Macrobenthic community structure and the distribution of the green sea urchin (Strongylocentrotus droebachiensis) were recorded inside and outside (=barrens) of kelp patches (Alaria esculenta) at Kongsfjordneset, Svalbard between August 2002 and October 2006. In manipulative field experiments, conducted at Kongsfjordneset, Svalbard in August 2002, the effect of the presence of the brown seaweed Desmarestia viridis on sea urchin distribution and kelp grazing was determined. Additionally, we studied the effect of sulphuric acid, which is produced and stored by D. viridis, on sea urchin movements in the laboratory at Ny-Ålesund, Svalbard, in May 2006. Sea urchin densities were two- to threefold lower in kelp patches than on barrens. The macrobenthic community inside kelp patches hosted 39% more species and was of different species composition than on barrens. Anchored pieces of the kelp A. esculenta were less consumed when surrounded by D. viridis than non-surrounded conspecifics. Changes in pH affected the behaviour of sea urchins. Exposing sea urchins to 500 μl seawater at pH 7.5 caused them to stop, while the exposure of as little as 25 μl at pH 1 caused sea urchins to move in the opposite direction. Acid-mediated escape responses in sea urchin behaviour suggest chemical protection by D. viridis as an additional mechanism to mechanical protection in the generation of kelp refuges. These results improve our understanding of how isolated kelp beds can persist over a wide range of environmental conditions, like wave-sheltered sites, and suggest that changes in community structure may be in part attributable to altered trophic interactions. 相似文献

The generalized additive model is extended to handle negative binomial responses. The extension is complicated by the fact that the negative binomial distribution has two parameters and is not in the exponential family. The methodology is applied to data involving DNA adduct counts and smoking variables among ex-smokers with lung cancer. The life history, reproduction and development of Desmarestia menziesii J. Agardh from Antarctica is described. Unilocular sporangia occur singly or in small groups in the outermost cortical layer of the sporophyte. They are formed by periclinal division of cortex cells into a stalk cell and the sporangium initial. Meiospores germinate into dioecious microscopic filamentous gametophytes. As in other perennial Antarctic species of the Desmarestiales, gametangia are formed in culture under short-day conditions or in darkness. In nature, juvenile sporophytes should therefore be formed in winter. They develop only attached to the oogonium. At first they are uniseriate and elongate by means of an intercalary meristem located in their middle part. Laterals are formed predominantly in this region, and they subsequently give rise to secondary laterals. The branching pattern is opposite to alternate in both young and adult plants. Cortication of the main axis is initiated by filaments growing out from the lowermost cells of the primary laterals. In sporophytes of this developmental stage the meristem of the main axis is confined to a small region where cortication starts and above. Lateral branches elongate and become corticated in the same way as the main axis. In mature plants, cells of the inner cortex can become meristematic again and form a meristoderm which contributes to axis thickness by periclinal and anticlinal divisions. The interactive effects of an 8 h exposure to UV radiation and altered temperatures on the ultrastructure and germination of zoospores of the sublittoral brown alga Laminaria hyperborea (Gunn.) Foslie were investigated for the first time. Spores were exposed to four temperatures (2, 7, 12 and 17 degrees C) and three light regimes (PAR, PAR + UV-A, PAR + UV-A+UV-B). Freshly-released spores of L. hyperborea lack a cell wall and contain a nucleus with fine granular nucleoplasm and a nucleolus, one chloroplast, several mitochondria, dictyosomes and an endoplasmatic reticulum. Further, several kinds of so-called adhesive vesicles, lipid globuli and physodes containing UV-absorbing phlorotannins are embedded in the cytoplasm. No eye-spot is present. Physodes were found but they were rare and small. After an 8 h exposure to UV-B, the nucleoplasm had a mottled structure, chloroplasts contained plastoglobuli, the structure of the mitochondria changed from crista- to sacculus-type and germination was strongly inhibited at all temperatures. UV-A only had an impact on the ultrastructure at the highest temperature tested. The strongest effects were found at 17 degrees C, where germination was reduced to 35%, 32% and 9% after exposure to PAR, PAR+UV-A and PAR + UV-A + UV-B, respectively. This study indicates that UV-B radiation has strong damaging effects on the physiology and ultrastructure of zoospores of L. hyperborea. The results are important for developing scenarios for the effect of enhanced UV radiation and increasing temperatures caused by global climate changes. 相似文献

