ABSTRACT: BACKGROUND: Asymptomatic carriage of Plasmodium falciparum and Plasmodium vivax is common in both low-and high-transmission settings and represents an important reservoir of infection that needs to be targeted if malaria elimination is to succeed. METHODS: Mass blood examinations (475 individuals) were conducted in two villages in Mae Hong Son, an area of endemic but low-transmission malaria in the north-west of Thailand. The microscopist at the local malaria clinic did not detect any infections. Pools of four samples were screened by real-time PCR; individual members of all of the positive pools were then re-examined by expert microscopy and by a second species-specific PCR reaction. RESULTS: Eight subjects were found to be positive by both PCR and expert microscopy and one was found to be positive by PCR alone. The slides contained asexual stage parasites of P. vivax, P. falciparum and Plasmodium malariae, but no gametocytes. The local clinic was notified within two to eight days of the survey. CONCLUSION: A combination of pooling, real-time PCR and expert microscopy provides a feasible approach to identifying and treating asymptomatic malaria infections in a timely manner.  相似文献

Objective: To determine the association of four simple anthropometric indices with coronary heart disease (CHD) in Thai men, and to determine the optimal cut‐off points for each index in the prediction of CHD. Research Methods and Procedures: This is a cohort study with 17 years of follow‐up. A total of 2536 male employees from the Electricity Generating Authority of Thailand 35 to 59 years of age at baseline were included in the study. Height, weight, waist circumference, and hip circumference were measured to generate BMI, waist circumference (WC), waist‐to‐hip ratio (WHR), and waist‐to‐height ratio (WHtR). Cox regression models were used to estimate hazard ratios by thirds of each index. Receiver operating characteristic curves were used to assess discrimination of CHD. Results: WHtR was most strongly associated with CHD events in Thai men. The age‐adjusted hazard ratio for those in the highest, compared with the lowest, third was 2.89 (1.37, 6.11). Although WHtR had the largest area under the receiver operating characteristic curve (AUC) with the optimal cut‐off estimated to be 0.51 (sensitivity, 55%; specificity, 61%), no statistically significant difference (p > 0.10) was found between the AUC for WHtR and that for the other three indices. Conclusion: WHtR is, marginally, the best of the four indices considered to predict CHD events in Thai men.  相似文献

Pueraria mirifica is a tuberous plant enriched with active phytoestrogens. There is no established information about the factors influencing isoflavonoid storage in the tubers. We investigated the tuberous storage of the major isoflavonoids of 1-year-old plants. Four cultivars of P. mirifica were cultivated in the same field trial during the same period to establish a unique plant age and differentiation under the same environment and soil conditions. The tubers collected from the 1-year-old plants in the summer, rainy season and winter were submitted to an HPLC analysis with a gradient system comprising 0.1% acetic acid and acetonitrile. Five major isoflavonoids, puerarin, daidzin, genistin, daidzein and genistein, were adopted as standards. P. mirifica tubers of different cultivars collected in the same season exhibited significant differences in individual and total isoflavonoid contents, showing chemovariety. P. mirifica tubers of the same cultivar collected from different seasons also exhibited significant differences in individual and total isoflavonoid contents, showing the influence of season. In conclusion, the tuberous storage of major isoflavonoids in 1-year-cultivated plants was greatly diverse and was strongly influenced by the season and plant genetics.  相似文献

用改进的斑点免疫结合法,检测了烟草花叶病毒、黄瓜花叶病毒、豇豆花叶病毒、芋花叶病毒、柑桔速衰病毒和柑桔顽固病螺原体。无论用单克隆抗体还是多克隆抗体,全血清还是提纯的IgG,均获得满意的结果。直接法与间接法结果相同。在测定豇豆花叶病毒和芋花叶病毒时,与酶联免疫吸附试验和免疫电镜进行比较。表明无论是对纯化病毒还是感病植物汁液,其测定的敏感性均优于后二法,豇豆花叶病毒的可测感度是0.35ng,芋花叶病毒为0.83ng。以含0.5%吐温20的Tris缓冲液封闭硝基纤维素薄膜固相载体的未结合部位,其效果与牛血清白蛋白相同。应用碱性磷酸酶标记抗体以及羟基吲哚磷酸盐和氮蓝四唑为底物较合适,这种底物可在室温下长期保存,且反应产物为不褪色的紫色。易于观察和保存。  相似文献

