Tumor cells use broad spectrum proteolytic activity of plasmin to invade tissue and form metastatic foci. Cell surface-associated enolase-1 (ENO-1) enhances plasmin formation and thus participates in the regulation of pericellular proteolysis. Although increased levels of cell surface bound ENO-1 have been described in different types of cancer, the molecular mechanism responsible for ENO-1 exteriorization remains elusive. In the present study, increased ENO-1 protein levels were found in ductal breast carcinoma and on the cell surface of highly metastatic breast cancer cell line MDA-MB-231. Elevated cell surface-associated ENO-1 expression correlated with augmented MDA-MB-231 cell migratory and invasive properties. Exposure of MDA-MB-231 cells to LPS potentiated translocation of ENO-1 to the cell surface and its release into the extracellular space in the form of exosomes. These effects were independent of de novo protein synthesis and did not require the classical endoplasmic reticulum/Golgi pathway. LPS-triggered ENO-1 exteriorization was suppressed by pretreatment of MDA-MB-231 cells with the Ca2+ chelator BAPTA or an inhibitor of endoplasmic reticulum Ca2+-ATPase pump, cyclopiazonic acid. In line with these observations, the stromal interaction molecule (STIM) 1 and the calcium release-activated calcium modulator (ORAI) 1-mediated store-operated Ca2+ entry were found to regulate LPS-induced ENO-1 exteriorization. Pharmacological blockage or knockdown of STIM1 or ORAI1 reduced ENO-1-dependent migration of MDA-MB-231 cells. Collectively, our results demonstrate the pivotal role of store-operated Ca2+ channel-mediated Ca2+ influx in the regulation of ENO-1 exteriorization and thus in the modulation of cancer cell migratory and invasive properties.  相似文献

Hageman factor (FXIIa) initiates the intrinsic coagulation pathway and triggers the kallikrein-kinin and the complement systems. In addition, it functions as a growth factor by expressing promitogenic activities toward several cell types. FXIIa binds to the cell surface via a number of structurally unrelated surface receptors; however, the underlying mechanisms are not yet fully understood. Here, we demonstrate that FXIIa utilizes cell membrane-bound glycosaminoglycans to interact with the cell surface of human lung fibroblasts (HLF). The combination of enzymatic, inhibitory, and overexpression approaches identified a heparan sulfate (HS) component of proteoglycans as an important determinant of the FXIIa binding capacity of HLF. Moreover, cell-free assays and competition experiments revealed preferential binding of FXIIa to HS and heparin over dextran sulfate, dermatan sulfate, and chondroitin sulfate A and C. Finally, we demonstrate that fibroblasts isolated from the lungs of the patients suffering from idiopathic pulmonary fibrosis (IPF) exhibit enhanced FXIIa binding capacity. Increased sulfation of HS resulting from elevated HS 6-O-sulfotransferase-1 expression in IPF HLF accounted, in part, for this phenomenon. Application of RNA interference technology and inhibitors of intracellular sulfation revealed the cooperative action of cell surface-associated HS and urokinase-type plasminogen activator receptor in the accumulation of FXIIa on the cell surface of IPF HLF. Moreover, FXIIa stimulated IPF HLF migration, which was abrogated by pretreatment of cells with heparinase I. Collectively, our study uncovers a novel role of HS-type glycosaminoglycans in a local accumulation of FXIIa on the cell membrane. The enhanced association of FXIIa with IPF HLF suggests its contribution to fibrogenesis.  相似文献

Until now a stable long-lived electronanopore could be generated in a lipid membrane only under current-clamp conditions, and stochastic properties of a single nanopore have been studied by the chronopotentiometry. The current-clamp experiment introduces negative feedback, which could be responsible for the electropore fluctuations and observed 1/fB power spectrum. A new electroporation method, chronoamperometry after current clamp (CACC), prevents irreversible rupture of the membrane and eliminates the feedback by clamping the voltage after previous electroporation. The experiments show that the electropore size can also be stabilized under constant potential. The electropore fluctuations do not need feedback to appear. The fluctuations are self-similar with a short memory. CACC provides an effective tool for studying the natural dynamics of an electropore in various environments, which was tested with Na+ and Al3+ ions. Comparison between chronopotentiometry and CACC reveals that the feedback mainly shortens the memory of the stochastic fluctuations. Statistical analysis shows that the conductance fluctuations can be approximately modeled as a fractional Lévy stable motion for a small hydrophilic electropore, which tends to fractional Brownian motion when the electropore increases its size. A hypothesis is presented that this transition reflects a more regular shape of big nanopores.  相似文献