BACKGROUND: Benzo(a)pyrene (BaP), anthracene (ANTH) and chrysene (CHRY) are polynuclear aromatic hydrocarbons (PAHs) implicated in renal toxicity and carcinogenesis. These PAHs elicit cell type-specific effects that help predict toxicity outcomes in vitro and in vivo. While BaP and ANTH selectively injure glomerular mesangial cells, and CHRY targets cortico-tubular epithelial cells, binary or ternary mixtures of these hydrocarbons markedly reduce the overall cytotoxic potential of individual hydrocarbons. METHODS: To study the biochemical basis of these antagonistic interactions, renal glomerular mesangial cells were challenged with BaP alone (0.03 - 30 microM) or in the presence of ANTH (3 microM) or CHRY (3 microM) for 24 hr. Total RNA and protein will be harvested for Northern analysis and measurements of aryl hydrocarbon hydroxylase (AHH) and ethoxyresorufin-O-deethylase (EROD) activity, respectively, to evaluate cytochrome P450 mRNA and protein inducibility. Cellular hydrocarbon uptake and metabolic profiles of PAHs were analyzed by high performance liquid chromatography (HPLC). RESULTS: Combined hydrocarbon treatments did not influence the cellular uptake of individual hydrocarbons. ANTH or CHRY strongly repressed BaP-inducible cytochrome P450 mRNA and protein expression, and markedly inhibited oxidative BaP metabolism. CONCLUSION: These findings indicate that antagonistic interactions among nephrocarcinogenic PAHs involve altered expression of cytochrome P450s that modulate bioactivation profiles and nephrotoxic/ nephrocarcinogenic potential. 相似文献

Epiphytic red algae of the order Ceramiales from mangroves and salt marshes (nine species from Bostrychia, three from Stictosiphonia and four from Caloglossa) produce varying levels of the UV-absorbing compounds mycosporine-glycine, shinorine, porphyra-334, palythine, asterina-330 and palythinol, a suite of substances chemically assigned as mycosporine-like amino acids (MAAs). Mean MAA levels varied from 0.02 to 12.8 mg g–1 DW in field-collected and laboratory cultured specimens. While in field samples of Bostrychia montagneiHarvey, Bostrychia radicans (Montagne) Montagne and Caloglossa apomeiotica J.West et G.Zuccarello MAA concentrations were generally higher compared to cultured plants of the same taxa, Bostrychia tenella(Lamouroux) J.Agardh did not show such a difference. Catenella caespitosa (Withering) L.Irvine, Catenella impudica (Montagne) J.Agardh and Catenella nipae Zanardini (Gigartinales, Caulacanthaceae) produce two novel UV-absorbing compounds: MAA-1 (1.4–4.3 mg g –1 DW) and MAA-2 (0.1–1.0 mg g–1 DW), which absorb at 334 nm and 320 nm, respectively. In laboratory culture of Bostrychia moritziana when photosynthetically active radiation (PAR) was increased from 20 to 40 mol photons m–2 s–1, the total level of palythinol increased by 85% (from 2.0 to 3.7 mg g–1 DW). In a culture of Caloglossa leprieurii when PAR was increased from 40 to 80 mol m–2 s–1the porphyra-334 content increased by 77% (from 3.1 to 5.5 mg g–1 DW). Extremely high MAA contents of >30 mg g–1 DW were detected in mature tetrasporangial sori prepared from two isolates of laboratory-cultured reproductive Caloglossa apomeiotica compared to vegetative plants (about 10 mg MAAs g–1 DW) indicating tetraspores loaded up with UV-sunscreens. All data demonstrate that mangrove red algae contain high MAA concentrations, particularly the reproductive structures, and hence these compounds may act as biochemical photoprotectants against exposure to UV-radiation. 相似文献

