By immunocytochemistry and in situ hybridization at the electron microscopy level, and by the PCR technique, we have shown that HIV-1 binds and enters normal sperm; that viral particles, their antigens, and nucleic acid are present in sperm from HIV-1 infected men; and that such sperm can transfer HIV-1 like particles to normal human oocytes. We also present evidence that a galactosylceramide-like compound is present on the sperm membrane and could function as an alternative receptor for HIV. 相似文献

Bacterial ice nucleation activity (INA+ phenotype) can be traced to the
product of a single gene, ina. A remarkably sparse distribution of this
phenotype within three bacterial genera indicates that the ina gene may
have followed an unusual evolutionary path. Southern blot analyses, coupled
with assays for ice-nucleating ability, revealed that within four bacterial
species an ina gene is present in some strains but absent from others.
Results of hybridization experiments using DNA fragments that flank the ina
gene suggested that the genotypic dimorphism of ina may be anomalous. A
phylogenetic analysis of 16S ribosomal RNA gene sequences from a total of
14 ina+ and ina- bacterial strains indicated that the ina+ bacteria are not
monophyletic but instead phylogenetically interspersed among ina- bacteria.
The relationships of ina+ bacteria inferred from ina sequence did not
coincide with those inferred from the 16S data. These results suggest the
possibility of horizontal transfer in the evolution of bacterial ina genes.

The method of secretory granuleformation in the acinar cells of the rat exorbital lacrimal gland was studied by electron microscope morphological and cytochemical techniques. Immature secretory granules at the inner face of the Golgi apparatus were frequently attached to a narrow cisternal structure similar to GERL as described in neurons by Novikoff et al. (Novikoff, P. M., A. B. Novikoff, N. Quintana, and J.-J. Hauw. 1971. J. Cell Bio. 50:859-886). In the lacrimal gland. GERL was located adjacent to the inner Golgi saccule, or separated from it by a variable distance. Portions of GERL were often closely paralleled by modified cisternae of rough endoplasmic reticulum (RER), which lacked ribosomes on the surface adjacent to GERL. Diaminobenzidine reaction product of the secretory enzyme peroxidase was localized in the cisternae of the nuclear envelope, RER, peripheral Golgi vesicles, Golgi saccules, and immature and mature secretory granules. GERL was usually free of peroxidase reaction product or contained only a small amount. Thiamine pyrophosphatase reaction product was present in two to four inner Golgi saccules; occasionally, the innermost saccule was dilated and fenestrated, and contained less reaction product than the next adjacent saccule. Acid phosphatase (AcPase) reaction product was present in GERL, immature granules, and, rarely, in the innermost saccule, but not in the rest of the Golgi saccules. Thick sections of AcPase preparations viewed at 100 kV revealed that GERL consisted of cisternal, and fenestrated or tublular portions. The immature granules were attached to GERL by multiple connections to the tublular portions. These results suggest that, in the rat exorbital lacrimal gland, the Golgi saccules participate in the transport of secretory proteins, and that GERL is involved in the formation of secretory granules. 相似文献

Remedial sport hunting of predators is often used to reduce predator populations and associated complaints and livestock depredations. We assessed the effects of remedial sport hunting on reducing cougar complaints and livestock depredations in Washington from 2005 to 2010 (6 years). The number of complaints, livestock depredations, cougars harvested, estimated cougar populations, human population and livestock populations were calculated for all 39 counties and 136 GMUs (game management units) in Washington. The data was then analyzed using a negative binomial generalized linear model to test for the expected negative relationship between the number of complaints and depredations in the current year with the number of cougars harvested the previous year. As expected, we found that complaints and depredations were positively associated with human population, livestock population, and cougar population. However, contrary to expectations we found that complaints and depredations were most strongly associated with cougars harvested the previous year. The odds of increased complaints and livestock depredations increased dramatically (36 to 240%) with increased cougar harvest. We suggest that increased young male immigration, social disruption of cougar populations, and associated changes in space use by cougars - caused by increased hunting resulted in the increased complaints and livestock depredations. Widespread indiscriminate hunting does not appear to be an effective preventative and remedial method for reducing predator complaints and livestock depredations. 相似文献

For most neurodegenerative diseases the precise duration of an individual cell''s death is unknown, which is an obstacle when counteractive measures are being considered. To address this, we used the rd1 mouse model for retinal neurodegeneration, characterized by phosphodiesterase-6 (PDE6) dysfunction and photoreceptor death triggered by high cyclic guanosine-mono-phosphate (cGMP) levels. Using cellular data on cGMP accumulation, cell death, and survival, we created mathematical models to simulate the temporal development of the degeneration. We validated model predictions using organotypic retinal explant cultures derived from wild-type animals and exposed to the selective PDE6 inhibitor zaprinast. Together, photoreceptor data and modeling for the first time delineated three major cell death phases in a complex neuronal tissue: (1) initiation, taking up to 36 h, (2) execution, lasting another 40 h, and finally (3) clearance, lasting about 7 h. Surprisingly, photoreceptor neurodegeneration was noticeably slower than necrosis or apoptosis, suggesting a different mechanism of death for these neurons. 相似文献

There has been limited attention to estimating maternity rate because it appears to be relatively simple. However, when used for multi-annual breeder species, such as the largest carnivores, the most common estimators introduce an upward bias by excluding unproductive females. Using a simulated dataset based on published data, we compare the accuracy of maternity estimates derived from standard methods against estimates derived from an alternative method. We show that standard methods overestimate maternity rates in the presence of unsuccessful pregnancies. Importantly, population growth rates derived from a matrix model parameterized with the biased estimates may indicate increasing populations although the populations are stable or even declining. We recommend the abandonment of the biased standard methods and to instead use the unbiased alternative method for population projections and assessments of population viability. 相似文献

In social species, the transmission and maintenance of infectious diseases depends on the contact patterns between individuals within groups and on the interactions between groups. In southern Africa, the Cape buffalo (Syncerus caffer caffer) is a vector for many pathogens that can infect sympatric livestock. Although intra-group contact patterns of Cape buffalo have been relatively well described, how groups interact with each other and risks for pathogen transmission remain poorly understood. We identified and compared spatial behavior and contact patterns between neighboring groups of Cape buffalo under contrasting environments: within the seasonally flooded environment of the Okavango Delta in Botswana and the semi-arid environment of northern Kruger National Park in South Africa. We used telemetry data collected between 2007 and 2015 from 10 distinct groups. We estimated seasonal overlap and proximity between home ranges of pairwise neighboring groups, and we quantified seasonal contact patterns between these groups. We defined contact patterns within variable spatiotemporal windows compatible with the transmission of diseases carried by the Cape buffalo: bovine tuberculosis, brucellosis, and Rift Valley fever (mosquito-borne transmission). We examined the effects of habitat and distance to water on contact location. In both study populations, neighboring buffalo groups were highly spatially segregated in the dry and rainy seasons. Inter-group contact patterns were characterized by very few direct and short-term indirect (within 0–2 days) contacts, lasting on average 1 hour and 2 hours, respectively. Contact patterns were generally consistent across populations and seasons, suggesting species-specific behavior. In the drier study site, the probability of indirect and vector-borne contacts generally decreased during the dry season with increasing distance to water. In the seasonally flooded area, only the probability of vector-borne contact decreased with increasing distance to water. Our results highlight the importance of dry season water availability in influencing the dynamics of indirectly transmitted Cape buffalo pathogens but only in areas with low water availability. The results from this study have important implications for future modeling of pathogen dynamics in a single host, and the ecology and management of Cape buffalo at the landscape level. © 2021 The Authors.
