Abstract The Open Bay Island skink (Oligosoma taumakae) is one of New Zealand's rarest lizard species. Until 2010, it was known only from two small islands in the Open Bay Island Group, a Māori-owned wildlife sanctuary in South Westland, New Zealand. Skinks on these islands are threatened by predation from weka (Gallirallus australis), a flightless native rail thought to have been introduced to the Open Bay Islands c. 100 years ago. Here, we describe the discovery of Open Bay Island skinks on two vegetated rock stacks located off the coast of Barn Bay, 52 km southwest of the Open Bay Islands. Although small (c. 0.10 and 0.36 ha), the Barn Islands appear to be predator-free, providing an important sanctuary for the skinks. We recommend: (1) a survey of mainland sites with suitable habitat; and (2) an evaluation of the need for island biosecurity measures for detecting and responding to incursions of small mammals. 相似文献

Egg activation is a series of highly coordinated processes that prepare the mature oocyte for embryogenesis. Typically associated with fertilization, egg activation results in many downstream outcomes, including the resumption of the meiotic cell cycle, translation of maternal mRNAs and cross-linking of the vitelline membrane. While some aspects of egg activation, such as initiation factors in mammals and environmental cues in sea animals, have been well-documented, the mechanics of egg activation in insects are less well-understood. For many insects, egg activation can be triggered independently of fertilization. In Drosophila melanogaster, egg activation occurs in the oviduct resulting in a single calcium wave propagating from the posterior pole of the oocyte. Here we use physical manipulations, genetics and live imaging to demonstrate the requirement of a volume increase for calcium entry at egg activation in ex vivo mature Drosophila oocytes. The addition of water, modified with sucrose to a specific osmolarity, is sufficient to trigger the calcium wave in the mature oocyte and the downstream events associated with egg activation. We show that the swelling process is regulated by the conserved osmoregulatory channels, aquaporins and DEGenerin/Epithelial Na+ channels. Furthermore, through pharmacological and genetic disruption, we reveal a concentration-dependent requirement of transient receptor potential M channels to transport calcium, most probably from the perivitelline space, across the plasma membrane into the mature oocyte. Our data establish osmotic pressure as a mechanism that initiates egg activation in Drosophila and are consistent with previous work from evolutionarily distant insects, including dragonflies and mosquitos, and show remarkable similarities to the mechanism of egg activation in some plants. 相似文献

Both the Entamoeba histolytica lectin, a virulence factor for the causative
agent of amebiasis, and the mammalian hepatic lectin bind to
N-acetylgalactosamine (GalNAc) and galactose (Gal) nonreducing termini on
oligosaccharides, with preference for GalNAc. Polyvalent GalNAc-
derivatized neoglycoproteins have >1000-fold enhanced binding affinity
for both lectins (Adler,P., Wood,S.J., Lee,Y.C., Lee,R.T., Petri,W.A.,Jr.
and Schnaar,R.L.,1995, J. Biol. Chem ., 270, 5164-5171). Substructural
specificity studies revealed that the 3-OH and 4-OH groups of GalNAc were
required for binding to both lectins, whereas only the E.histolytica lectin
required the 6-OH group. Whereas GalNAc binds with 4-fold lower affinity to
the E.histolytica lectin than to the mammalian hepatic lectin,
galactosamine and N-benzoyl galactosamine bind with higher affinity to the
E. histolytica lectin. Therefore, a synthetic scheme for converting
polyamine carriers to poly-N-acyl galactosamine derivatives (linked through
the galactosamine primary amino group) was developed to test whether such
ligands would bind the E.histolytica lectin with high specificity and high
affinity. Contrary to expectations, polyvalent derivatives including
GalN6lys5, GalN4desmosine, GalN4StarburstTMdendrimer, and
GalN8StarburstTMdendrimer demonstrated highly enhanced binding to the
mammalian hepatic lectin but little or no enhancement of binding to the
E.histolytica lectin. We propose that the mammalian hepatic lectin binds
with greatest affinity to GalNAc "miniclusters," which mimic branched
termini of N-linked oligosaccharides, whereas the E.histolytica lectin
binds most effectively to "maxiclusters," which may mimic more widely
spaced GalNAc residues on intestinal mucins.

The importance of managing land to optimize carbon sequestration for climate change mitigation is widely recognized, with grasslands being identified as having the potential to sequester additional carbon. However, most soil carbon inventories only consider surface soils, and most large‐scale surveys group ecosystems into broad habitats without considering management intensity. Consequently, little is known about the quantity of deep soil carbon and its sensitivity to management. From a nationwide survey of grassland soils to 1 m depth, we show that carbon in grassland soils is vulnerable to management and that these management effects can be detected to considerable depth down the soil profile, albeit at decreasing significance with depth. Carbon concentrations in soil decreased as management intensity increased, but greatest soil carbon stocks (accounting for bulk density differences), were at intermediate levels of management. Our study also highlights the considerable amounts of carbon in subsurface soil below 30 cm, which is missed by standard carbon inventories. We estimate grassland soil carbon in Great Britain to be 2097 Tg C to a depth of 1 m, with ~60% of this carbon being below 30 cm. Total stocks of soil carbon (t ha?1) to 1 m depth were 10.7% greater at intermediate relative to intensive management, which equates to 10.1 t ha?1 in surface soils (0–30 cm), and 13.7 t ha?1 in soils from 30 to 100 cm depth. Our findings highlight the existence of substantial carbon stocks at depth in grassland soils that are sensitive to management. This is of high relevance globally, given the extent of land cover and large stocks of carbon held in temperate managed grasslands. Our findings have implications for the future management of grasslands for carbon storage and climate mitigation, and for global carbon models which do not currently account for changes in soil carbon to depth with management. 相似文献