The Effect of Elevated UV Radiation on Fucus spp. (Fucales,Phaeophyta) Zygote and Embryo Development

Abstract: This study has shown that in Fucus serratus and Fucus distichus, young zygotes and embryos are highly susceptible to elevated levels of both UVA (UVAR) and UVB radiation (UVBR). Zygotes treated with UVAR are able to polarise and germinate, but are very slow to divide; if they do, they often have skewed division planes or deformed rhizoids. Those treated with UVAR and UVBR remain spherical, they do not polarise, germinate to form rhizoids or undergo cell division. We suggest that the UVR may be affecting the cytoskeleton. Conversely, zygotes and embryos of Fucus spiralis are able to withstand these same UVR levels and, at the light microscope level, appear to develop normally. When the brown algal phenolic compound phloroglucinol was placed in a filter covering the developing embryos, normal development was seen under all treatments. Phenolic compounds protect the developing fucoids from UVR. In comparison with the other two species, Fucus spiralis grows high up on the shore and is exposed for much longer periods of time and, presumably, to higher levels of natural UVR. The failure of the juvenile stages of F. serratus and F. distichus to withstand UVR stress may have implications for the continued survival of these species in the intertidal, and may prove detrimental to the population as a whole if UVR levels increase. 相似文献

Background
Irreversible airflow obstruction in Chronic Obstructive Pulmonary Disease (COPD) is thought to result from airway remodelling associated with aberrant inflammation. Patients who experience frequent episodes of acute deterioration in symptoms and lung function, termed exacerbations, experience a faster decline in their lung function, and thus over time greater disease severity However the mechanisms by which these episodes may contribute to decreased lung function are poorly understood.This study has prospectively examined changes in sputum levels of inflammatory cells, MMP-9 and TIMP-1 during exacerbations comparing with paired samples taken prior to exacerbation.Methods
Nineteen COPD patients ((median, [IQR]) age 69 [63 to 74], forced expiratory volume in one second (FEV1) 1.0 [0.9 to1.2], FEV1% predicted 37.6 [27.3 to 46.2]) provided sputa at exacerbation. Of these, 12 were paired with a samples collected when the patient was stable, a median 4 months [2 to 8 months] beforehand.Results
MMP-9 levels increased from 10.5 μg/g [1.2 to 21.1] prior to exacerbation to 17.1 μg/g [9.3 to 48.7] during exacerbation (P < 0.01). TIMP-1 levels decreased from 3.5 μg/g [0.6 to 7.8] to 1.5 μg/g [0.3 to 4.9] (P = 0.16). MMP-9/TIMP-1 Molar ratio significantly increased from 0.6 [0.2 to 1.1] to 3.6 [2.0 to 25.3] (P < 0.05). Neutrophil, eosinophil and lymphocyte counts all showed significant increase during exacerbation compared to before (P < 0.05). Macrophage numbers remained level. MMP-9 levels during exacerbation showed highly significant correlation with both neutrophil and lymphocyte counts (Rho = 0.7, P < 0.01).Conclusion
During exacerbation, increased inflammatory burden coincides with an imbalance of the proteinase MMP-9 and its cognate inhibitor TIMP-1. This may suggest a pathway connecting frequent exacerbations with lung function decline. 相似文献69.

Stratospheric ozone depletion and the concomitant increase in irradiance of ultraviolet-B radiation (UVB) at the earth’s surface represent major threats to terrestrial and aquatic ecosystems. In costal rocky shore environments, seaweeds constitute a group of organisms of particular significance to ecosystem function. Thus, impairment of seaweed performance by UVB-exposure may result in severe changes in the functioning of coastal ecosystems. Here we present our view on how UVB radiation affects seaweed physiology and ecology and, thus, shapes the coastal environment by affecting the spatial, species and functional structure of seaweed communities. 相似文献
