Panicum sonorum in Sonoran Desert agriculture

Evidence of prehistoric domestication of plants native to southwestern United States is rare. Presented here are archaeological, historic and contemporary ethnobotanical data suggesting that a native grain, Panicum sonorum,was harvested, sown and culturally selected in the Sonoran Desert Region. This indicator crop, other new clarifications of indigenous crops, and water management practices all point to the distinctiveness of the region’s agricultural complex. As redefined here, the Sonoran Desert Agricultural Region is diverse in both native crops and in cultivars of introduced crops originating elsewhere.     

New map of bacteriophage lambda DNA.

A map of bacteriophage lambda was constructed, including accurate positions for all 41 cut sites made by 12 different restriction enzymes. Over 100 fragments from single, multiple, and partial enzyme digestions were measured versus standards that were calibrated with respect to DNA molecules of known sequence. The data were subjected to least-squares analysis to assign map coordinates. In no case did a fragment size predicted from the map differ from the measurement of the fragment by more than +/- 5%. This low error rate was consistent in all size ranges of fragments. The total length of lambda was calculated as 49,133 nucleotide pairs. This probably is accurate to within 500 base pairs.     

The heterologous expression potential of an acid-tolerant <Emphasis Type="Italic">Talaromyces pinophilus</Emphasis> β-glucosidase in <Emphasis Type="Italic">Saccharomyces cerevisiae</Emphasis>

A filamentous fungus displaying high cellulase activity was isolated from a compost heap with triticale (a wheat-rye hybrid) as the main constituent. It was preliminarily identified as a Talaromyces pinophilus species. A 2577 base pair β-glucosidase gene was cloned from complementary DNA and heterologously expressed in Saccharomyces cerevisiae. The recombinant β-glucosidase production profile was assessed and compared to that of the Saccharomycopsis fibuligera β-glucosidase which served as a benchmark. The enzyme was also characterised in terms of pH and temperature tolerance as well as response to inhibitors. Maximal extracellular β-glucosidase activity of 0.56 nkat/mg total protein was measured using p-nitrophenyl-β-D-glucopyranoside as substrate. The recombinant protein displayed a pH optimum of 4.0, and good thermostability as 70% of maximal enzyme activity was retained after 1 h at 60 °C. Activity of the recombinant β-glucosidase was adversely affected by the presence of glucose and ethanol at higher concentrations while xylose had no effect. The expression of the T. pinophilus β-glucosidase did not reach the same titres as for the benchmark; however, in the context of constructing a yeast strain for bioethanol production in a consolidated bioprocess, the enzyme may still show good potential.
