Dichelobacter nodosus is the essential causative agent of footrot in sheep. The major D. nodosus-encoded virulence factors that have been implicated in the disease are type IV fimbriae and extracellular proteases. To examine the role of the fimbriae in virulence, allelic exchange was used to insertionally inactivate the fimA gene, which encodes the fimbrial subunit protein, from the virulent type G D. nodosus strain VCS1703A. Detailed analysis of two independently derived fimA mutants revealed that they no longer produced the fimbrial subunit protein or intact fimbriae and did not exhibit twitching motility. In addition, these mutants were no longer capable of undergoing natural transformation and did not secrete wild-type levels of extracellular proteases. These effects were not due to polar effects on the downstream fimB gene because insertionally inactivated fimB mutants were not defective in any of these phenotypic tests. Virulence testing of the mutants in a sheep pen trial conducted under controlled environmental conditions showed that the fimA mutants were avirulent, providing evidence that the fimA gene is an essential D. nodosus virulence gene. These studies represent the first time that molecular genetics has been used to determine the role of virulence genes in this slow growing anaerobic bacterium.  相似文献

Immunoglobulins (Ig) in serum from barramundi vaccinated with bovine serum albumin (BSA) were purified by ammonium sulphate precipitation and affinity chromatography using BSA as the ligand. The BSA-binding activity of eluted putative Ig fractions was assessed by enzyme-linked immunosorbent assay (ELISA) before being pooled and characterised by sodium dodecyl sulphate polyacrylamide gel electrophoresis (SDS-PAGE). Double affinity purification did not improve the purity of the Ig preparation compared to single affinity purification. Barramundi Ig were injected into sheep to produce anti-Ig antisera which were assessed in an indirect ELISA as the secondary antibody to detect serum Ig in barramundi vaccinated with Cryptocaryon irritans theronts. Affinity-purified Ig induced a more specific reagent for use as secondary antibody in ELISA than did normal whole-barramundi sera. The heavy (H) chain of barramundi Ig had an apparent molecular weight of 70 kDa while that of the light (L) chain was 27 kDa in SDS-PAGE studies. Under non-reducing conditions 2 putative populations of Ig were identified, at 768 and 210 kDa. The N-terminal sequence of the barramundi Ig H chain showed 78% homology with channel catfish Ictalurus punctatus Ig H chain sequence.  相似文献

Pathological findings associated with helminth and arthropod parasites of four wild platypuses (Ornithorhynchus anatinus) are described. Fourth-stage larval rhabditoid nematodes (?Cylindrocorporidae) found partially embedded in the stratum corneum were associated with mild acanthosis, orthokeratotic hyperkeratosis and inconstant subacute dermatits. A filarioid nematode (?Lemdaninae) was found in hypodermal lymphoid tissue. A larval trichostrongyloid nematode was found in the dermis, with no evidence of host response. An undescribed species of trombiculid mite ("chigger") was found in the pelage and attached to superficial stratum corneum. Focal inclusions of eosinophilic material within stratum corneum, interpreted as mite gel saliva produced during formation of the "stylostome," were also observed. The tick Ixodes ornithorhynchi punctured the epidermis causing adnexal trauma, dermal haemorrhage and chronic active dermatitis. The digenean Mehlisia ornithorhynchi was present in the small intestine of one animal and was associated with mild catarrhal enteritis.  相似文献

The morphology of three nematode taxa found in the skin of the platypus Ornithorhynchus anatinus (Monotremata) in southeastern New South Wales, Australia, is described and illustrated. Fourth-stage larvae belonging to the Rhabditoidea, ?Cylindrocorporidae, occurred primarily in superficial keratin and epidermal layers of the skin and in hair follicles. A nematode recognised by its conspicuous synlophe as a fourth-stage larva of the Trichostrongyloidea, of unknown genus and species but probably belonging to the molineid evolutionary line, was observed in histological section in the hypodermis. A gravid female nematode belonging to the Filarioidea, ?Lemdaninae, occurred in a lymphoid nodule in the hypodermis. O. anatinus is a protected species and these data are presented to draw the attention of parasitologists to skin as an important habitat for nematodes of the platypus.  相似文献

The analysis of two rodent X human somatic cell hybrids, carrying different inborn translocations of the human chromosome 14 long arm, has permitted us to narrow down the localization of the structural locus for alpha-1-antitrypsin (PI) to band 14q32.1, proximally to the highly polymorphic DNA locus D14S1 which has been localized by previous studies between 14q32.1 and 14q32.2. These data, evaluated in conjunction with other published information, suggest that the D14S1 locus is cytologically equidistant from both the PI locus and the complex locus for the immunoglobulin heavy chains (IGH) but, genetically, it appears much closer to the latter since the recombination frequency reported between the IGH complex and PI is six times greater than that between the IGH complex and D14S1 (lod score peaks respectively at 26% and 4% with narrow fiducial limits). The present report adds further strength to the frequently proposed hypothesis of a nonlinear relationship between cytologic and genetic distances of human genes. The possibility that this phenomenon may be a feature of frequent occurrence throughout the entire human genome is discussed.  相似文献

