Huntington disease (HD) is a genetically dominant condition caused by expanded CAG repeats coding for glutamine in the HD gene product huntingtin. Although HD symptoms reflect preferential neuronal death in specific brain regions, huntingtin is expressed in almost all tissues, so abnormalities outside the brain might be expected. Although involvement of nuclei and mitochondria in HD pathophysiology has been suggested, specific intracellular defects that might elicit cell death have been unclear. Mitochondria dysfunction is reported in HD brains; mitochondria are organelles that regulates apoptotic cell death. We now report that lymphoblasts derived from HD patients showed increased stress-induced apoptotic cell death associated with caspase-3 activation. When subjected to stress, HD lymphoblasts also manifested a considerable increase in mitochondrial depolarization correlated with increased glutamine repeats.  相似文献

The COVID-19 pandemic has presented significant challenges and implications for the sports community. Thus, this study aimed to describe the prevalence of COVID-19 in Brazilian athletes and identify the epidemiological, clinical, athletic, life and health factors associated with the disease in these individuals. A cross-sectional study was performed involving 414 athletes from 22 different sports using an online questionnaire from August to November 2020. The association between the athletes’ characteristics and COVID-19 was evaluated using a logistic regression model. The prevalence of COVID-19 was 8.5%, although only 40% of athletes reported having been tested. Being under 27 years of age (3-fold), having children (~5-fold), having a teammate test positive for COVID-19 (2.5-fold), and smoking (14-fold) were associated with a possible higher risk of disease. Almost 20% of athletes self-reported musculoskeletal injuries during the period of the pandemic that was studied. Athletes with a university education (P = 0.02), a profession other than sports (P < 0.001), those from a low-income family (P = 0.01), and public health system users (P = 0.04) were significantly less frequently tested for COVID-19, whereas international competitors, athletes who received a wage, and athletes who had a teammate who tested positive for COVID-19 were 2-, 3-, and 15-fold more likely to be tested for COVID-19, respectively. Approximately 26% of the athletes who tested negative or were untested reported more than three characteristic COVID-19 symptoms, and 11% of athletes who tested positive for COVID-19 were asymptomatic. The identification of modifiable (have children, smoking, and teammates positively tested) and non-modifiable (age under 27 years) factors related to COVID-19 in athletes can contribute to implementing surveillance programmes to decrease the incidence of COVID-19 in athletes and its negative impacts in sports.  相似文献

DNP (dynamic nuclear polarization)-enhanced solid-state NMR is employed to directly detect protein–DNA and protein–ATP interactions and identify the residue type establishing the intermolecular contacts. While conventional solid-state NMR can detect protein–DNA interactions in large oligomeric protein assemblies in favorable cases, it typically suffers from low signal-to-noise ratios. We show here, for the oligomeric DnaB helicase from Helicobacter pylori complexed with ADP and single-stranded DNA, that this limitation can be overcome by using DNP-enhanced spectroscopy. Interactions are established by DNP-enhanced 31P–13C polarization-transfer experiments followed by the recording of a 2D 13C–13C correlation experiment. The NMR spectra were obtained in less than 2 days and allowed the identification of residues of the motor protein involved in nucleotide binding.  相似文献

Coexistence of trees and grasses in savannas should be possible if competition between the woody and the grassy components is less intense than the competition within each component. Although several studies have investigated competition between trees and grasses, little is known about tree–tree interactions. We used a multi-proxy approach to examine the spatial pattern of Acacia mellifera and other savanna woody species in a semi-arid savanna in South Africa. Spatial analysis of the point patterns of young and reproductively mature shrubs detected decreasing aggregation with size/age over all spatial scales. This indicated the prevalence of competition although the overall spatial shrub pattern was aggregated. In contrast to point pattern statistics that detect changes only when competition has led to the death of the inferior competitor, we also applied methods identifying the competitive effect on sizes of individual trees. Competition should lead to a negative spatial autocorrelation in size, which we observed in half of the studied cases. Quantile regressions show that nearest-neighbour distance increased steeply with combined size of the target shrub and its neighbours indicating strong competitive effects. The medians of the distributions of maximum root lengths of A. mellifera, of the scale of regular patterns, and of negative autocorrelations were not significantly different, suggesting that overlapping root systems mediate competitive interactions. A competitor removal experiment did not lead to increased shrub sizes, which may be due to the limited duration of the experiment. From the nearest neighbour and autocorrelation analyses, we conclude that competition had a strong impact on growth rates of savanna woody species. Competition-induced mortality only becomes obvious when analysing the shift towards less aggregated spatial patterns when shrubs become reproductively mature. As the overall clustered spatial pattern masks the perceptible effect of competition, a time component should always be included in spatial pattern-based inference of competition.  相似文献

