Silencing of tumor suppressor genes (TSGs) by DNA promoter hypermethylation is an early event in carcinogenesis and a potential target for personalized cancer treatment. In head and neck cancer, little is known about the role of promoter hypermethylation in survival. Using methylation specific multiplex ligation-dependent probe amplification (MS-MLPA) we investigated the role of promoter hypermethylation of 24 well-described genes (some of which are classic TSGs), which are frequently methylated in different cancer types, in 166 HPV-negative early oral squamous cell carcinomas (OSCC), and 51 HPV-negative early oropharyngeal squamous cell carcinomas (OPSCC) in relation to clinicopathological features and survival. Early OSCC showed frequent promoter hypermethylation in RARB (31% of cases), CHFR (20%), CDH13 (13%), DAPK1 (12%), and APC (10%). More hypermethylation (≥ 2 genes) independently correlated with improved disease specific survival (hazard ratio 0.17, P = 0.014) in early OSCC and could therefore be used as prognostic biomarker. Early OPSCCs showed more hypermethylation of CDH13 (58%), TP73 (14%), and total hypermethylated genes. Hypermethylation of two or more genes has a significantly different effect on survival in OPSCC compared with OSCC, with a trend toward worse instead of better survival. This could have a biological explanation, which deserves further investigation and could possibly lead to more stratified treatment in the future. 相似文献

The reproductive system of the important basal gastrotrich Neodasys chaetonotoideus is described and reconstructed on the basis of light microscopy, serial ultrathin sections (ultrastructure) and scanning electron microscopy. Starting frontally, the hermaphroditic reproductive system consists of paired and tube shaped lateral testes that do not possess elongated seminal ducts but most likely open directly via paired ventral pores. The unpaired, medio‐dorsal ovary region contains early oogenic stages that mature caudally towards the uterus region, where the most mature egg is positioned laterally to the midgut. The ovary region is not covered with an epithelial lining whereas the uterus region possesses a distinct epithelial wall. Between ovary and uterus region, we have detected a conspicuous section of the female gonad, the vitellogenic oviduct that consists of a thick epithelial wall which forms cellular protuberances into the developing oocytes passing the oviduct. We interpret this as a special, hitherto undescribed mode of vitellogenesis in Gastrotricha. Further caudally, the uterus continues with the fronto‐caudal organ, a complex of two substructures that are apparently homologous to the frontal organ and the caudal organ of many species of the Gastrotricha Macrodasyida. Neodasys chaetonotoideus obviously engages in spermatophore formation and transfer. In this study we develop a morpho‐functional scenario for the gonads and accessory organs in terms of spermatophore production, exchange and oviposition. We compare our newly obtained data with already published results on the reproductive organs of several species of Gastrotricha by means of a species‐character matrix and provide a computer aided evaluation by a parsimonious character optimization. A reconstruction of the reproductive system of the stem species of Gastrotricha on the basis of three recent phylogenetic analyses is presented. These reconstructions give support for a Neodasys‐like reproductive system in the ground pattern of Gastrotricha with slight morphological differences and direct transfer of spermatozoa rather than spermatophore transfer. The evolution of selected characters is traced thus revealing some incidents of convergent evolution as well as the evolutionary replacement of the ancestral frontal organ by the derived frontal sac in at least two separated lineages. 相似文献

Human colon cancer harbors a small subfraction of tumor-initiating cells (TICs) that is assumed to be a functionally homogeneous stem-cell-like population driving tumor maintenance and metastasis formation. We found unexpected cellular heterogeneity within the TIC compartment, which contains three types of TICs. Extensively self-renewing long-term TICs (LT-TICs) maintained tumor formation in serial xenotransplants. Tumor transient amplifying cells (T-TACs) with limited or no self-renewal capacity contributed to tumor formation only in primary mice. Rare delayed contributing TICs (DC-TICs) were exclusively active in secondary or tertiary mice. Bone marrow was identified as an important reservoir of LT-TICs. Metastasis formation was almost exclusively driven by self-renewing LT-TICs. Our results demonstrate that tumor initiation, self-renewal, and metastasis formation are limited to particular subpopulations of TICs in primary human colon cancer. We identify LT-TICs as a quantifiable target for therapies aimed toward eradication of self-renewing tumorigenic and metastatic colon cancer cells. 相似文献

