Background

Application of plasmid DNA for immunization of food-producing animals established new standards of food safety. The addition of foreign products e.g. pDNA into the food chain should be carefully examined to ensure that neither livestock animals nor consumers develop unpredicted or undesirable side-effects.

Methods

A quantitative real-time PCR (QRTPCR) methodology was developed to study the biodistribution and persistence of plasmid DNA vaccine pDNAX (pVAX-Hsp60 TM814) in mice and beef cattle. The linear quantification range and the sensitivity of the method was found to be 10 – 109 copies per reaction (500 ng/gDNA) and 3 copies per reaction, respectively.

Results

Persistence of pDNAX in mice muscle tissue was restricted to injection site and the amount of pDNAX showed delivery formulation dependent (naked pDNA, electroporation, cationic liposome complexes) and mouse age-dependent clearance form injection site but pDNAX was still detectable even after 365 days. The QRTPCR analysis of various muscle tissue samples of vaccinated beef bulls performed 242–292 days after the last revaccination proved that residual pDNAX was found only in the injection site. The highest plasmid levels (up to 290 copies per reaction) were detected in the pDNAX:CDAN/DOPE group similarly to mice model. No pDNA was detected in the samples from distant muscles and draining lymph nodes.

Conclusion

Quantitative real-time PCR (QRTPCR) assay was developed to assess the residual pDNA vaccine pVAX-Hsp60 TM814 in mice and beef cattle. In beef cattle, ultra low residual level of pDNA vaccine was only found at the injection site. According to rough estimation, consumption of muscles from the injection site represents almost an undetectable intake of pDNA (400 fg/g muscle tissue) for consumers. Residual plasmid in native state will hardly be found at measurable level following further meat processing. This study brings supportive data for animal and food safety and hence for further approval of pDNA vaccine field trials.  相似文献

Human adenovirus-36 (Ad-36) increases adiposity in chickens, mice, and nonhuman primates and is associated with human obesity. Ad-36 paradoxically reduces serum cholesterol and triglycerides in animal models. Ad-36 increases adipocyte differentiation and triglyceride accumulation in 3T3-L1 cells in vitro. This study evaluated whether three other human adenoviruses increase adiposity in chickens and in 3T3-L1 cells in vitro. We inoculated chickens with human Ad-2, Ad-31, Ad-37, or media at age 3 wk. Food intake and weights were recorded for 3.5 wk, and then chickens were killed and visceral fat, body composition, serum lipids, and viral antibody status were determined. Visceral fat and total body fat were significantly elevated (P < 0.001) in the Ad-37 group compared with all other groups. Final body weights were higher in chickens inoculated with Ad-37 compared with Ad-2, but not significantly higher than in control or Ad-31 groups. Food intake did not differ among groups. Serum cholesterol was elevated in Ad-37 chickens compared with control (P < 0.01) but was not affected by other viruses. Triglycerides were reduced in Ad-37 chickens (P < 0.0001) but were not affected by other viruses. In 3T3-L1 cells in vitro, Ad-31, Ad-36, and Ad-37, but not Ad-2, increased adipocyte differentiation and triglycerides accumulation. In summary, Ad-37 is another human adenovirus that increases adiposity and reduces serum triglycerides in an animal model. However, the response of serum cholesterol is opposite that of Ad-36. Evaluation of other human adenoviruses to determine their effects on adiposity and serum lipids is warranted, but in vitro assays may not be definitive for this purpose.  相似文献

BACKGROUND: Serological tests to detect antibodies specific to Plasmodium vivax could be a valuable tool for epidemiological studies, for screening blood donors in areas where the malaria is not endemic and for diagnosis of infected individuals. Because P. vivax cannot be easily obtained in vitro, ELISA assays using total or semi-purified antigens are rarely used. Based on this limitation, we tested whether recombinant proteins representing the 19 kDa C-terminal region of the merozoite surface protein-1 of P. vivax (MSP119) could be useful for serological detection of malaria infection. METHODS: Three purified recombinant proteins produced in Escherichia coli (GST-MSP119, His6-MSP119 and His6-MSP119-PADRE) and one in Pichia pastoris (yMSP119-PADRE) were compared for their ability to bind to IgG antibodies of individuals with patent P. vivax infection. The method was tested with 200 serum samples collected from individuals living in the north of Brazil in areas endemic for malaria, 53 serum samples from individuals exposed to Plasmodium falciparum infection and 177 serum samples from individuals never exposed to malaria. RESULTS: Overall, the sensitivity of the ELISA assessed with sera from naturally infected individuals was 95%. The proportion of serum samples that reacted with recombinant proteins GST-MSP119, His6-MSP119, His6-MSP119-PADRE and yMSP119-PADRE was 90%, 93.5%, 93.5% and 93.5%, respectively. The specificity values of the ELISA determined with sera from healthy individuals and from individuals with other infectious diseases were 98.3% (GST-MSP119), 97.7% (His6-MSP119 and His6-MSP119-PADRE) or 100% (yMSP119-PADRE). CONCLUSIONS: Our study demonstrated that for the Brazilian population, an ELISA using a recombinant protein of the MSP119 can be used as the basis for the development of a valuable serological assay for the detection of P. vivax malaria.  相似文献

