Human impacts on the environment at local or regional scales largely depend on intrinsic characteristics of the population, such as household size, household number, and human population growth. These demographic factors can vary considerably among ethnic groups sharing similar ecological landscapes, yet the role of traditional cultural practices in shaping local environmental impacts is not well known for many parts of the world. We here quantify land-cover changes and their relation to the habitat of the endangered Rhinopithecus bieti in Tibet, in 2 areas populated by different ethnic groups (polyandrous Tibetans and monogamous Naxi) from 1986 to 2006. Results indicate that habitat of the monkey decreased greatly within our study area over the 20-yr period. Polyandrous and monogamous ethnic communities differed in household size, household number, population growth, and per capita and per household land use. The practice of polyandry by ethnic Tibetan appears to have reduced per capita resource consumption by reducing the growth of overall household number and increasing household size, which can mitigate the negative effects of higher human density and population growth on the environment. Ethnic Tibetan may also reduce land impacts by adhering to Buddhist customs and alternative, more sustainable means of livelihood. Accordingly, the protection of traditional cultural resources, such as polyandry and Buddhist beliefs, could be an effective way to aid biodiversity and environmental conservation efforts in this key ecosystem.  相似文献

Aquatic photosynthetic organisms, such as the green alga Chlamydomonas reinhardtii, respond to low CO(2) conditions by inducing a CO(2) concentrating mechanism (CCM). Carbonic anhydrases (CAs) are important components of the CCM. CAs are zinc-containing metalloenzymes that catalyze the reversible interconversion of CO(2) and HCO(3)(-). In C. reinhardtii, there are at least 12 genes that encode CA isoforms, including three alpha, six beta, and three gamma or gamma-like CAs. The expression of the three alpha and six beta genes has been measured from cells grown on elevated CO(2) (having no active CCM) versus cells growing on low levels of CO(2) (with an active CCM) using northern blots, differential hybridization to DNA chips and quantitative RT-PCR. Recent RNA-seq profiles add to our knowledge of the expression of all of the CA genes. In addition, protein content for some of the CA isoforms was estimated using antibodies corresponding to the specific CA isoforms: CAH1/2, CAH3, CAH4/5, CAH6, and CAH7. The intracellular location of each of the CA isoforms was elucidated using immunolocalization and cell fractionation techniques. Combining these results with previous studies using CA mutant strains, we will discuss possible physiological roles of the CA isoforms concentrating on how these CAs might contribute to the acquisition and retention of CO(2) in C. reinhardtii.  相似文献

The predominant strategy for using algae to produce biofuels relies on the overproduction of lipids in microalgae with subsequent conversion to biodiesel (methyl-esters) or green diesel (alkanes). Conditions that both optimize algal growth and lipid accumulation rarely overlap, and differences in growth rates can lead to wild species outcompeting the desired lipid-rich strains. Here, we demonstrate an alternative strategy in which cellulose contained in the cell walls of multicellular algae is used as a feedstock for cultivating biofuel-producing microorganisms. Cellulose was extracted from an environmental sample of Cladophora glomerata-dominated periphyton that was collected from Lake Mendota, WI, USA. The resulting cellulose cake was hydrolyzed by commercial enzymes to release fermentable glucose. The hydrolysis mixture was used to formulate an undefined medium that was able to support the growth, without supplementation, of a free fatty acid (FFA)-overproducing strain of Escherichia coli (Lennen et. al 2010). To maximize free fatty acid production from glucose, an isopropyl β-D-1-thiogalactopyranoside (IPTG)-inducible vector was constructed to express the Umbellularia californica acyl-acyl carrier protein (ACP) thioesterase. Thioesterase expression was optimized by inducing cultures with 50 μM IPTG. Cell density and FFA titers from cultures grown on algae-based media reached 50% of those (~90 μg/mL FFA) cultures grown on rich Luria-Bertani broth supplemented with 0.2% glucose. In comparison, cultures grown in two media based on AFEX-pretreated corn stover generated tenfold less FFA than cultures grown in algae-based media. This study demonstrates that macroalgal cellulose is a potential carbon source for the production of biofuels or other microbially synthesized compounds.  相似文献

