The transition from G1 to S phase of the cell cycle in Saccharomyces cerevisiae requires the activity of the Ubc3 (Cdc34) ubiquitin-conjugating enzyme. S. cerevisiae cells lacking a functional UBC3 (CDC34) gene are able to execute the Start function that initiates the cell cycle but fail to form a mitotic spindle or enter S phase. The Ubc3 (Cdc34) enzyme has previously been shown to catalyze the attachment of multiple ubiquitin molecules to model substrates, suggesting that the role of this enzyme in cell cycle progression depends on its targeting an endogenous protein(s) for degradation. In this report, we demonstrate that the Ubc3 (Cdc34) protein is itself a substrate for both ubiquitination and phosphorylation. Immunochemical localization of the gene product to the nucleus renders it likely that the relevant substrates similarly reside within the nucleus.  相似文献

The genome of Drosophila bifasciata harbours two distinct subfamilies of P-homologous sequences, designated M-type and O-type elements based on similarities to P element sequences from other species. Both subfamilies have some general features in common: they are of similar length (M-type: 2935 bp, O-type: 2986 bp), are flanked by direct repeats of 8 by (the presumptive target sequence), contain terminal inverted repeats, and have a coding region consisting of four exons. The splice sites are at homologous positions and the exons have the coding capacity for proteins of 753 amino acids (M-type) and 757 amino acids (O-type). It seems likely that both types of element represent functional transposons. The nucleotide divergence of the two P element subfamilies is high (31%). The main structural difference is observed in the terminal inverted repeats. Whereas the termini of M-type elements consist of 31 by inverted repeats, the inverted repeats of the O-type elements are interrupted by non-complementary stretches of DNA, 12 by at the 5 end and 14 by at the 3 end. This peculiarity is shared by all members of the O-type subfamily. Comparison with other P element sequences indicates incongruities between the phylogenies of the species and the P transposons. M-type and O-type elements apparently have no common origin in the D. bifasciata lineage. The M-type sequence seems to be most closely related to the P element from Scaptomyza pallida and thus could be considered as a more recent invader of the D. bifasciata gene pool. The origin of the O-type elements cannot be unequivocally deduced from the present data. The sequence comparison also provides new insights into conserved domains with possible implications for the function of P transposons.  相似文献

Abstract Three layers of different electron density can be distinguished in the periostracum. Periostracal units of up to 900 nm length are merged into the outer fibrous layer and binding of gold-labelled lectin-WGA indicates the presence of chitin because it is labile to chitinase treatment. The periostracum is formed by the epithelia of the groove and the belt at the mantle edge. The distal and basal epithelium of the groove consists mainly of type A cells with an extended Golgi apparatus and apical vesicles. The presence of peroxidase and phenol oxidase indicates a function in tanning of the periostracum. In the proximal epithelium of the groove, type B cells with protruding apices add more material for periostracum formation. Type C cells secrete single periostracal units which are formed within single vesicles or larger vacuoles. Type D cells secrete electron-dense vesicles which also contain WGA-positive material. The distal cells of the belt are characterized by predominating strands of the rER while subapical vacuoles, to some of which WGA binds, dominate in the cells of the central part. In the belt, phenol oxidase and peroxidase can be localized in cisternae of the rER and the Golgi apparatus. Numerous control incubations indicate that, indeed, two different enzymes are localized.  相似文献

Species identification is pivotal in biodiversity assessments and proteomic fingerprinting by MALDI-TOF mass spectrometry has already been shown to reliably identify calanoid copepods to species level. However, MALDI-TOF data may contain more information beyond mere species identification. In this study, we investigated different ontogenetic stages (copepodids C1–C6 females) of three co-occurring Calanus species from the Arctic Fram Strait, which cannot be identified to species level based on morphological characters alone. Differentiation of the three species based on mass spectrometry data was without any error. In addition, a clear stage-specific signal was detected in all species, supported by clustering approaches as well as machine learning using Random Forest. More complex mass spectra in later ontogenetic stages as well as relative intensities of certain mass peaks were found as the main drivers of stage distinction in these species. Through a dilution series, we were able to show that this did not result from the higher amount of biomass that was used in tissue processing of the larger stages. Finally, the data were tested in a simulation for application in a real biodiversity assessment by using Random Forest for stage classification of specimens absent from the training data. This resulted in a successful stage-identification rate of almost 90%, making proteomic fingerprinting a promising tool to investigate polewards shifts of Atlantic Calanus species and, in general, to assess stage compositions in biodiversity assessments of Calanoida, which can be notoriously difficult using conventional identification methods.  相似文献

