Heating locally the hypocotyl of Bidens pilosa L. elicits awave of depolarization. The mechanism of the wave has been investigatedby means of microelectrophysiological techniques. The amplitudeof the transmembrane potential variation induced by an extracellularion concentration change (K+, Na+, Ca2+, Cl) was thesame in the resting conditions as during the slow wave. At pH4.0, the amplitude of the slow wave was reduced by 56% comparedwith the control performed at pH 7.0. In the presence of theuncoupler CCCP, the slow wave was not observed. The Ca2+ -chelatorEGTA and the Ca22+ -channel blocker La3+ reduced, respectively,the amplitude of the slow wave by 78% and 68%. These resultsindicate the involvement of Ca2+ in triggering the slow wave.A transient modification of the electrogenic H+ pump activity(inactivation-activation) and of the transmembrane H+ flux inthe slow wave are discussed. Three-day-old cowpea seedlings were inoculated with the severestrain of Cowpea Mosaic Virus (CpMV) and 24 h later with Bradyrhizobiumsp. cowpea, strain I-125, when virus translocation to rootsstill had not taken place. Plants were harvested at 22, 30,45 and 59 d after germination. Active virus replication wasassociated with increased protein content, detected in the leavesof 22-d-old plants. CpMV infection reduced the total leaf area,dry weight of shoots and the chlorophyll content of the firsttrifoliolate leaf at all experimental times. Low sugar contentwas recorded in leaves of 22- and 30-d-old CpMV-infected plantsand in nodules and roots of 30- and 45-d-old CpMV-infected plants.Up to 45 d, the nodule mass of CpMV-infected plants was lowerthan in controls, but reached control values at the 4th harvest.In CpMV-infected nodules, massive agglomeration of virus particles,crystalline virus inclusions and the proliferation of the endoplasmicreticulum were observed only in those cells containing bacteroids.In 30- and 45-d-old plants, CpMV infection decreased the contentof ureides in nodules, roots, and petioles. Virus infectiondid not alter the -amino-N content of roots and nodules butinduced a transient 74% reduction in the level of -amino-N inpetioles of 45-d-old plants. ) in nodulesalso showed the non-effect of CpMV on this enzyme, except fora temporary 16% reduction at the 2nd harvest. As compared tocontrols, the relative abundance of ureides in 30-d-old CpMV-infectedplants indicated a 15%, 10%, and 51% reduction in the nodules,roots, and petioles, respectively. Results indicate that atthe time of the 4th harvest the symbiotic process, measuredin terms of ureide content and enzymatic activities, was functioningat a near normal level despite nodule infection by CpMV. Abstract. Gravistimulation was investigated as a potential and unwanted component in the interpretation of physiological investigations on plants. Using both seedlings and mature sunflower plants, two situations are described where gravistimulation contributes significantly to the outcome of an experiment not initially designed to include this parameter. The number of adventitious roots formed in derooted seedings decreased when the tops of the plants were allowed to bend over under their own weight, and the effect correlated positively with the rate of ethylene production by non-vertical stems. In droughted mature plants, and increase in leaf and stem ethylene caused by water stress was supplemented by additional ethylene produced in the lower halves of stems. Drought had caused these stem tissues to wilt, become gravistimulated, and thus produce more ethylene. Other situations in which gravistimulated ethylene production and its physiological consequences are likely to be complicating factors in experiments are discussed. It is concluded that procedures that unnecessarily place experimental material in non-vertical orientations should be avoided. 相似文献

Animal bioerosion trace fossils upon mineral substrates are analyzed from the point of view of the Seilacherian ethological classification. Several of the currently accepted ethological classes: cubichnia, fugichnia, repichnia, fodinichnia, agrichnia, calichnia and aedificichnia are not represented in these substrates. This fact points out a lower behavioral diversity of hard substrate trace fossils when compared with soft sediment trace fossils. Bioerosion traces can be classified in just five classes: domichnia, pascichnia, equilibrichnia, praedichnia and fixichnia. Fixichnia is here erected to gather superficial etching scars resulting from the anchoring of fixation of sessile epiliths by means of a soft or skeletal body part. The location of major quantitative trait loci (QTL) contributing to stem and leaf [Na+] and [K+] was previously reported in chromosome 7 using two connected populations of recombinant inbred lines (RILs) of tomato. HKT1;1 and HKT1;2, two tomato Na+‐selective class I‐HKT transporters, were found to be closely linked, where the maximum logarithm of odds (LOD) score for these QTLs located. When a chromosome 7 linkage map based on 278 single‐nucleotide polymorphisms (SNPs) was used, the maximum LOD score position was only 35 kb from HKT1;1 and HKT1;2. Their expression patterns and phenotypic effects were further investigated in two near‐isogenic lines (NILs): 157‐14 (double homozygote for the cheesmaniae alleles) and 157‐17 (double homozygote for the lycopersicum alleles). The expression pattern for the HKT1;1 and HKT1;2 alleles was complex, possibly because of differences in their promoter sequences. High salinity had very little effect on root dry and fresh weight and consequently on the plant dry weight of NIL 157‐14 in comparison with 157‐17. A significant difference between NILs was also found for [K+] and the [Na+]/[K+] ratio in leaf and stem but not for [Na+] arising a disagreement with the corresponding RIL population. In cardiomyocytes, certain extracellular stimuli that activate heterotrimeric G protein-coupled receptors (GPCRs) can induce hypertrophy by regulating gene expression and increasing protein synthesis. We investigated if rat embryonic cardiomyocytes (H9c2) underwent variations in the expression levels and subcellular distribution of key components of GPCR-activated signaling pathways during endothelin-1 (ET-1)-induced hypertrophic response. A significant increase of p115RhoGEF protein level was evident in ET-1-treated cells. Real-time quantitative PCR showed RhoGEF mRNA levels were significantly increased. Inhibition of the Rho-associated kinase (ROCK) caused a significant decrease of p115RhoGEF protein in the nuclear fraction, whereas an inhibitor of PKC induced a redistribution of the protein between membrane/organelle and nuclear fractions. The ROCK inhibitor also decreased H9c2 cell hypertrophic response. These results indicate that ROCK and its downstream target molecules, which are involved in inducing the hypertrophic response, are also implicated in signaling the up-regulation of the p115RhoGEF protein. 相似文献
