We describe a new method for the transfer of carbohydrate moieties to polypeptides in which complex carbohydrate, in the form of glycosyl amino acid, is removed from an available glycoprotein, derivatized, and reacted with a polypeptide via an iodoacetylated alpha-amino group. A family of oligomannose chains, N-linked to the side chain of Asn, was obtained from ovalbumin by pronase digestion and purified as previously described. A reactive sulfhydryl group was specifically placed on these molecules by reaction of 2-iminothiolane with the Asn alpha-amino group. Separately, the alpha-amino group of the peptide GGYR was specifically iodoacetylated by reaction with iodoacetic anhydride at pH 6. Reaction of the thiol-containing carbohydrate with iodoacetylated peptide at pH 8 gave in high yield the corresponding oligomannosyl-peptides, whose structures were confirmed by mass spectrometry. A peptide inhibitor of HIV protease was also oligomannosylated by this procedure. The principle advantage of this method is the efficiency of the reaction even when performed with stoichiometric amounts of the two molecules at low concentration. It should be feasible to extend this chemistry to larger polypeptides.  相似文献

1. The loss of weight in pigeons subjected to prolonged starvation on water alone may be accurately represented by an appropriate form of the law of autocatakinesis. 2. This function, within proper limitations, gives a definite indication as to the nature and character of the fundamental reactions involved. In addition, it provides quantitative measures of the relative progress and importance of these reactions. 3. Analytically, the process of starvation consists of two nearly symmetrical cycles, corresponding chemically to the destruction of the carbohydrate-fat reserves and to the decomposition of body protein. 4. A suitable transformation overcoming certain difficulties inherent in the general form of the law is discussed and applied to the data reported.  相似文献

Construction of mathematical simulation models helps to organize current information and extend inferences from available data. During the past two decades, microbial ecology has undergone rapid developments in both quantity and quality of available data. In particular, considerable advances have been made in our knowledge of microbial food web dynamics in the Duplin River watershed at Sapelo Island, Georgia. Here we provide examples of how modeling and microbial ecology have interfaced. In the early 1970s, construction of a 14-compartment model of carbon flow through a salt marsh ecosystem aided in directing method development and field experiments on the sediment microbial community. In turn, the results of field experiments corroborated the model's postulated controls on the community. Also, during the past 12 years we have developed a series of simulation models reflecting the growing information on the aquatic microbial food web. Early models provided evidence for the microbial loop but illustrated the paucity of knowledge concerning controls for bacterial growth on detritus. Results from newer methods in microbial ecology and studies from the Duplin River have allowed us to construct a model which provides realistic simulations but is also highly sensitive to certain parameter value changes (e.g., in organic matter availability and grazing by protozoans). Thus improvements in model structure and corroboration of the models with extant data have been closely tied to methodological and conceptual advances in microbial ecology. The relationship is viewed as synergistic, as needs for model parameter values and equation forms have directed further development of methods, experimentation, and field observations.  相似文献

With populations of land snails of very small size like Vertiginidae,questions have arisen as to whether populations of relativelydistant islands in archipelagos are really isolated from eachother. Apart from other flight agencies, airborne transportof loose specimens is not improbable in stormy weather conditions.Currently, mechanisms of wind-borne transport of sand particlesover short and long distances have been intensively studied.The results are available in the literature on sediments, allowingthe calculation of probable flight distances for particles insuspension. For living snails of the Aegean species Truncatel-lina rothi,an average fall velocity of 2.6–2.7 m s–1 has beendetermined in experiments under laboratory conditions. Applyingthese results, Truncatellina living on an island at 100 m altitudeand close to the coast could be transported up to several kilometersin heavy storms, which are not uncommon in the Aegean archipelago(Greece). This would imply that many of the Aegean islands arenot effectively isolated for minute snail species, and thatgenetic interchange between island populations is probably frequent. DNA content and p53 protein expression in ductal breast cancer
The DNA content of 85 ductal breast cancers of different histological grades was evaluated using static cytometry and correlated with immunocytochemical expression of p53 protein in tumour cells in cytological material. A statistically significant difference was observed between p53 protein expression and grade of malignancy ( P <0.001). The percentage of euploid tumours significantly decreased from grade I through grade II to grade III tumours ( P <0.001). Clonal DNA heterogeneity was observed in 26.6% of cases analysed and was correlated with p53 protein expression ( P <0.001). These changes probably reflect genomic alterations which may affect potential malignancy of breast cancer.  相似文献

Creation of the big trophical niches of the winter pine shoot moth (Rhyacionia buoliana Schiff.) on susceptible trees of Pinus silvestris L. provoked the mass outbreak of the pest in the south areas in Ukraine. In its turn, high density of the pest induced processes of the aggressivity's growth of population. In so doing the wide range of adaptive zones represented by groups of the pines with different resistance is favourable for formation of aggressive biotypes. The spreading of the pest from weakened to more resistant groups on the pines is similar to the process of “drift”.