Zusammenfassung Als Hauptergebnis der Ameisenversuche, die damit auch ein allgemeines Interesse verdienen, läßt sich folgendes feststellen: Es ist möglich, in Kunstnestern der dafür besonders geeigneten Gattung Pheidole durch experimentelle Methoden Soldatenentwicklung künstlich hervorzurufen, d.h. die Entwicklung der jungen Larven so zu beeinflussen, daß nicht, wie gewöhnlich, kleine Arbeiterinnen entstehen, sondern größere, kräftigere, widerstandsfähigere Tiere, die sich außerdem durch eine längere Lebensdauer auszeichnen. Man kann aber auch in natürlichen, ganz unbeeinflußten Pheidole-Nestern die Zahl solcher Soldaten auf das 4- und 5fache erhöhen, wenn man den Nestern ein bestimmtes Futter vorlegt. Die Möglichkeiten zur Soldatenentstehung sind bei Pheidole im Ei vererbungsmäßig festgelegt; sie verwirklichen sich aber nur unter gewissen Bedingungen. Ausschlaggebend ist dabei ein besonderer Wirkstoff, der Faktor T. Er konnte bisher durch keinen anderen Biokatalysator ersetzt werden, und ebensowenig durch bisher angewandte Gemische von Vitaminen und Aminosäuren-Cholin, Carnitin und Desoxyribonucleinsäure. Dies gilt nicht nur für die Beobachtungen und Versuche an Ameisen selbst, sondern auch für die zu Modellexperimentell herangezogenen anderen Tiere, insbesondere Drosophila melanogaster. Dieser Faktor T hat nach allen vorliegenden Erfahrungen bis hinauf zum Menschen in bisher noch nicht absteckbaren Grenzen die Fähigkeit, andere Wirkstoffe zu mobilisieren und damit vorhandene Reserven herauszuholen. Die Ameisengattung Pheidole, die schon bei der Auffindung dieses Faktors eine bedeutende Eolle spielte, ist durch ihre Soldaten-Kaste bis jetzt das eindrucksvollste Beispiel dafür, wie durch den Faktor T schlummerende, nicht genützte biologische Möglichkeiten verwirklicht werden können.Herrn Prof. Dr. K. von Frisch zu seinem 70. Five clones, containing polymorphic CA-repeat sequences, have been isolated from a specific human chromosome 21 phage library and have been localised to band q21 of chromosome 21 using a somatic cell hybrid panel. These highly repetitive sequences (D21S1263, D21S1264, D21S1415, D21S1417 and D21S1420) have been characterised in the CEPH reference parents and have heterozygosities ranging from 0.30 to 0.81 and an average polymorphism information content (PIC) of 0.62. The relative order of these markers, based on the somatic cell hybrid panel, is cen-D21S1417, D21S1420-D21S1263, D21S1415-D21S1264-tel. The most polymorphic marker (D21S1264) has been included in the chromosome 21 genetic map. They have also been localised in the CEPH/ Généthon YAC panel, providing a refined localisation of these polymorphic sequences. These five CA-repeat markers should provide a better characterisation of the q21 region of chromosome 21.  相似文献

A fragment of Bacillus subtilis DNA coding for xylose isomerase and xylulokinase was isolated from a BamHI restriction pool by complementation of an isomerase-defective Escherichia coli strain. The spontaneous insertion of IS5, which occurred during the very slow growth of the E. coli xyl- cells on xylose, allowed the expression of the cloned Bacillus genes in E. coli. Without IS5 insertion, the xylose genes were inactive in E. coli. Deletion experiments indicated that the control of the expression resides within a 270-bp long region at the right end of IS5. Deletion of this region led to a loss of expression, which could be restored by insertion of the lacUV5 promoter fragment at the deletion site. Sequence analysis showed that the site of IS5 insertion is 195 bp upstream from the putative ATG initiation codon of the xylose isomerase structural gene. This ATG is preceded by a ribosome binding sequence and two hexamers also found in promoter regions of other Bacillus genes. Deletion and mutagenesis analysis led to a preliminary map of the Bacillus xylose operon.  相似文献

Forty–nine species of myxomycetes are reported as new to Norway. Of these, 22 species are new to Scandinavia. Several have not, or only rarely, been reported from Europe. Two species (viz. Diderma subfloriformis and Comatricha anomala) were previously known only from their type localities. Brief discussions are given for some of the more interesting species.  相似文献

The binding of the natural and unnatural diastereoisomers 25-hydroxyvitamin D3-26,23-lactone and 1,25 dihydroxyvitamin D3-26,23-lactone to the vitamin D-binding protein (DBP) and 1,25 dihydroxyvitamin D3 [1,25(OH)2D3] chick intestinal receptor have been investigated. Also, the biological activities, under in vivo conditions, of these compounds, in terms of intestinal calcium absorption (ICA) and bone calcium mobilization (BCM), in the chick are reported. The presence of the lactone ring in the C23-C26 position of the seco-steroid side chain increased two to three times the ability of both 25(OH)D3 and 1,25(OH)2D3 to displace 25(OH)[3H]D3 from the D-binding protein; however, the DBP could not distinguish between the various diastereoisomers. In contrast, the unnatural form (23R,25S) of the 25-hydroxy-lactone was found to be 10-fold more potent than the natural form, and the unnatural (23R,25S)1,25(OH)2D3-26,23-lactone three times more potent than the natural 1,25-dihydroxy-lactone in displacing 1,25(OH)2[3H]D3 from its intestinal receptor. While studying the biological activity of these lactone compounds, it was found that the natural form of the 25-hydroxy-lactone increased the intestinal calcium absorption 48 h after injection (16.25 nmol), while bone calcium mobilization was decreased by the same dose of the 25-hydroxy-lactone. The 1,25-dihydroxyvitamin D3-26,23-lactone in both its natural and unnatural forms was found to be active in stimulating ICA and BCM. These results suggest that the 25-hydroxy-lactone has some biological activity in the chick and that 1,25(OH)2D3-26,23-lactone can mediate ICA and BCM biological responses, probably through an interaction with 1,25-(OH)2D3 specific receptors in these target tissues.  相似文献