Sphingomyelin signal transduction pathway regulates cell cycle through a number of lipid second messengers, which stimulate cell proliferation (sphingosine-1-phosphate), initiate growth arrest or induce apoptosis (sphingosine, ceramide). To asses the functioning of sphingomyelin pathway during liver regeneration after partial hepatectomy in rat (PH) we measured the content of sphingomyelin (SM), ceramide (CER), sphingosine (SPH), sphingosine-1-phosphate (S1P), the activity of neutral Mg(2+)-dependent and acidic sphingomyelinases and ceramidases, in the remnant liver lobes during the first 24h after PH in rat. The activity of acidic ceramidase was highest at 4th hour after PH, whereas the activity of neutral ceramidases peaked at 12th hour after the operation. At these time points the activity Mg(2+)-dependent sphingomyelinase was also elevated, together with the content of SPH, S1P and the ratio of S1P to CER. The activity of acidic sphingomyelinase increased gradually from 4th to 24th hour after the operation. This was accompanied by significant increase in the content of ceramide between 4th and 24th hour and reduction in the content of S1P and S1P to CER ratio. It is concluded that partial hepatectomy induces production of the pro-mitogenic intermediates of sphingomyelin signaling pathway during the first 12h of liver regeneration in rat.  相似文献

Both cytosolic PLA(2) (cPLA(2)) and secretory PLA(2) (sPLA(2)) have been implicated in pathology of cerebral ischemia. However, which of PLA(2) isoforms in astrocytes is responsible for arachidonic acid (AA) release contributing to their ischemic injury remains to be determined. The aim of the present study was to investigate the time-dependent activation of cPLA(2) and sPLA(2) in astrocytes exposed to combined oxygen glucose deprivation (OGD) as well as to evaluate the effectiveness of their pharmacological blockage as a method of preventing ischemic damage of the glial cells. It was shown that exposure of cultured astrocytes to OGD (0.5-24h) causes an increase in cPLA(2) and sPLA(2) expression and activity. The role of AA liberated mainly by cPLA(2) in the process of apoptosis was also demonstrated. To confirm the specific role of cPLA(2) and sPLA(2) in the mechanism of cells injury by OGD exposure, the effect of AACOCF(3) as cPLA(2) inhibitor and 12-epi-scalaradial as sPLA(2) inhibitor on AA release was examined. It was proved that simultaneous pharmacological blockade of enzymatic activity of cPLA(2) and sPLA(2) during OGD by AACOCF(3) and 12-epi-scalaradial substantially improves survival of ischemic injured glial cells.  相似文献

The differentiation and formation of the primitive endoderm in early embryos can be mimicked in vitro by the aggregation of embryonic stem cells to form embryoid bodies. We present morphological evidence that primitive endoderm cells often first locate in the interior of embryoid bodies and subsequently migrate to the surface. Cell mixing experiments indicate that surface positioning is an intrinsic property of endoderm epithelial cells. Moreover, Disabled-2 (Dab2) is required for surface sorting and positioning of the endoderm cells: when Dab2 expression was eliminated, the differentiated endoderm epithelial cells distributed throughout the interior of the embryoid bodies. Surprisingly, E-cadherin is dispensable for primitive endoderm differentiation and surface sorting in embryoid bodies. These results support the model that primitive endoderm cells first emerge in the interior of the inner cell mass and are subsequently sorted to the surface to form the primitive endoderm.  相似文献

BACKGROUND: Internucleosomal DNA fragmentation is one of the hallmarks of apoptosis. Because the low molecular weight DNA fragments are extracted during cell staining in aqueous solutions, apoptotic cells can be identified on DNA content frequency histograms as cells with fractional ("sub-G(1)") DNA content. The aim of the present study was to explore whether in situ DNA fragmentation during apoptosis is discontinuous or progresses incessantly and if it is discontinuous, to define the resistant to cleavage fraction of DNA that remains stainable with the fluorochrome. MATERIALS AND METHODS: The model of activation-induced apoptosis of human lymphocytes was chosen as it provides uniform cell population with identical DNA content (DI = 1.00) that undergo apoptosis. Their apoptosis was induced by multivalent mitogen phytohemagglutinin (PHA) in the absence and presence of geldanamycin (GA), the benzoquinone ansamycin antibiotic which binds to Hsp90 (Heat Shock Protein 90) and alters its function. The cells were stained with acridine orange, the metachromatic fluorochrome that differentially stains cellular DNA and RNA. RESULTS: A sharp, discrete peak representing the subpopulation of "sub-G(1)" cells with highly reproducible DI = 0.42 +/- 0.02 (CV = 5.5 +/- 1.2) was observed on DNA content histograms of lymphocytes whose apoptosis was induced by PHA alone. Two distinct peaks, one representing cell subpopulations with DI = 0.42 (as above) and another, with DI = 0.79 +/- 0.04 (CV = 5.8 +/- 0.4), respectively, were seen in apoptotic cells from cultures stimulated with PHA in the presence of GA. The frequency of cells represented by the sub-G(1) peaks varied depending on time of induction of apoptosis and GA concentration. CONCLUSIONS: Apoptosis-induced DNA fragmentation is discontinuous; approximately 42% of DNA is relatively stable and remains within the cell. The data suggest that the stable DNA is associated with nuclear matrix while the degradable fraction represents DNA in loop domains. A transient DNA stabilization is apparent in the presence of GA as evidenced by the presence of cell subpopulations with 79% of DNA retained in the cell. The observed discontinuity of DNA fragmentation appears to reflect sequential involvement of different nucleases and may also be modulated by chromatin structure.  相似文献
