The growth of Rio Bravo virus (RBV) in eight cell culture systems was studied. Highest yields of virus were produced in BHK-21 (C13), L, and Vero cell lines, but L cells were resistant to low doses of virus. LLC-MK(2), HeLa, and human embryo skin cells produced moderate amounts of virus, but FL amnion and primary chick embryo fibroblasts supported little virus growth. Virus was rapidly inactivated by exposure to pH values below 7.0. Single-cycle growth in BHK-21, L, and LLC-MK(2) cell monolayers was characterized by a latent period of about 12 hr followed by rapid virus production that peaked at 36 to 48 hr. Vero cell cultures can remain chronically infected with RBV for more than 100 days. Such cultures show evidence of cell destruction, and their supernatant fluids contain virus at 10(4) to 10(5) log(10) per ml. 相似文献

We describe a few mathematical tools which allow to investigate whether air-water interfaces exist(under prescribedconditions)and are mechanically stable and temporally persistent.In terms of physics,air-water interfaces are governed by theYoung-Laplace equation.Mathematically they are surfaces of constant mean curvature which represent solutions of a nonlinearelliptic partial differential equation.Although explicit solutions of this equation can be obtained only in very special cases,it is-under moderately special circumstances-possible to establish the existence of a solution without actually solving thedifferential equation.We also derive criteria for mechanical stability and temporal persistence of an air layer.Furthermorewe calculate the lifetime of a non-persistent air layer.Finally,we apply these tools to two examples which exhibit the symmetriesof 2D lattices.These examples can be viewed as abstractions of the biological model represented by the aquatic fernSalvinia. 相似文献

Giardia intestinalis is a ubiquitous parasitic protist that is the causative agent of giardiasis, one of the most common protozoan diarrheal diseases in the world. Giardia trophozoites attach to the intestinal epithelium using a specialized and elaborate microtubule structure, the ventral disc. Surrounding the ventral disc is a less characterized putatively contractile structure, the lateral crest, which forms a continuous perimeter seal with the substrate. A better understanding of ventral disc and lateral crest structure, conformational dynamics, and biogenesis is critical for understanding the mechanism of giardial attachment to the host. To determine the components comprising the ventral disc and lateral crest, we used shotgun proteomics to identify proteins in a preparation of isolated ventral discs. Candidate disc-associated proteins, or DAPs, were GFP-tagged using a ligation-independent high-throughput cloning method. Based on disc localization, we identified eighteen novel DAPs, which more than doubles the number of known disc-associated proteins. Ten of the novel DAPs are associated with the lateral crest or outer edge of the disc, and are the first confirmed components of this structure. Using Fluorescence Recovery After Photobleaching (FRAP) with representative novel DAP::GFP strains we found that the newly identified DAPs tested did not recover after photobleaching and are therefore structural components of the ventral disc or lateral crest. Functional analyses of the novel DAPs will be central toward understanding the mechanism of ventral disc-mediated attachment and the mechanism of disc biogenesis during cell division. Since attachment of Giardia to the intestine via the ventral disc is essential for pathogenesis, it is possible that some proteins comprising the disc could be potential drug targets if their loss or disruption interfered with disc biogenesis or function, preventing attachment. 相似文献

Because of the relative impermeability of the blood‐brain barrier (BBB), many drugs are unable to reach the CNS in therapeutically relevant concentration. One method to deliver drugs to the CNS is the osmotic opening of the BBB using mannitol. Hyperosmotic mannitol induces a strong phosphorylation on tyrosine residues in a broad spectrum of proteins in cerebral endothelial cells, the principal components of the BBB. Previously, we have shown that among targets of tyrosine phosphorylation are β‐catenin, extracellular signal‐regulated kinase 1/2 and the non‐receptor tyrosine kinase Src. The aim of this study was to identify new signalling pathways activated by hypertonicity in cerebral endothelial cells. Using an antibody array and immunoprecipitation we identified the receptor tyrosine kinase Axl to become tyrosine phosphorylated in response to hyperosmotic mannitol. Besides activation, Axl was also cleaved in response to osmotic stress. Degradation of Axl proved to be metalloproteinase‐ and proteasome‐dependent and resulted in 50–55 kDa C‐terminal products which remained phosphorylated even after degradation. Specific knockdown of Axl increased the rate of apoptosis in hyperosmotic mannitol‐treated cells; therefore, we assume that activation of Axl may be a protective mechanism against hypertonicity‐induced apoptosis. The nutritive value of whole crop forage maize is influenced by the proportion of ears and stover in the whole crop and by the nutrient composition and digestibility characteristics of the plant parts. An experiment investigating the impact of variety, harvest date and year on the nutritive value of ensiled maize ears was carried out in three consecutive years (2007, 2008 and 2010). Nine different maize varieties were harvested at three different maturity stages (50, 55 and 60% dry matter (DM) content in the ears). After harvest, ears and stover were ensiled separately and afterwards nutrient composition and ruminal nutrient degradability (organic matter (OM), crude protein (CP) and non-fibre carbohydrates (NFC)) were analysed. Variety had a significant influence on content of CP and effective ruminal degradability (ED) of OM at low passage rates, whereas ED of CP and NFC was not affected by variety. In contrast, harvest date and year significantly influenced nutrient composition and ruminal degradability of ensiled maize ears. The content of NFC increased and the content of fibre components as well as ED of OM, CP and NFC declined with processing maturity of the maize plants. At a passage rate of 5% h?1, ED of OM declined from 75.9% to 68.4%, ED of CP from 82.5% to 73.8% and ED of NFC from 88.0% to 82.3% between the early and late harvest date. The results of this study indicate that the nutrient composition and ruminal degradability of ensiled maize ears are affected mainly by maturity stage at harvest and by year, whereas variety has only little influence. 相似文献

Summary Neuron populations in the retina of the toad, Bufo marinus, were labelled with a monoclonal antibody raised against microtubule-associated protein 2 (MAP2). A subpopulation of cones, probably corresponding to the blue-sensitive small single cones, large diameter amacrine cells in the most proximal row of the inner nuclear layer and some large ganglion cells in the ganglion cell layer were labelled. Double labelling experiments were carried out to establish the colocalisation of MAP2 with known putative transmitter substances of the anuran amacrine cells. MAP2 was colocalised in a subpopulation of serotonin-immunoreactive and in all tyrosine hydroxylase-immunoreactive amacrine cells.