Background

For membrane proteins, lipids provide a structural framework and means to modulate function. Paired connexin hemichannels form the intercellular channels that compose gap junction plaques while unpaired hemichannels have regulated functions in non-junctional plasma membrane. The importance of interactions between connexin channels and phospholipids is poorly understood.

Results

Endogenous phospholipids most tightly associated with purified connexin26 or connexin32 hemichannels or with junctional plaques in cell membranes, those likely to have structural and/or modulatory effects, were identified by tandem electrospray ionization-mass spectrometry using class-specific interpretative methods. Phospholipids were characterized by headgroup class, charge, glycerol-alkyl chain linkage and by acyl chain length and saturation. The results indicate that specific endogenous phospholipids are uniquely associated with either connexin26 or connexin32 channels, and some phospholipids are associated with both. Functional effects of the major phospholipid classes on connexin channel activity were assessed by molecular permeability of hemichannels reconstituted into liposomes. Changes to phospholipid composition(s) of the liposome membrane altered the activity of connexin channels in a manner reflecting changes to the surface charge/potential of the membrane and, secondarily, to cholesterol content. Together, the data show that connexin26 and connexin32 channels have a preference for tight association with unique anionic phospholipids, and that these, independent of headgroup, have a positive effect on the activity of both connexin26 and connexin32 channels. Additionally, the data suggest that the likely in vivo phospholipid modulators of connexin channel structure-function that are connexin isoform-specific are found in the cytoplasmic leaflet. A modulatory role for phospholipids that promote negative curvature is also inferred.

Conclusion

This study is the first to identify (endogenous) phospholipids that tightly associate with connexin channels. The finding that specific phospholipids are associated with different connexin isoforms suggests connexin-specific regulatory and/or structural interactions with lipid membranes. The results are interpreted in light of connexin channel function and cell biology, as informed by current knowledge of lipid-protein interactions and membrane biophysics. The intimate involvement of distinct phospholipids with different connexins contributes to channel structure and/or function, as well as plaque integrity, and to modulation of connexin channels by lipophilic agents.  相似文献

Collecting natural data at regular, fine scales is an onerous and often costly procedure. However, there is a basic need for fine scale data when applying inductive methods such as neural networks or genetic algorithms for the development of ecological models. This paper will address the issues involved in interpolating data for use in machine learning methods by considering how to determine if a downscaling of the data is valid. The approach is based on a multi-scale estimate of errors. The resulting function has similar properties to a time series variogram; however, the comparison at different scales is based on the variance introduced by rescaling from the original sequence. This approach has a number of properties, including the ability to detect frequencies in the data below the current sampling rate, an estimate of the probable average error introduced when a sampled variable is downscaled and a method for visualising the sequences of a time series that are most susceptible to error due to sampling. The described approach is ideal for supporting the ongoing sampling of ecological data and as a tool for assessing the impact of using interpolated data for building inductive models of ecological response.  相似文献

This 2-year field study examined stomatal conductance, photosynthesis, and biomass allocation of Liquidambar styraciflua saplings in response to below- and aboveground competition with the vines Lonicera japonica and Parthenocissus quinquefolia. Vine competition did not affect stomatal conductance of the host trees. The leaf photosynthetic capacity and photosynthetic nitrogen-use efficiency were significantly reduced by root competition with vines, either singly or in combination with aboveground competition, early in the second growing season. However, such differences disappeared by the end of the second growing season. Trees competing below ground with vines also had lower allocation to leaves compared with steins. Aboveground competition with vines resulted in reduced photosynthetic capacity per unit leaf area, but not per unit leaf weight, in trees. No correlation was found between single leaf photosynthetic capacity and tree growth. In contrast, a high positive correlation existed between allocation to leaves and diameter growth. Results from this study suggest that allocation patterns are more affected than leaf photosynthesis in trees competing with vines.  相似文献