UDP-3-O-((R)-3-hydroxymyristoyl)-N-acetylglucosamine deacetylase (LpxC) is a zinc-dependent enzyme that catalyzes the deacetylation of UDP-3-O-((R)-3-hydroxymyristoyl)-N-acetylglucosamine to form UDP-3-O-(R-hydroxymyristoyl)glucosamine and acetate. The structural similarity of the active site of LpxC to metalloproteases led to the proposal that LpxC functions via a metalloprotease-like mechanism. The pH dependence of k(cat)/Km catalyzed by Escherichia coli and Aquifex aeolicus LpxC displayed a bell-shaped curve (EcLpxC yields apparent pKa values of 6.4+/-0.1 and 9.1+/-0.1), demonstrating that at least two ionizations are important for maximal activity. Metal substitution and mutagenesis experiments suggest that the basic limb of the pH profile is because of deprotonation of a zinc-coordinated group such as the zinc-water molecule, whereas the acidic limb of the pH profile is caused by protonation of either Glu78 or His265. Furthermore, the magnitude of the activity decreases and synergy observed for the active site mutants suggest that Glu78 and His265 act as a general acid-base catalyst pair. Crystal structures of LpxC complexed with cacodylate or palmitate demonstrate that both Glu78 and His265 hydrogen-bond with the same oxygen atom of the tetrahedral intermediate and the product carboxylate. These structural features suggest that LpxC catalyzes deacetylation by using Glu78 and His265 as a general acid-base pair and the zinc-bound water as a nucleophile.  相似文献

Experiments were conducted in commercial tomato, Lycopersicon esculentum Miller (Solanaceae), greenhouses to compare the relative foraging effort of two bumble bee species, Bombus occidentalis Greene and Bombus impatiens Cresson, to examine interspecific competition between B. occidentalis and B. impatiens, and to determine whether bumble bee colonies grew to their full population potential in commercial tomato greenhouses. B. impatiens colonies had more brood and workers and made more foraging trips per hour than B. occidentalis colonies. However, B. impatiens returned to the colony without pollen loads and left their colonies without dropping off their pollen loads more frequently than B. occidentalis greenhouse colonies. Our data also suggest that the presence of B. impatiens had a detrimental effect on B. occidentalis populations. Furthermore, B. occidentalis colonies did not grow to their full population potential in tomato greenhouses, with fewer workers in greenhouse colonies than in colonies placed outside in a natural environment, or in colonies that were physically enclosed and protected from external mortality. Together, this study suggests that B. impatiens is a better pollinator than B. occidentalis. It also shows that unknown factors are limiting the size of B. occidentalis colonies in tomato greenhouses.  相似文献

Dendromonocotyle pipinna n. sp. (Monogenea: Monocotylidae) is described from the dorsal skin surface of Taeniura meyeni Müller & Henle from a public aquarium in Mooloolaba, Queensland, Australia. D. torosa n. sp. was found on the dorsal skin surface of a heavily infected specimen of Aetobatus narinari (Euphrasen) during a post-mortem at the Mooloolaba aquarium and on a specimen of A. narinari kept in an aquarium in Cairns. Both of these new Dendromonocotyle species are distinguished from each other and others in the genus by the morphology of the terminal papillary sclerite and the male copulatory organ.  相似文献

Empruthotrema stenophallus n. sp. (Monogenea: Monocotylidae) is described from specimens from the nasal tissue of the blue-spotted maskray Dasyatis kuhlii (Muller and Henle, 1841) collected in shallow waters off Pulau Banggi and Pulau Mabul, Sabah, Borneo, Malaysia. This is the first monogenean species to be described from an elasmobranch collected from Sabah. E. stenophallus can be distinguished from the other 6 members of the genus by the morphology of the sclerotized male copulatory organ, which is narrow, short, and distally tapered. E. dasyatidis Whittington and Kearn, 1992, previously documented from the nasal tissue of several of elasmobranch species from Australia, is recorded from 8 host species distributed around Malaysian Borneo. These represent new host and locality records for this monocotylid. The difficulties in identifying species of Empruthotrema and the apparent lack of host specificity by some members of the genus are discussed.  相似文献

The effects of non-native plants on habitat use are well studied; however, whether introduced plants negatively influence reproductive output of animal populations has received much less attention. We conducted a systematic literature review to evaluate the influence of non-native plants on reproductive performance in songbirds. Our global search resulted in 32 relevant articles, from which we compiled a dataset of 133 songbird responses to nesting in or around non-native vegetation. Reproductive metrics examined included measures of nest survival/mortality, productivity, fledgling survival, adult survival, nestling condition, and brood parasitism. Thirty-five percent of songbird reproductive responses were negative (n = 47), with 31% positive (n = 41) and 34% neutral (n = 45) responses found. Only 15% of responses were statistically significant effects (n = 20), and of these, negative effects were reported three times as often as positive effects. Non-significant trends were more prevalent (51% of responses), and the frequency of negative and positive trends was similar. The probability of finding a negative response (significant effect or non-significant trend) was higher for birds using introduced shrubs and wetland habitats. Mechanisms underlying responses were diverse, though similar drivers, such as differences in vegetation characteristics, predation pressure, and resource availability, were offered to explain both positive and negative effects. We found evidence for non-native plants as ecological traps in 39% of articles that assessed these phenomena (n = 16). This review highlights the sparsity of research targeting reproductive responses to plant invasion and synthesizes existing information to enhance our understanding of how birds respond to non-native plants. Our findings could be used to inform future research priorities in a world increasingly dominated by novel ecosystems.  相似文献
