The major histocompatibility complex (MHC) has been studied in a multitude of mammals by now, but much less is known about
its organisation and variation in other vertebrate species. The mammalian MHC is organised as a single gene cluster, but recent
studies on birds suggest that this paradigm of MHC organisation has to be supplemented. The domestic chicken thus possesses
two separate gene clusters which both contain MHC class I and class II B genes, and we have shown that the ring‐necked pheasant
Phasianus colchicus also has two unlinked clusters of class II B genes. We are studying the effect of the MHC on mate choice,
survival and reproductive success in natural populations of birds and reptiles. For this reason, we are developing DNA techniques
to determine the animals' MHC genotype. The amplification of the hypervariable exon 3 of the class I gene from songbirds and
reptiles has provided us with species specific probes that can be used in Southern blot analysis. The first results indicate
very extensive variation in all studied species, that is starlings Sturnus vulgaris, great reed warblers Acrocephalus arundinaceus
and water pythons Liasis fuscus. The restriction fragment length polymorphism (RFLP) analysis also suggests that the number
of MHC genes is significantly larger in these species than in pheasants and domestic chickens.

Research was conducted to determine whether pelletized hyphae ofHirsutella rhossiliensissuppressed invasion of roots by the sugarbeet cyst nematodeHeterodera schachtiiin field microplots. The loamy sand in the microplots was infested withH. schachtiibut not withH. rhossiliensis.Alginate pellets, with or without hyphae ofH. rhossiliensis,were mixed into soil removed from the microplots (1 pellet/cm3of soil). The soil was placed in cylinders positioned vertically in microplots; cylinders (6/microplot) were 10.1 cm wide and 15.3 cm deep and contained 1200 cm3of soil. Pellets and soil also were placed in soil observation chambers, which were buried in the cylinders or kept at 20°C in moisture chambers in the laboratory. After 12 days, cabbage seeds were planted in each cylinder, and after 10 days of growth, the seedlings were removed from the soil andH. schachtiiin the roots were counted. The number ofH. schachtiiin roots was large and was unaffected by addition ofH. rhossiliensis.In soil observation chambers,H. rhossiliensisgrew vigorously from the pellets in heat-treated soil but not in nonheated soil, and enchytraeids and collembolans were observed near damaged pellets. A major goal in evolutionary biology is to understand the genetic basis of adaptive traits. In migratory birds, wing morphology is such a trait. Our previous work on the great reed warbler (Acrocephalus arundinaceus) shows that wing length is highly heritable and under sexually antagonistic selection. Moreover, a quantitative trait locus (QTL) mapping analysis detected a pronounced QTL for wing length on chromosome 2, suggesting that wing morphology is partly controlled by genes with large effects. Here, we re‐evaluate the genetic basis of wing length in great reed warblers using a genomewide association study (GWAS) approach based on restriction site‐associated DNA sequencing (RADseq) data. We use GWAS models that account for relatedness between individuals and include covariates (sex, age and tarsus length). The resulting association landscape was flat with no peaks on chromosome 2 or elsewhere, which is in line with expectations for polygenic traits. Analysis of the distribution of p‐values did not reveal biases, and the inflation factor was low. Effect sizes were however not uniformly distributed on some chromosomes, and the Z chromosome had weaker associations than autosomes. The level of linkage disequilibrium (LD) in the population decayed to background levels within c. 1 kbp. There could be several reasons to why our QTL study and GWAS gave contrasting results including differences in how associations are modelled (cosegregation in pedigree vs. LD associations), how covariates are accounted for in the models, type of marker used (multi‐ vs. biallelic), difference in power or a combination of these. Our study highlights that the genetic architecture even of highly heritable traits is difficult to characterize in wild populations. 相似文献

Sexual selection hypotheses stipulate that the major histocompatibility complex genes (MHC) constitute a key molecular underpinning for mate choice in vertebrates. The last four decades saw growing empirical literature on the role of MHC diversity and dissimilarity in mate choice for a wide range of vertebrate animals, but with mixed support for its significance in natural populations. Using formal phylogenetic meta‐analysis and meta‐regression techniques, we quantitatively review the existing literature on MHC‐dependent mating preferences in nonhuman vertebrates with a focus on the role of MHC diversity and dissimilarity. Overall, we found small, statistically nonsignificant, average effect sizes for both diversity‐ and dissimilarity‐based mate choice (r = 0.113 and 0.064, respectively). Importantly, however, meta‐regression models revealed statistically significant support regarding female choice for diversity, and choice for dissimilarity (regardless of choosy sex) only when dissimilarity is characterized across multiple loci. Little difference was found among vertebrate taxa; however, the lack of statistical power meant statistically significant effects were limited to some taxa. We found little sign of publication bias; thus, our results are likely to be robust. In light of our quantitative assessment, methodological improvements and fruitful future avenues of research are highlighted. 相似文献