Mediterranean ecosystems are hotspots of species richness where fire is one of the key processes influencing their structure, composition and function. Post‐fire seedling emergence constitutes a crucial event in the life cycle of plants and species‐specific temporal and spatial patterns of seedling emergence have been hypothesized to contribute to the high diversity in these ecosystems. Here we study the temporal and spatial patterns of seedling emergence observed for the four dominant species (Cistus albidus, Ulex parviflorus, Helianthemum marifolium, Ononis fruticosa) after an experimental fire in a Mediterranean gorse shrubland. In a first analysis we compared the timing of emergence of each species using the Kaplan‐Meier method. The spatial component of seedling emergence and the spatiotemporal relationship between different cohorts of the same species were analyzed using recent techniques of spatial point pattern analyses. We found a bimodal temporal pattern of emergence. Emergence of Cistaceae species (H. marifolium and C. albidus) occurred predominantly early after the fire while Fabaceae (O. fruticosa and U. parviflorus) emerged mainly during the following autumn. Individually, all species showed an aggregated spatial pattern and, when testing for pair interactions, we found that the clusters of individual species were spatially segregated. Additionally, the clusters of individual species showed an internal spatial structure where seedlings of different cohorts were spatially segregated. Theoretical models predict that these patterns will promote species coexistence. We identified a number of mechanisms that all have the potential to contribute to the observed pattern formation. However, the potential interaction among these mechanisms are complex and not easy to predict. Our analyses take a significant step forward in studying seedling emergence in fire prone ecosystems since, to our knowledge, this is the first time that both spatial and temporal patterns of all dominant species have been studied together.  相似文献

We examined spatial patterns of fire-caused mortality and after-fire establishment of two dominant shrub species, Baccharis dracunculifolia and Eupatorium buniifolium in a humid temperate South American savannah. Our objective was to determine whether fires mediate in interactions between these two species. After a natural fire burned a large tract of savannah, we established two plots (respectively 550 and 500 m2) within which we mapped all surviving and dead shrubs as well as all individuals of shrub species that recruited in the following year. We used techniques of point-pattern analysis to test specific null hypotheses about spatial associations in the distribution, mortality, and establishment of shrubs. Results support the notions that fire mediates interactions between these two species. Fire-caused death of E. buniifolium tended to occur selectively in the vicinities of Baccharis individuals, and recruitment of B. dracunculifolia tended to be concentrated in the places of dead shrubs. These responses, however, were contingent on local abundances of shrubs which depend in part from the recent fire history. Anthropogenic perturbation of the natural fire regime would have therefore distorted the role of fire mediated interactions as drivers of the dynamics of the vegetation of this temperate savannah.  相似文献

Liver and gills of roach (Rutilus rutilus) and silver carp (Hypophthalmichthys molitrix) were examined for glutathione S-transferases (GSTs) contents and their substrate specificity and capacity to biotransform microcystin-LR (MC-LR). GSTs and other glutathione (GSH) affine proteins were purified using a GSH-agarose matrix and separated by anionic chromatography (AEC). Substrate specificities were determined photometrical for 1-chloro-2,4-dinitrobenzene (CDNB), 1,2-dichloro-4-nitrobenzene (DCNB), 4-nitrobenzyl chloride (pNBC) and ethacrynic acid (ETHA). Biotransformation rate of MC-LR was determined by high performance liquid chromatography (HPLC). Roach exhibited different hepatic and branchial GST activities for used substrates (DNB, pNBC and DCNB) compared to silver carp but not for ethacrynic acid. It suggests that, both fish species have similar amount of pi and/or alpha class, which were the dominant GST classes in liver and gills. Gills of both fish species contained a higher number of GST isoenzymes, but with lower activities and ability of MC-LR biotransformation than livers. GST isoenzymes from roach had higher activity to biotransform MC-LR (conversion rate ranging up to 268 ng MC-LR min? 1 mL? 1 hepatic enzyme) than that isolated from silver carp. Without any prior contact to MC-LR or another GST inducer, roach seems to be better equipped for microcystin biotransformation than silver carp.  相似文献

A 41-year study (1968–2008) of the orchids of the Catoctin Mountains, Frederick County, Maryland reveals that 19 of 21 species have experienced precipitous declines. Four of these species are currently considered Threatened or Endangered by the State of Maryland and another two are considered Rare. Annual census data at 167 sites from throughout the Catoctin Mountains on protected and unprotected lands (private and public) show a loss of three species from the study area, a decline of >90 % (ranging from 99 to 91 %) in seven species, and a decline of <90 % (ranging from 51 to 87 %) for nine species. Each species was analyzed using Ordinary Least Squares Analysis to show trends and document corresponding R2 and p values. We tested the hypothesis that this decline is due to intensified herbivory by white-tailed deer. The overall orchid census data is significantly inversely-correlated (R = ?0.93) to the white-tailed deer harvest data of Frederick County (a surrogate for population size), which includes the entirety of the study area. Platanthera ciliaris showed a huge expansion at a single site explicitly managed for this species otherwise this orchid showed a decline similar to the other species. Proper management is critical for the continuation of the orchid species in this study, be it control of the white-tailed deer herd or combating woody plant succession in the case of P. ciliaris.  相似文献
