A series of cationic nickel complexes [(η3-methally)Ni(PP(O))]SbF6 (1–4) [PP(O) = Ph2P(CH2)P(O)Ph2 (dppmO) (1), Ph2P(CH2)2P(O)Ph2 (dppeO) (2), Ph2P(CH2)3P(O)Ph2 (dpppO) (3), pTol2P(CH2)P(O)pTol2 (dtolpmO) (4)] has been synthesized in good yields by treatment of [(η3-methally)NiBr]2 with biphosphine monoxides and AgSbF6. The ligands are coordinated in a bidentate way. Starting from [(η3-all)PdI]2 the cationic complexes [(η3-all)PP(O))]Y (8–14). [PP(O) = dppmO, dppeO, dpppO, dtolpmO;Y = BF4, SbF6, CF3SO3, pTolSO3] were synthesized in good yields. The coordination mode of the ligand is dependent on the backbone and the anion, revealing a monodentate coordination with dppmO for stronger coordinating anions. The intermediates [(η3-all)Pd(I)(PP(O)-κ1-P)] (5–7) [PP(O) = dppmO (5), dppeO (6), dtolpmO (7)] were isolated and characterized. Neutral methyl complexes [(Cl)(Me)Pd(PP(O))] (15–18). [PP(O) = dppmO (15), dppeO (16), dpppO (17), dtolpmO (18)] can easily be obtained in high yields starting from [(cod)PdCl2]. For dppmO two different routes are presented. The structure of [(Me)(Cl)Pd{;Ph2P(CH2-P(O)Ph22-P,O};] · CH2Cl2 (15) with the chlorine atom trans to phosphorus was determined by X-ray diffraction.  相似文献

Leukemic cells of a 20 year old patient, suffering from acute lymphoblastic leukemia, were characterized by surface marker and functional analysis. A significant cell population within this type of leukemia expresses concomitantly the CD4 and CD8 antigen on the same cell and might represent a new differentiation stage of T-cells with the gamma/delta receptor. The leukemic cells show a distinct pattern of growth response to mitogens and lymphokines, which might correlate to their differentiation stage. Moreover, a "natural killer"-like activity can be induced in these cells by IL-2.  相似文献

Transgenic soybean (Glycine max [L.] Merr.) plants were regenerated from calli derived from protoplasts electroporated with plasmid DNA-carrying genes for a selectable marker, neomycin phosphotransferase (NPTII), under the control of the cauliflower mosaic virus 35-Svedberg unit promoter, linked with a nonselectable mannityl opine synthesis marker. Following electroporation and culture, the protoplast-derived colonies were subjected to kanamycin selection (50 micrograms per milliliter) beginning on day 15 for 6 weeks. Approximately, 370 to 460 resistant colonies were recovered from 1 × 106 electroporated protoplasts, giving an absolute transformation frequency of 3.7 to 4.6 × 10−4. More than 80% of the kanamycin-resistant colonies showed NPTII activity, and about 90% of these also synthesized opines. This indicates that the linked marker genes were co-introduced and co-expressed at a very high frequency. Plants were regenerated from the transformed cell lines. Southern blot analysis of the transformed callus and leaf DNA demonstrated the integration of both genes. Single-plant assays performed with different plant parts showed that both shoot and root tissues express NPTII activity and accumulate opines. Experiments with NPTII and mannityl opine synthesis marker genes on separate plasmids resulted in a co-expression rate of 66%. These results indicate that electroporation can be used to introduce both linked and unlinked genes into the soybean to produce transformed plants.  相似文献

The function of the eyespot in phototaxis of the flagellate green alga Chlamydomonas reinhardtii Dangeard was studied using quantitative reflection confocal laser scanning microscopy and photoelectric measurements. The reflective properties of the eyespot and the photoreceptor current of the C. reinhardtii eyespot mutant ey 627, mt were compared with those of Chlamydomonas strains possessing a well-developed eyespot. Under growth conditions in which strongly disorganized eyespots were observed in the mutant by electron microscopy, there was a significant reduction in the reflection intensity of the eyespot and in the amplitude ratio (500440 nm) of photoreceptor currents induced by flashes of 500- and 440-nm light in non-oriented cells. Photoelectrical responses of pre-oriented cells revealed that the latter effect is caused by an altered directional sensitivity of the antenna complex, whereas the functional state of the photoreceptor pigment is not strongly affected in mutant cells. Both the reflection intensity and the amplitude ratio of photoreceptor currents increased to the level of reference strains under conditions supporting the development of a well-organized eyespot in the mutant. Furthermore, incubation of the mutant with high concentrations of all-trans-retinal (10 M), independent of whether carotenoid biosynthesis was inhibited or not, was found to increase the reflection intensity of the eyespot. An increase in the rate of photoorientation of the mutant occurred concomitant with the increase in the reflective properties of the mutant eyespot. These observations demonstrate the importance of an intact eyespot for interference reflection and absorption of phototactically active light, and thus for the directional sensitivity of the eyespot apparatus.Abbreviations HSM high-salt medium This study was supported by the Deutsche Forschungsgemeinschaft. Summary Dynamic activities of the endoplasmic reticulum (ER) and of the cell surface were analyzed in living epithelial cells (pinacocytes) ofSpongilla lacustris by contrast-enhanced video microscopy with the AVEC-DIC or the ACE equipment. Long term sequences revealed the ER to be a highly unstable system undergoing permanent alterations of the reticular patterns in that tubules merge and split or polygons open and close again. Treatment with colcemid or colchicine causes distinct changes of the typical motile phenomena, whereas cytochalasin D has no influence. On the other hand, the dynamic behavior of the cell surface is characterized by distinct ruffling activities as well as the formation and retraction of spiky filopodia. In contrast to the described ER dynamics, cell surface phenomena are clearly influenced by cytochalasin D but not by colcemid or colchicine. Altogether, results of the present paper are similar to correspondent observations on mammalian cells and point to microtubules and microfilaments as the cytoskeletal elements being responsible for ER and cell surface dynamics, respectively.  相似文献

The action of ATP and its analogs as well as the effects of alkali ions were studied in their action on the ouabain receptor. One single ouabain receptor with a dissociation constant (KD) of 13 nM was found in the presence of (Mg2+ + Pi) and (Na+ + Mg2+ + ATP). pH changes below pH 7.4 did not affect the ouabain receptor. Ouabain binding required Mg2+, where a curved line in the Scatchard plot appeared. The affinity of the receptor for ouabain was decreased by K+ and its congeners, by Na+ in the presence of (Mg2+ + Pi), and by ATP analogs (ADP-C-P, ATP-OCH3). Ca2+ antagonized the action of K+ on ouabain binding. It was concluded that the ouabain receptor exists in a low affinity (Rα) and a high affinity conformational state (Rβ). The equilibrium between both states is influenced by ligands of (Na+ + K+)-ATPase. With 3 mM Mg2+ a mixture between both conformational states is assumed to exist (curved line in the Scatchard plot).  相似文献

[14C]ADP binding to EDTA-washed ox brain cell membranes was increased by Na+, but decreased by K+, Mg2+ and Ca2+. Na+ abolished the effect of K+ on ADP binding by a competitive mechanism, but could not reverse the inhibitory action of Mg2+ and Ca2+. It is concluded that the cation-induced changes in ADP binding reflect properties of (Na+ + K+)-activated ATPase.  相似文献