Abdominal obesity is characterized by sympathetic nerve activation (SNA), probably mediated by elevated insulin and leptin levels. Resting heart rate (RHR) is a marker of sympathetic tone, and independently associated with cardiovascular events and death in various populations. We investigated and quantified the relation between visceral adipose tissue (VAT) and RHR in patients with vascular disease. In 3,723 patients with manifest vascular disease, visceral and subcutaneous fat tissue was measured with ultrasonography. RHR was obtained from an electrocardiogram (ECG). The association between quartiles of VAT and RHR was quantified using linear regression analysis with adjustments for potential confounding factors. Separate analyses were performed for men and women and for location of vascular disease. Visceral fat was categorized into sex-pooled quartiles (Q) ranging from 2.7-8.0 cm in Q1 (reference) to 9.4-20.6 cm in Q4. High visceral fat thickness was associated with increased RHR, in men (Q4 vs. Q1, β = 4.36; 95% confidence interval (CI) = 3.11-5.61) and women (β = 1.48; 95% CI = -0.70 to 3.66), after full adjustment. Waist circumference and BMI had a significant relation with RHR in men (β = 3.51; 95% CI = 2.21-4.81 and β = 2.80; 95% CI = 1.51-4.08, respectively) but these relations were smaller and not significant in women (β = 0.71; 95% CI = -1.44 to 2.85 and β = 0.24; 95% CI = -1.90 to 2.37, respectively). There was no relation between subcutaneous fat and RHR in men and women. The relation between visceral fat and RHR was similar in patients with different locations of vascular diseases. Increased visceral fat is associated with increased RHR in male and female patients with vascular disease, independent of the location. 相似文献

The study contributes to the discussion of mastax evolution within Rotifera by giving an insight into the ultrastructure of the mastax in the rotifer species Filinia longiseta (Flosculariacea) and additionally into the bdelloid rotifer species Adineta vaga and Zelinkiella synaptae. The existence of cuticularized jaw elements (trophi) in the mastax, a muscular pharynx, is one of the defining rotiferan characters and the basis on which the monophyletic taxon Gnathifera Ahlrichs 1995a, comprising Rotifera, Gnathostomulida, Micrognathozoa and Acanthocephala, was erected. By means of SEM observations of the trophi and ultrathin serial sections (TEM) of the mastax, the internal and external organization of the jaw elements of F. longiseta is reconstructed. TEM sections of the incus of Filinia demonstrate that the fulcrum and the rami are built up by multitudes of tiny cuticular tubes. While tubular substructures in the rotiferan fulcrum have been described previously, distinct cuticular tubes as a substructure of the ramus have only been described for species belonging to the taxa Seisonidea and Bdelloidea so far (
[Koehler and Hayes, 1969] and [Ahlrichs, 1995b]). By comparing the appearance and arrangement of the cuticular tubes in the rami of F. longiseta to those found in species of Seisonidea and Bdelloidea, a higher degree of resemblance between the structures in F. longiseta and Bdelloidea can be reported. The occurrence of the ramus substructures in species of Seisonidea (Paraseison annulatus and Seison nebaliae) is given consideration to represent an intermediate between the ramus substructure of Bdelloidea/Flosculariacea and Ploima. Additionally, the mastax musculature of F. longiseta, being associated with the trophi, is described: A total of seven muscles are found that directly insert the jaw elements or are indirectly associated with them via muscle-to-muscle connections. 相似文献

The ribbon complex of retinal photoreceptor synapses represents a specialization of the cytomatrix at the active zone (CAZ) present at conventional synapses. In mice deficient for the CAZ protein Bassoon, ribbons are not anchored to the presynaptic membrane but float freely in the cytoplasm. Exploiting this phenotype, we dissected the molecular structure of the photoreceptor ribbon complex. Identifiable CAZ proteins segregate into two compartments at the ribbon: a ribbon-associated compartment including Piccolo, RIBEYE, CtBP1/BARS, RIM1, and the motor protein KIF3A, and an active zone compartment including RIM2, Munc13-1, a Ca2+ channel alpha1 subunit, and ERC2/CAST1. A direct interaction between the ribbon-specific protein RIBEYE and Bassoon seems to link the two compartments and is responsible for the physical integrity of the photoreceptor ribbon complex. Finally, we found the RIBEYE homologue CtBP1 at ribbon and conventional synapses, suggesting a novel role for the CtBP/BARS family in the molecular assembly and function of central nervous system synapses. 相似文献

A significant part of global primary productivity is provided by cyanobacteria, which are abundant in most marine and freshwater habitats. In many oceanographic regions, however, the concentration of iron can be so low that it limits growth. Cyanobacteria respond to this condition by expressing a number of iron stress inducible genes, of which the isiA gene encodes a chlorophyll-binding protein known as IsiA or CP43'. It was recently shown that 18 IsiA proteins encircle trimeric photosystem I (PSI) under iron-deficient growth conditions. We report here that after prolonged growth of Synechocystis PCC 6803 in an iron-deficient medium, the number of bound IsiA proteins can be much higher than previously known. The largest complexes bind 12-14 units in an inner ring and 19-21 units in an outer ring around a PSI monomer. Fluorescence excitation spectra indicate an efficient light harvesting function for all PSI-bound chlorophylls. We also find that IsiA accumulates in cyanobacteria in excess of what is needed for functional light harvesting by PSI, and that a significant part of IsiA builds supercomplexes without PSI. Because the further decline of PSI makes photosystem II (PSII) increasingly vulnerable to photooxidation, we postulate that the surplus synthesis of IsiA shields PSII from excess light. We suggest that IsiA plays a surprisingly versatile role in cyanobacteria, by significantly enhancing the light harvesting ability of PSI and providing photoprotection for PSII. 相似文献

The studies concern the phenotypic variability, density, distribution in space, dry matter production and seed production in relation to some habitat conditions.
