Linocarpon species are reported from Pandanaceae in Australia, Brunei, Hong Kong, Nepal, New Zealand, Philippines, Seychelles, Thailand and Vanuatu. Linocarpon lammiae sp. nov. were collected on decaying leaves of Pandanus tectorius in Hong Kong. Linocarpon siamensis sp. nov. and L. suthepensis sp. nov. were collected from decaying leaves of P. penetrans in Thailand. These taxa are described, illustrated and compared with Linocarpon species with similar ascospore morphology and dimensions. Included are a synoptic table, which compares the new species to similar known species, and a dichotomous key to species of Linocarpon known from members of the Pandanaceae. 相似文献

Neonates are thought to mount less vigorous adaptive immune responses than adults to antigens and infectious agents. This concept has led to a delay in the administration of many currently available vaccines until late infancy or early childhood. It has recently been shown that vaccines composed of plasmid DNA can induce both humoral and cell-mediated antimicrobial immunity when administered within hours of birth. In most of these studies, immune responses were measured weeks or months after the initial vaccination, and it is therefore questionable whether the observed responses were actually the result of priming of splenocytes within the neonatal period. Here we show that DNA vaccination at birth results in the rapid induction of antigen-specific CD8(+) T cells within neonatal life. Analyses of T-cell effector functions critical for the resolution of many viral infections revealed that neonatal and adult CD8(+) T cells produce similar arrays of cytokines. Furthermore, the avidities of neonatal and adult CD8(+) T cells for peptide and the rapidity with which they upregulate cytokine production after recall encounters with antigen are similar. Protective immunity against the arenavirus lymphocytic choriomeningitis virus, which is mediated by CD8(+) cytotoxic T cells, is also rapidly acquired within the neonatal period. Collectively these data imply that, at least in the case of CD8(+) T cells, neonates are not as immunodeficient as previously supposed and that DNA vaccines may be an effective and safe means of providing critical cell-mediated antiviral immunity extremely early in life. 相似文献

Kinetic parameters for nitrate reduction in intact sediment cores were investigated by using the acetylene blockage method at five sites along the Swale-Ouse river system in northeastern England, including a highly polluted tributary, R. Wiske. The denitrification rate in sediment containing added nitrate exhibited a Michaelis-Menten-type curve. The concentration of nitrate for half-maximal activity (Kmap) by denitrifying bacteria increased on passing downstream from 13.1 to 90.4 μM in the main river, but it was highest (640 μM) in the Wiske. The apparent maximal rate (Vmaxap) ranged between 35.8 and 324 μmol of N m−2 h−1 in the Swale-Ouse (increasing upstream to downstream), but it was highest in the Wiske (1,194 μmol N m−2 h−1). A study of nitrous oxide (N2O) production at the same time showed that rates ranged from below the detection limit (0.05 μmol of N2O-N m−2 h−1) at the headwater site to 27 μmol of N2O-N m−2 h−1 at the downstream site. 1. A survey was made of denitrification and nitrous oxide (N2 O) production in river sediments at fifty sites in north‐east England during one season in order to investigate the relationship between rates and environmental factors likely to influence these processes. The sites were chosen to represent a wide range of physical and chemical conditions. Denitrification rate and N2 O production were measured within 5 h of sampling using the slurry acetylene blockage technique.
2. Denitrification rate ranged from less than 0.005–260 nmol N g–1 DW h–1 , tending to increase in a downstream direction. N2 O production ranged from negative values (net consumption) to 13 nmol N2 O‐N g–1 DW h–1 and accounted for 0–115% of the N gases produced.
3. Denitrification rate and N2 O concentration in the sediment were correlated positively with nitrate concentration in the water column, water content of the sediment and percentage of fine (< 100 μm) particles in the sediment.
4. The variation in denitrification rate was satisfactorily explained (64% total variance) by a model employing measurements of water nitrate and water content of sediments. No simple or multiple relationship was found for N2 O production. 相似文献