The fungus Cochliobolus carbonum causes leaf spot disease of maize. Highly virulent isolates of the pathogen produce a host-selective, peptide toxin that is active against susceptible genotypes of maize. Prior to infection, spores must germinate and differentiate appressoria, structures specialized for leaf penetration. Analysis of spore germination fluids by plasma desorption mass spectrometry, which allowed detection of as little as 0.5 ng toxin, revealed that spores induced to form appressoria in vitro synthesized and released the toxin at a time coincident with maturation of appressoria. Spores incubated under conditions that did not induce appressorium formation failed to produce toxin. These observations indicate that synthesis of the host-selective toxin, which is essential for successful pathogenesis of maize by C. carbonum, is regulated by infection-related morphogenesis.  相似文献

  1. Energy pathways in stream food webs are often driven by allochthonous basal resources. However, allochthonous dissolved organic carbon (DOC) is generally viewed as a minor if not insignificant basal resource because much of the DOC pool comprises high molecular weight, recalcitrant compounds and is inefficiently incorporated into biomass. Nevertheless, there is increasing evidence that the relatively small, labile fraction of DOC may indeed fuel microbial activity to a level that stimulates productivity across multiple trophic levels, resulting in a trophic upsurge. Here, we tested the trophic upsurge hypothesis by subsidising the labile DOC pool of an Alaskan boreal stream that had relatively high nutrient availability but low levels of naturally occurring DOC.
  2. We continuously added ecologically relevant (0.250 mg C/L, c. 10% increase above ambient bulk DOC) concentrations of labile DOC (acetate-C) for 62 days to a treatment reach that was statistically indistinguishable in its channel form and chemistry from an upstream reference reach. We measured responses of periphyton production and biomass, whole reach metabolism and nutrient uptake, benthic invertebrate abundances, and juvenile salmonid (Dolly Varden, Salvelinus malma) abundance and growth.
  3. Measurements of basal ecosystem responses collectively indicated increased energy mobilization at the base of the food web in response to labile DOC addition. Periphyton bacterial production in the treatment reach was generally >1.5× reference reach values, and periphyton ash-free dry mass, chlorophyll-a, and chlorophyll-a:ash-free dry mass were all greater in the treatment reach by the end of the study. Throughout dosing, ecosystem respiration was 1.3× greater in the treatment reach and dissolved inorganic nitrogen uptake was greater in the treatment reach on eight out of nine measurements.
  4. Benthic invertebrate counts, dominated by Baetis spp. and Chironomidae, were c. 4× greater after 28 dosing days and c. 8× greater after 56 days in the upstream portion of the treatment reach. Abundance generally declined with increasing distance from the dosing station. Dolly Varden fry and parr age classes were nearly 2× more abundant in the upstream portion of the treatment reach than in any section of the reference reach and also declined with increasing distance from the dosing station. Further, Dolly Varden tagged with passive integrated transponders prior to the experiment had significantly higher instantaneous growth rates in the treatment reach than those recaptured in the reference reach.
  5. The strong consumer responses to small quantities of labile DOC mirrored significant treatment reach increases in basal ecosystem function and therefore demonstrated a response consistent with a trophic upsurge. Terrestrial DOC has historically been viewed as contributing little to metazoan consumers, instead modulating the influence of nutrients and being respired out of a disconnected microbial loop. Because we dosed the treatment reach with a relevant concentration of labile DOC, based on measurements in nearby peatland-draining streams, we suggest that terrestrial DOC deserves more attention as a basal resource for whole food webs, akin to nutrients fuelling green (autochthonous) pathways.

The effects of petal-size manipulations on the behavior of pollinators and pollen/seed predators, and on pollen removal and deposition, were studied in Hibiscus moscheutos (Malvaceae) populations. The ultimate effects on the female reproductive success of flowers, such as fruit set, seed predation rate, and final seed set were also measured. We applied three levels of petal removal (100%, 50%, and 0% size reduction in radius) to flowers in natural populations. Two pollinators (Bombus pennsylvanicus and Ptilothrix bombiformis) ignored flowers without petals, suggesting that pollinators use petals as a visual cue to locate flowers. Consequently, 100% petal removal reduced female reproductive success considerably, mainly through a higher rate of fruit abortion due to failure of pollen deposition on stigmas. No significant differences between the 50% petal removal treatment and uncut control were detected in any components of female success examined. The results, therefore, suggest that differences in petal size have little influence on female reproductive success of Hibiscus flowers at our study site. Final seed set varied considerably depending on the larval densities of two coleopteran seed predators (Althaeus hibisci and Conotrachelus fissunguis). A. hibisci responded to petal size, and a higher density of adults was found in flowers in which petal size had not been reduced. Because Althaeus feed on pollen as adults and no effect of petal size on seed predation was detected, the preference of Althaeus for larger flowers may represent a foraging strategy for adult beetles and may exert counteracting selection pressure on petal size through male reproductive success of flowers.