The ability of plants to increase their net CO2 assimilation rate in response to increased irradiance is due to morphological and physiological changes, which might be related to their shade tolerance and leaf ontogeny, but few studies have considered morphology and physiology. Two sympatric oak species (the shade-tolerant Q. petraea and the comparatively shade-intolerant Q. pyrenaica) were grown in hydroponic solution in low-light (LL) and high-light (HL) conditions. 5 months after leaf expansion under these conditions, half of the LL plants were transferred to high light (TLH). Transfer of Q. pyrenaica, from low- to high light led to photoinhibition and after 21 days in higher light there was little acclimation of the maximum rate of carboxylation (VCmax) or the maximum rate of electron transport (Jmax). Q. pyrenaica TLH plants showed lower stomatal conductance at all times compared to plants growing in LL. Stomatal closure was the main limitation to photosynthesis after transfer in Q. pyrenaica. The increase in evaporative demand upon TLH did not affect hydraulic conductivity of Q. pyrenaica. In contrast, the more shade-tolerant Q. petraea showed a greater degree of acclimation of gas exchange in TLH than Q. pyrenaica and two weeks after transfer gas-exchange rates were as high as in LL plants. In Q. petraea, the most important changes occurred at the level of leaf biochemistry with significant increase in VCmax that decreased the Jmax/VCmax ratio below values recorded in HL plants. However, this potential increase in photosynthesis was at least partially hamstrung by a decrease in internal conductance, which highlights the importance of internal conductance in acclimation to higher light in mature leaves. Neither oak species reached the photosynthetic rates of HL plants; however a trend towards leaf acclimation was observed in Q. petraea while the transfer was harmful to the leaves of Q. pyrenaica developed in the shade.  相似文献

Upper extremity musculoskeletal disorders represent an important health issue across all industry sectors; as such, the need exists to develop models of the hand that provide comprehensive biomechanics during occupational tasks. Previous optical motion capture studies used a single marker on the dorsal aspect of finger joints, allowing calculation of one and two degree-of-freedom (DOF) joint angles; additional algorithms were needed to define joint centers and the palmar surface of fingers. We developed a 6DOF model (6DHand) to obtain unconstrained kinematics of finger segments, modeled as frusta of right circular cones that approximate the palmar surface. To evaluate kinematic performance, twenty subjects gripped a cylindrical handle as a surrogate for a powered hand tool. We hypothesized that accessory motions (metacarpophalangeal pronation/supination; proximal and distal interphalangeal radial/ulnar deviation and pronation/supination; all joint translations) would be small (less than 5° rotations, less than 2mm translations) if segment anatomical reference frames were aligned correctly, and skin movement artifacts were negligible. For the gripping task, 93 of 112 accessory motions were small by our definition, suggesting this 6DOF approach appropriately models joints of the fingers. Metacarpophalangeal supination was larger than expected (approximately 10°), and may be adjusted through local reference frame optimization procedures previously developed for knee kinematics in gait analysis. Proximal translations at the metacarpophalangeal joints (approximately 10mm) were explained by skin movement across the metacarpals, but would not corrupt inverse dynamics calculated for the phalanges. We assessed performance in this study; a more rigorous validation would likely require medical imaging.  相似文献

Until recently, facultative automictic parthenogenesis within the squamate reptiles exhibiting ZZ:ZW genetic sex determination has resulted in single reproductive events producing male (ZZ) or female (ZW) offspring. With the recent discovery of viable parthenogenetically produced female (WW) Boa constrictors, the existence of further parthenogenetic events resulting in WW females was questioned. Here, we provide genetic evidence for consecutive virgin births by a female Colombian rainbow boa (Epicrates maurus), resulting in the production of WW females likely through terminal fusion automixis. Samples were screened at 22 microsatellite loci with 12 amplifying unambiguous products. Of these, maternal heterozygosity was observed in 4, with the offspring differentially homozygous at each locus. This study documents the first record of parthenogenesis within the genus Epicrates, a second within the serpent lineage Boidae, and the third genetically confirmed case of consecutive virgin births of viable offspring within any vertebrate lineage. Unlike the recent record in Boa constrictors, the female described here was isolated from conspecifics from birth, demonstrating that males are not required to stimulate parthenogenetic reproduction in this species and possibly other Boas.  相似文献

The extent and causes of crucian carp Carassius carassius decline were assessed during an initial study of c. 25 ponds in north Norfolk, eastern England, U.K., which was then replicated (a validation study) on another c. 25 ponds in an adjacent area. Of these ponds, c. 40 are known to have contained C. carassius during the 1970s-1980s. In the initial and validation studies, C. carassius were found in only 11 of these ponds, yielding declines of 76% (five of 21 ponds) and 68% (six of 19 ponds), respectively (72% decline overall). Non-native cyprinids, including goldfish Carassius auratus and common carp Cyprinus carpio and their hybrids with C. carassius, were observed in 20% of the ponds. Causes of C. carassius local extinction from 21 ponds were confidently determined as desiccation due to drought, terrestrialization and habitat deterioration, hybridization and competition with non-native cyprinids, agricultural land reclamation and predation (after the introduction of pike Esox lucius). This study led to C. carassius being designated as a Biodiversity Action Plan (BAP) species in the county of Norfolk, the first formal conservation designation for the species in the U.K. The C. carassius BAP plan aims to halt the decline of this much overlooked species through reintroductions and selective stocking of suitable ponds within the native range of the species.  相似文献