Studies in IFN-gamma-deficient mice suggest that the delivery of IFN-gamma to CD8(+) T cells early in virus infection programs their eventual contraction, thereby reducing the abundance of CD8(+) memory T cells. In this study, we show that such mice fail to completely eliminate virus infection and that, when evaluated without the confounding factor of persisting Ag, both CD4(+) and CD8(+) T cells undergo profound contraction when they are unable to receive IFN-gamma signals. Furthermore, the abundance of CD4(+) and CD8(+) memory cells that express the IFN-gamma receptor is approximately 100-fold higher than cells lacking this molecule. Thus, direct IFN-gamma signaling is not required for T cell contraction during virus infection, and it enhances, rather than suppresses, the development of virus-specific CD4(+) and CD8(+) T cell memory. 相似文献

A study was made of the aquatic environment, tissue nutrient composition and surface phosphatase activities of the aquatic
moss Warnstorfia fluitans in Brandon Pithouse Stream, a small acidic stream in N-E England. The water, which originates from an underground spring,
had been pH 2.6 for at least 30 years, but about 3.9 during the present study. The moss was by far the most abundant phototroph
during all this period. Seasonal changes in aqueous nitrogen and phosphorus fractions were measured over a 2-year period near
the source. Most of the filtrable N and P were at times organic, but the very high N:P ratio (even if organic N is excluded)
suggests that only organic phosphate is likely to be important for the moss. There was a high peak in organic phosphate in
late spring in both study years. Surface phosphomonoesterase (PMEase) and phosphodiesterase (PDEase) activities were highly
correlated in the field and in axenic culture, though there were some differences in response to environmental factors. Axenic
material showed higher PMEase and PDEase activities when grown with organic P than with inorganic P. Although the data suggest
that internal P content is an important factor influencing phosphatase activities, PDEase activity was especially marked when
the moss was grown with the diester, DNA, as P source, indicating that at least one of its surface phosphatases can also respond
directly to the environment.

Individual chitin synthase enzymes synthesize microfibrils of differing structure at specific locations in the Candida albicans cell wall 总被引:1,自引:1,他引:0
The shape and integrity of fungal cells is dependent on the skeletal polysaccharides in their cell walls of which beta(1,3)-glucan and chitin are of principle importance. The human pathogenic fungus Candida albicans has four genes, CHS1, CHS2, CHS3 and CHS8, which encode chitin synthase isoenzymes with different biochemical properties and physiological functions. Analysis of the morphology of chitin in cell wall ghosts revealed two distinct forms of chitin microfibrils: short microcrystalline rodlets that comprised the bulk of the cell wall; and a network of longer interlaced microfibrils in the bud scars and primary septa. Analysis of chitin ghosts of chs mutant strains by shadow-cast transmission electron microscopy showed that the long-chitin microfibrils were absent in chs8 mutants and the short-chitin rodlets were absent in chs3 mutants. The inferred site of chitin microfibril synthesis of these Chs enzymes was corroborated by their localization determined in Chsp-YFP-expressing strains. These results suggest that Chs8p synthesizes the long-chitin microfibrils, and Chs3p synthesizes the short-chitin rodlets at the same cellular location. Therefore the architecture of the chitin skeleton of C. albicans is shaped by the action of more than one chitin synthase at the site of cell wall synthesis. 相似文献

辽宁是东北林蛙Rana dybowskii主要分布地之一,种群数量巨大,其群体遗传多样性有待评估.本研究应用ISSR标记技术对东北林蛙4个种群105个样本进行研究,5个引物共获得44条清晰谱带,4个种群的多态位点率均大于75%,Nei's基因多样性为0.2851,Shannon信息指数为0.4476,显示了较高的遗传多样性.对遗传分化系数、Nei's遗传距离、AMOVA分子变异巢式方差分析和F-统计量等遗传参数的统计结果表明,辽宁东北林蛙种群间已经出现一定程度的遗传分化,分析认为,自然屏障(高山和平原等)以及栖息地片段化是其遗传分化形成的主要因素. 相似文献