Inbreeding depression may drive the evolution of inbreedingavoidance through dispersal and mate choice. In birds, manyspecies show female-biased dispersal, which is an effectiveinbreeding avoidance mechanism. In contrast, there is scarceevidence in birds for kin discriminative mate choice, whichmay, at least partly, reflect difficulties detecting it. First,kin discrimination may be realized as dispersal, and this isdifficult to distinguish from other causes of dispersal. Second,even within small, isolated populations, it is often difficultto determine the potential candidates available to a femalewhen choosing a mate. We sought evidence for inbreeding avoidancevia kin discrimination in a breeding population of great reedwarblers (Acrocephalus arundinaceus) studied over 17 years.Inbreeding depression is strong in the population, suggestingthat it would be adaptive to avoid relatives as mates. Detaileddata on timing of settlement and mate search movements madeit possible to identify candidate mates for each female, andlong-term pedigrees and resolved parentage enabled us to estimaterelatedness between females and their candidate mates. We foundno evidence for kin discrimination: mate choice was random withrespect to relatedness when all mate-choice events were considered,and, after correction for multiple tests, also in all breedingyears. We suggest that dispersal is a sufficient inbreedingavoidance mechanism in most situations, although the lack ofkin discriminative mate choice has negative consequences forsome females, because they end up mating with closely relatedmales that lowers their fitness. 相似文献

Proliferation of direct repeats near the Oenothera chloroplast DNA origin of replication 总被引:1,自引:0,他引:1
The spacer between the 16S and 23S rRNA genes of the chloroplast DNA has
been implicated as an origin of replication in several species of plants.
In the evening primrose, Oenothera, this site was found to vary greatly in
size, with plastid genomes (plastomes) being readily distinguished. To
determine whether plastome "strength" in transmission could be correlated
with variation at oriB, the 16S rRNA-trnI spacer was sequenced from five
plastomes. The size variation was found to be due to differential
amplification (and deletion) of combinations of sequences belonging to
seven families of direct repeats. From these comparisons, one short series
of direct repeats and one region capable of forming a hairpin structure
were identified as candidates for the factor that could be responsible for
the differences between strong and weak plastome types. Ample sequence
variation allowed phylogenetic inferences to be made about the
relationships among the plastomes. Phylogenetic trees also could be
constructed for most of the families of direct repeats. The amplifications
and deletions of repeats that account for the size variation at oriB are
proposed to have occurred through extensive replication slippage at this
site.

Isolation and characterization of turbot (Scophtalmus maximus)-associated bacteria with inhibitory effects against Vibrio anguillarum. 下载免费PDF全文
More than 400 isolates from the intestine and the external surface of farmed Scophtalmus maximus as well as from fish food and hatchery water were screened for inhibitory effects against the fish pathogen Vibrio anguillarum HI 11345 and seven other fish pathogens. The bacteria with inhibitory effects were then characterized with regard to their sites of colonization, especially the intestinal regions and sites within each region. Of the total number of bacterial isolates from the intestine, 28% were inhibitory against V. anguillarum HI 11345. A marine biochemical assay was used to order the inhibitory strains into different phena. Most inhibitory bacteria were found in the rinse and mucus fractions of the gastrointestinal tract. No correlations among the different phena, site of colonization, and inhibitory effect could be found; however, a biochemical diversity was noted in the strains with an inhibitory effect. Of the isolates with an inhibitory effect against V. anguillarum HI 11345, 60% had an inhibitory effect on five other fish-pathogenic serotypes of V. anguillarum. Inhibitory effects of the isolates were also shown against Aeromonas salmonicida and Aeromonas hydrophila. 相似文献

When the adult sex ratio differs between years in local populations, but still is predictable between adjacent years, it has been proposed that the best strategy would be to bias the offspring sex ratio in favour of the rare sex. We tested this hypothesis using a data set of great reed warbler offspring, sexed by molecular techniques, that were collected over 11 breeding seasons at two adjacent reed marshes. Three important assumptions for this hypothesis are fulfilled in the studied great reed warbler population. First, a substantial proportion of great reed warblers are living in small local populations where sex ratio distortions would be sufficiently large and common. Second, breeding adults and their offspring return to breed in the local population to a high degree. Third, females have a possibility to assess the breeding sex ratio before laying their eggs. At our study site, the breeding sex ratio was positively correlated between successive years. However, contrary to our prediction, female great reed warblers seemed not to adjust their offspring sex ratio in relation to the local breeding sex ratio. 相似文献