l-Sorbose reductase from Gluconobacter frateurii (SR) is an NADPH-dependent oxidoreductase. SR preferentially catalyzes the reversible reaction between d-sorbitol and l-sorbose with high substrate specificity. To elucidate the structural basis of the catalytic mechanism and the substrate specificity of SR, we have determined the structures of apo-SR, SR in complex with NADPH, and the inactive mutant (His116Leu) of SR in complex with NADPH and l-sorbose at 2.83 Å, 1.90 Å, and 1.80 Å resolutions, respectively. Our results show that SR belongs to the short-chain dehydrogenase/reductase (SDR) family and forms a tetrameric structure. Although His116 is not conserved among SDR family enzymes, the structures of SR have revealed that His116 is important for the stabilization of the proton relay system and for active-site conformation as a fourth catalytic residue. In the ternary complex structure, l-sorbose is recognized by 11 hydrogen bonds. Site-directed mutagenesis of residues around the l-sorbose-binding site has shown that the loss of almost full enzymatic activity was caused by not only the substitution of putative catalytic residues but also the substitution of the residue used for the recognition of the C4 hydroxyl groups of l-sorbose (Glu154) and of the residues used for the construction of the substrate-binding pocket (Cys146 and Gly188). The recognition of the C4 hydroxyl group of l-sorbose would be indispensable for the substrate specificity of SR, which recognizes only l-sorbose and d-sorbitol but not other sugars. Our results indicated that these residues were crucial for the substrate recognition and specificity of SR.  相似文献

Pueraria mirifica-derived tuberous powder has been long-term consumed in Thailand as female hormone-replacement traditional remedies. The protein profiles of tubers collected in different seasons were evaluated. Phenol extraction, 2D-PAGE, and mass spectrometry were employed for tuberous proteome analysis. Out of the 322 proteins detected, over 59% were functionally classified as being involved in metabolism. The rest proteins were involved in defense, protein synthesis, cell structure, transportation, stress, storage, and also unidentified function. The proteins were found to be differentially expressed with respect to harvest season. Importantly, chalcone isomerase, isoflavone synthase, cytochrome p450, UDP-glycosyltransferase, and isoflavone reductase, which are all involved in the biosynthesis pathway of bioactive isoflavonoids, were most abundantly expressed in the summer-collected tubers. This is the first report on the proteomic patterns in P. mirifica tubers in relevant with seasonal variation. The study enlights the understanding of variance isoflavonoid production in P. mirifica tubers.  相似文献

Direct gene transfer to protoplasts is a widespread experimental procedure. In this review, it is discussed in relation to the fate and expression of transforming DNA and its genetic transmission upon integration, and expression (in)stability during successive meiotic generations. The results suggest that the major differences between Nicotiana plumbaginifolia and N. tabacum were not in DNA uptake but rather in the specific fate of the transforming DNA. Differences between integrative transformation versus transient gene expression are discussed. Transient gene expression experiments with cotransformed independent constructs enable accurate assays of gene activity modulation, including both activation and repression. The study of the time of integration of foreign DNA using haploid mesophyll protoplasts showed that foreign DNA integration took place during the first two division cycles of the protoplasts. Most of the transfomants inherited the foreign genes as a monogenic trait: N. plumbaginifolia exhibited a higher proportion of multiple insertions compared with N. tabacum . Furthermore, gene integration occurs as a random process. Compared to 'simple' transformation, cotransformation with independent constructs showed a higher level of rearrangement of the inserted DNA. Foreign gene transmission, followed through different generations, indicated that inactivation of the inserted gene could happen through a variety of processes. These results open up a series of questions concerning the applied aspects of direct gene transfer to plants.  相似文献

Eight species members of the Thai Hyrcanus Group were identified based on the intact morphology and molecular analysis (COI barcoding, 658 bp) of F1-progenies. Five iso-female lines of each species were pooled in order to establish stock colonies. A stenogamous colony of each species was investigated by making 200 and 300 newly emerged adult females and males co-habit in a 30 cm cubic cage for one week. After ovipositon, the spermathecae of females were examined for sperms. The results revealed that Anopheles argyropus, Anopheles crawfordi, Anopheles nitidus, Anopheles pursati, Anopheles sinensis, Anopheles nigerrimus, Anopheles paraliae and Anopheles peditaeniatus yielded insemination rates of 0%, 0%, 0%, 31%, 33%, 42%, 50% and 77%, respectively. Continuous selection to establish stenogamous colonies indicated that An. sinensis, An. pursati, An. nigerrimus, An. paraliae and An. peditaeniatus provided insemination rates of 33–34%, 27–31%, 42–58%, 43–57% and 61–86% in 1, 2, 5, 6 and 20 generations of passages, respectively.  相似文献