The prediction of pest-control functioning by multi-predator communities is hindered by the non-additive nature of species functioning. Such non-additivity, commonly termed an emergent multi-predator effect, is known to be affected by elements of the ecological context, such as the structure and composition of vegetation, in addition to the traits of the predators themselves. Here we report mesocosm experiments designed to test the influence of plant density and species composition (wheat monoculture or wheat and faba bean polyculture) on the emergence of multi-predator effects between Adalia bipunctata and Chrysoperla carnea, in their suppression of populations of the aphid Metopolophium dirhodum. The mesocosm experiments were followed by a series of behavioural observations designed to identify how interactions among predators are modified by plant species composition and whether these effects are consistent with the observed influence of plant species composition on aphid population suppression. Although plant density was shown to have no influence on the multi-predator effect on aphid population growth, plant composition had a marked effect. In wheat monoculture, Adalia and Chrysoperla mixed treatments caused greater suppression of M. dirhodum populations than expected. However this positive emergent effect was reversed to a negative multi-predator effect in wheat and faba bean polyculture. The behavioural observations revealed that although dominant individuals did not respond to the presence of faba bean plants, the behaviour of sub-dominants was affected markedly, consistent with their foraging for extra-floral nectar produced by the faba bean. This interaction between plant composition and predator community composition on the foraging behaviour of sub-dominants is thought to underlie the observed effect of plant composition on the multi-predator effect. Thus, the emergence of multi-predator effects is shown to be strongly influenced by plant species composition, mediated, in this case, by the provision of extra-floral nectar by one of the plant species. 相似文献

Although they are rare, fossilized gills are well known in Mesozoic coleoid cephalopods. In the Late Jurassic Solnhofen (South Germany) and Late Cretaceous Hâkel and Hâdjoula (Lebanon) plattenkalks, the feather-like gill remains are usually preserved as a yellowish staining. Small coleoids from Hâkel—tentatively determined as octopods—attracted our attention because these stains occur throughout the entire mantle sac in an unusual symmetrical pattern. Actualistic comparisons point to a compound of diverse vascular structures that most likely reflect central parts of the venous blood system (afferent branchial vessels, branchial hearts, vena pallialis, blood sinus) as well as the nephridial sacs. The nephridial sacs are clearly separated, which confirms the octopod nature of the fossils. A reticulated staining pattern in the rear of the mantle, which may reflect the gonad capillary system, suggests the presence of mature small-sized octopods. Based on its colour, its amorphous habit, and energy-dispersive X-ray spectroscopy (EDX) elemental analyses, the major components of the coelomic cavities have been replicated by an iron-rich phase (presumably goethite; copper was not detected). The goethite does not replace the tissues; rather, it traces their gross form as a well-defined “stain”. It is assumed the goethite is secondary after pyrite, which precipitated as a consequence of the oxygen-binding capacity of the copper-bearing haemocyanin and its ability to locally regulate redox potentials immediately postmortem. 相似文献

Sentinel prey (an artificially manipulated patch of prey) are widely used to assess the level of predation provided by natural enemies in agricultural systems. Whilst a number of different methodologies are currently in use, little is known about how arthropod predators respond to artificially manipulated sentinel prey in comparison with predation on free‐living prey populations. We assessed how attack rates on immobilized (aphids stuck to cards) and artificial (plasticine lepidopteran larvae mimics) sentinel prey differed to predation on free‐moving live prey (aphids). Predation was assessed in response to density of the common invertebrate predators, a foliar‐active ladybird Harmonia axyridis (Coleoptera: Coccinellidae), and a ground‐active beetle Pterostichus madidus (Coleoptera: Carabidae). Significant increases in attack rates were found for the immobilized and artificial prey between the low and high predator density treatments. However, an increased predator density did not significantly reduce numbers of free‐living live aphids included in the mesocosms in addition to the alternate prey. We also found no signs of predation on the artificial prey by the predator H. axyridis. These findings suggest that if our assessment of predation had been based solely on the foliar artificial prey, then no increase in predation would have been found in response to increased predator density. Our results demonstrate that predators differentially respond to sentinel prey items which could affect the level of predation recorded where target pest species are not being used. 相似文献

Cryobanking, the freezing of biological specimens to maintain their integrity for a variety of anticipated and unanticipated uses, offers unique opportunities to advance the basic knowledge of biological systems and their evolution. Notably, cryobanking provides a crucial opportunity to support conservation efforts for endangered species. Historically, cryobanking has been developed mostly in response to human economic and medical needs — these needs must now be extended to biodiversity conservation. Reproduction technologies utilizing cryobanked gametes, embryos and somatic cells are already vital components of endangered species recovery efforts. Advances in modern biological research (e.g. stem cell research, genomics and proteomics) are already drawing heavily on cryobanked specimens, and future needs are anticipated to be immense. The challenges of developing and applying cryobanking for a broader diversity of species were addressed at an international conference held at Trier University (Germany) in June 2008. However, the magnitude of the potential benefits of cryobanking stood in stark contrast to the lack of substantial resources available for this area of strategic interest for biological science — and society at large. The meeting at Trier established a foundation for a strong global incentive to cryobank threatened species. The establishment of an Amphibian Ark cryobanking programme offers the first opportunity for global cooperation to achieve the cryobanking of the threatened species from an entire vertebrate class. 相似文献
