipid metabolism in hyperlipidemic apoE−/− mice. The Chinese species of the subgenus Microchelonus Szépligeti of the genus Chelonus Panzer with the female carapace having an incised apex in dorsal view and/or in posterior view are revised. Two new species, Chelonus (Microchelonus) rhagius sp. n. and Chelonus (Microchelonus) tobiasi sp. n. are described. Chelonus (Microchelonus) elegantulus Tobias and Chelonus (Microchelonus) volgensis Tobias are recorded in China for the first time.  相似文献

Influence of ladder concentration of nickel (Ni) on the leaves of Hydrocharis dubia were studied after 3 days treatment. The accumulation of Ni, the content of polyamines, proline, malondialdehyde (MDA) and soluble protein, as well as the activities of superoxide dismutase (SOD), catalase (CAT) and peroxidase (POD) in the leaves were investigated. The result indicated that the toxicity of Ni manifested in respective aspect of physiological and biochemical characters. Significant increase of Ni concentration in the leaf tissue was observed, which was concentration dependent. Visible symptoms of Ni toxicity: chlorosis and necrosis occurred following the 3rd day. Meantime, treatment with Ni resulted in the increase in the generation rate of O2•− in the leaves. SOD and CAT activities decreased significantly in response to Ni treatment, it was possibly the reason of accumulation of O2•−. However, a several-fold decrease in POD activities was found. Our results indicated that because of prolonged increases in O2•− level, oxidative damage, measured as the level of lipid peroxidation, occured in the leaves of Ni treated fronds. The changes of the content of polyamines (PAs) were also investigated in the leaves of Hydrocharis dubia. Ni treatment significantly increased the putrescine (Put) level and lowered spermidine (Spd) and spermine (Spm) levels, thereby significantly reducing the ratio of free (Spd + Spm)/Put in leaves, which has been considered as the signal under stress. Although the trend that PS-conjugated PAs and PIS-bound PAs changed the same as free PAs, they changed in more less extent.  相似文献

The carrying capacity of food resources for migrating shorebirds was estimated at a stopover site in the Yangtze River Estuary during the two migratory seasons (spring and autumn). From March to May and September to November 2005, the macrobenthos resources of the Jiuduansha Wetland were investigated, and most of the macrobenthos species in the newly-formed shoal were found to be appropriate food for shorebirds. Biomass measurements showed that the total food resource was about 4541.20 kg AFDW (Ash-Free Dry Weight) in spring and about 2279.64 kg AFDW in autumn. Calculations were also done in the available habitats (intertidal bare mudflat and Scirpus × mariqueter/Scirpus triqueter zones) for the shorebirds. The food resources in the available areas were about 3429.03 kg AFDW in spring and about 1700.92 kg AFDW in autumn. Based on the classification (by lean weight, basic metabolic rate and body length) of the shorebird community, and using the energy depletion model, it was theorized that all of the food resources in the Jiuduansha Wetland could support about 3.5 million shorebirds during spring season and 1.75 million shorebirds during autumn season. The shorebird carrying capacities in terms of the available food were about 2.6 million and 1.3 million birds during the two respective migration seasons. Considering the effect of intake rate, the potential carrying capacity was about 0.13–0.26 million shorebirds in the study area. The main factor restricting use of the area by shorebirds was the scarcity of available habitats for roosting at high tide rather than availability of food supply. The migration of osteoclasts (OCs) from circulation and bone marrow into bone surface plays a critical role in the pathogenesis of some bone resorptive diseases, such as rheumatoid arthritis and osteoporosis. To date, how the migration of OCs remains unclear. We investigated gene expression profiling in osteoclastic differentiation of bone marrow–derived macrophages (BMMs) into OCs by microarray analysis. We identified 387 genes overexpressed in osteoclastic differentiation of BMMs. Among them, chemokine CCL4 showed a robust up‐regulation signal. High expression of CCL4 was validated in primary BMMs and OC precursor cell line RAW264.7 during differentiation into OCs. The CCL4 neutralization decreased RANKL‐induced OC precursor cell migration and invasion in Matrigel‐coated transwell membranes assay and in vitro wound healing assay. However, CCL4 inhibition did not affect OCs differentiation and differentiation associated gene expression. The CCL4 inhibition promoted the PI3K phosphorylation at 45 to 60 minutes after RANKL stimulation in RAW264.7. This study indicated that chemokine CCL4 is an important regulator for OCs migration via PI3K pathway, providing a novel therapy target for bone resorptive diseases.  相似文献
