Nurse sharks have not previously been known to migrate. Nurse sharks of the Dry Tortugas (DRTO) mating population have a highly predictable periodic residency cycle, returning to the Dry Tortugas Courtship and Mating Ground (DTCMG) annually (males) or bi- to triennially (females) during the June/July mating season. For 23 years we have followed the movements of 76 recaptured adults of a total of 115 tagged adults. Telemetry detections of 40 females tagged with acoustic transmitters show that most tagged and presumably post-partum females are continuously present in the DRTO in the fall, winter and early spring following the June mating season but these females depart in late March to early May. Detections reveal these females avoid the DTCMG completely during the next mating season, returning from late summer to fall. Telemetry records of nine of 17 adult males that co-habited with these females in the DTCMG depart DRTO waters every July. Both sexes may overwinter in the DRTO. Between 2011 and 2016 three males and five females with transmitters were detected to move up the west coast of Florida outside of the mating season as far north as the waters off Tampa Bay (335 km). Six others were only detected in the lower Florida Keys (292 km). Nine sharks returned to DRTO; one returned six times. Some overwintered and some resumed courtship in June, demonstrating both resident and migratory contingents within their population, partial migration and an ability to navigate with high spatial and temporal precision.  相似文献

The Disproportionate micromelia (Dmm) mouse has a three nucleotide deletion in Col2a1 in the region encoding the C-propeptide which results in the substitution of one amino acid, Asn, for two amino acids, Lys-Thr. Western blot and immunohistochemical analyses failed to detect type II collagen in the cartilage matrix of the homozygous mice and showed reduced levels in the matrix of heterozygous mice. Type II collagen chains localized intracellularly within the chondrocytes of homozygote and heterozygote tissues. These findings provide evidence that the expression of type II procollagen chains containing the defective C-propeptide results in an intracellular retention and faulty secretion of type II procollagen molecules. A complete absence of mature type II collagen from the homozygote cartilage and an insufficiency of type II collagen in the heterozygote cartilage explains the Dmm mouse phenotype. The integrity of the C-propeptide is thus crucial for the biosynthesis of normal type II collagen by chondrocytes.  相似文献

Zn2+-dependent deoxyribozymes that form natural and unnatural RNA linkages   总被引:1,自引:0,他引:1  
We report Zn(2+)-dependent deoxyribozymes that ligate RNA. The DNA enzymes were identified by in vitro selection and ligate RNA with k(obs) up to 0.5 min(-)(1) at 1 mM Zn(2+) and 23 degrees C, pH 7.9, which is substantially faster than our previously reported Mg(2+)-dependent deoxyribozymes. Each new Zn(2+)-dependent deoxyribozyme mediates the reaction of a specific nucleophile on one RNA substrate with a 2',3'-cyclic phosphate on a second RNA substrate. Some of the Zn(2+)-dependent deoxyribozymes create native 3'-5' RNA linkages (with k(obs) up to 0.02 min(-)(1)), whereas all of our previous Mg(2+)-dependent deoxyribozymes that use a 2',3'-cyclic phosphate create non-native 2'-5' RNA linkages. On this basis, Zn(2+)-dependent deoxyribozymes have promise for synthesis of native 3'-5'-linked RNA using 2',3'-cyclic phosphate RNA substrates, although these particular Zn(2+)-dependent deoxyribozymes are likely not useful for this practical application. Some of the new Zn(2+)-dependent deoxyribozymes instead create non-native 2'-5' linkages, just like their Mg(2+) counterparts. Unexpectedly, other Zn(2+)-dependent deoxyribozymes synthesize one of three unnatural linkages that are formed upon the reaction of an RNA nucleophile other than a 5'-hydroxyl group. Two of these unnatural linkages are the 3'-2' and 2'-2' linear junctions created when the 2'-hydroxyl of the 5'-terminal guanosine of one RNA substrate attacks the 2',3'-cyclic phosphate of the second RNA substrate. The third unnatural linkage is a branched RNA that results from attack of a specific internal 2'-hydroxyl of one RNA substrate at the 2',3'-cyclic phosphate. When compared with the consistent creation of 2'-5' linkages by Mg(2+)-dependent ligation, formation of this variety of RNA ligation products by Zn(2+)-dependent deoxyribozymes highlights the versatility of transition metals such as Zn(2+) for mediating nucleic acid catalysis.  相似文献

Previous studies have examined the isometric contraction properties of the two heads of the cat flexor carpi ulnaris acting as a single unit. In this study, the contraction properties and fiber architecture of each head of the flexor carpi ulnaris were determined separately and related to previous reports on the histochemical characteristics of this muscle. The morphology of retrograde-labeled motor nuclei for the two heads of the muscle was also examined. The humeral head had a significantly longer contraction time (48 msec) than the ulnar head (36 msec) as well as a significantly lower tetanic fusion frequency (28 Hz vs. 35 Hz). The maximum tetanic tension per gram of muscle tissue was 71% greater in the ulnar head. Motoneurons of the flexor carpi ulnaris formed a column 12 mm long and 0.5 mm wide in the center of the ventral grey in spinal segments C8 and T1. The ulnar head had alpha-motoneurons with greater soma diameters than those in the humeral head. The smaller soma diameter, slower contraction time, and weaker contraction in the humeral head correlate with the preponderance of oxidative-metabolic muscle fiber types found in the humeral head by other workers. These correlations suggest that the humeral head plays a major role in maintaining a sustained antigravity tension that prevents the wrist from buckling during standing.  相似文献

The response of marine bacteria to Corexit 9527, with and without Prudhoe Bay crude oil labeled withn–(1–14C)hexadecane, in a temperate pelagic environment was monitored over 22 days using controlled ecosystem enclosures. The results indicated that Corexit and Corexit-dispersed crude oil stimulated bacterial production by serving as substrates and/or by inducing the release of organic compounds from the indigenous phytoplankton population. Highest bacterial standing stock was observed in the enclosure treated with a mixture of Corexit and crude oil, in which a large fraction of the predominant bacterivores were eliminated. Biodegradation appeared to be more significant than abiotic processes in contributing to the loss of low volatility n-alkanes in Corexit-dispersed oil. Twenty-two days following its addition, 50% of the radiotracer was recovered: 3% in the suspended particulate fraction, 10% in sedimentary material, 36% as CO2, and less than 1% in the dissolved organic pool.  相似文献
