Abstract: Senescent squirrel monkey is a valuable model to study pathogenesis of cerebrovascular amyloid angiopathy (CAA). Cerebrovascular sequestration and blood-brain barrier (BBB) permeability to 125I-amyloid β(1-40) synthetic peptide (sAβ1-40) were studied in adult versus aged squirrel monkey 1 h after a single intravenous injection. In aged monkey, the half-time of elimination of sAβ1-40, t e1/2, was prolonged by 0.6 h, the systemic clearance, Cl SS, was reduced from 1.8 to 1.1 ml/min/kg, and the mean residence time of intact peptide in the circulation was increased by 1 h (45%). In adult monkey, cerebrovascular sequestration of intact sAβ1-40 was significant, and the BBB permeability was 18.6-fold higher than for inulin. In aged monkey, the sequestration of intact sAβ1-40 by cortical and leptomeningeal microvessels and the BBB permeability were increased by 5.9, 1.8-, and 2.1-fold, respectively, in the presence of an unchanged barrier to inulin. In brain parenchyma of aged animals, 76.1% of circulating sAβ1-40 remained intact versus 45.7% in adult. We conclude that multiple age-related systemic effects, i.e., reduced body elimination and systemic clearance of sAβ1-40, and reduced peripheral metabolism, may act in concert with BBB mechanisms, i.e., increased transendothelial transport and microvascular accumulation of blood-borne sAβ1-40, and reduced brain metabolism to enhance the development of CAA.  相似文献

BACKGROUND: The rationale for rubella vaccination in the general population and for screening for rubella in pregnant women is the prevention of congenital rubella syndrome. The objective of this study was to evaluate the effectiveness of the prenatal rubella screening program in Quebec. METHODS: A historical cross-sectional study was designed. Sixteen hospitals with obstetric services were randomly selected, 8 from among the 35 "large" hospitals in the province (500 or more live births/year) and 8 from among the 50 "small" hospitals (fewer than 500 live births/year). A total of 2551 women were randomly selected from all mothers of infants born between Apr. 1, 1993, and Mar. 31, 1994, by means of stratified 2-stage sampling. The proportions of women screened and vaccinated were ascertained from information obtained from the hospital chart, the physician''s office and the patient. RESULTS: The overall (adjusted) screening rate was 94.0%. The rates were significantly different between large and small hospitals (94.4% v. 89.6%). Five large hospitals and one small hospital had rates above 95.0%. The likelihood of not having been screened was statistically significantly higher for women who had been pregnant previously than for women pregnant for the first time (4.8% v. 1.4%; p < 0.001). Of the 200 women who were seronegative at the time of screening (8.4%), 79 had been vaccinated postpartum, had a positive serological result on subsequent testing or did not require vaccination, and 59 had not been vaccinated postpartum; for 62, subsequent vaccination status was unknown. INTERPRETATION: Continued improvement in screening practices is needed, especially in small hospitals. Because vaccination rates are unacceptably low, it is crucial that steps be taken to address this issue.  相似文献

Experimental ovarian carcinogenesis has been investigated in inbred and hybrid strains of mice and induced by a diversity of mechanisms including X-irradiation, oocytotoxic xenobiotic chemicals, ovarian grafting to ectopic or orthotopic sites, neonatal thymectomy, mutant genes reducing germ cell populations, and aging. The mechanisms are briefly reviewed whereby disruptions in the function of graafian follicles results in a spectrum of ovarian proliferative lesions including tumors. The findings in mutant mice support the concept of a secondary (hormonally-mediated) mechanism of ovarian carcinogenesis in mice associated with sterility. Multiple pathogenetic factors that either destroy or diminish the numbers of graafian follicles in the ovary result in decreased sex hormone secretion (especially estradiol-17β) leading to a compensatory over-production of pituitary gonadotrophins (particularly luteinizing hormone), which places the mouse ovary at an increased risk to develop tumors. The intense proliferation of ovarian surface epithelium and stromal (interstitial) cells with the development of unique tubular adenomas in response to sterility does not appear to have a counterpart in the ovaries of women.  相似文献

Oxygen isotopic data were used to assess how far waters from the Savannah River, a major river in the Southeastern United States, backed up and inundated the wetlands along a small Coastal Plain tributary during a flood. This approach worked because the water of this tributary, Unper Three Runs, had an oxygen isotopic composition (18O = –4.9 ) distinct from Savannah River water (18O = –3.2 ).Two sample surveys were taken from the mouth of Upper Three Runs to 2 km upstream. Waters were isotopically uniform along the length of the sample transect when both river and tributary water levels were below bankfull. Visual estimates of turbidity taken when both the river and tributary overflowed their channels and inundated adjacent wetlands indicated that Savannah River water extended about 300 meters up the tributary channel. In contrast, the isotopic data indicated that a mixing zone of river and tributary waters extended about 1100 meters upstream. Although this mixing zone was documented only in the channel of Upper Three Runs, it probably extended into adjacent parts of the riparian wetland, potentially affecting ecological processes due to the differing water quality. This study was conducted in a single river-tributary system and the approach is probably applicable in other large rivers.Corresponding Editor: R. Abstract— Phosphoglyceride and fatty acid composition was determined in the cellular membranes of isolated cerebral microvessels and brain parenchymal cells (neurons and glia) taken from 10-, 20-, and 27–30-month-old C57BL6/NNIA mice. Lipids were extracted from each fraction and the fatty acid profiles of ethanolamine, cho-line, serine, and inositol phosphoglycerides analyzed by gas chromatography. The results suggest that membrane phosphoglycerides from cerebral microvessels are significantly more affected by the aging process than are those of the brain parenchyma. Relative percentage for fatty acids in cerebral microvessels indicate an overall decline in membrane unsaturation with a concomitant elevation in the level of saturation. The decline in unsaturation is reflected primarily in the loss of precursor fatty acids for arachidonic (18:2n-6 and 20:3n-6) and docosahexaenoic (20:5n-3 and 22:5n-3) acids. Levels of arachidonic (20:4n-6) and docosahexaenoic (22:6n-3) acids in each phos-phoglyceride remained unchanged with age; however, mol% for ethanolamine plasmalogen, a major source of these fatty acids, was significantly reduced in 27–30-month-old mice. Conversely, mol% for choline phospho-glyceride increased with age. The age-related changes in fatty acid profile for microvessel membrane phosphoglycerides are reflected by increased saturation/unsaturation ratios and decreased unsaturation indices. These parameters were not affected by aging in parenchymal membranes.  相似文献

Fragmented tropical forest landscapes preserve much of the remaining biodiversity and carbon stocks. Climate change is expected to intensify droughts and increase fire hazard and fire intensities, thereby causing habitat deterioration, and losses of biodiversity and carbon stock losses. Understanding the trajectories that these landscapes may follow under increased climate pressure is imperative for establishing strategies for conservation of biodiversity and ecosystem services. Here, we used a quantitative predictive modelling approach to project the spatial distribution of the aboveground biomass density (AGB) by the end of the 21st century across the Brazilian Atlantic Forest (AF) domain. To develop the models, we used the maximum entropy method with projected climate data to 2100, based on the Intergovernmental Panel on Climate Change Representative Concentration Pathway (RCP) 4.5 from the fifth Assessment Report. Our AGB models had a satisfactory performance (area under the curve > 0.75 and p value < .05). The models projected a significant increase of 8.5% in the total carbon stock. Overall, the projections indicated that 76.9% of the AF domain would have suitable climatic conditions for increasing biomass by 2100 considering the RCP 4.5 scenario, in the absence of deforestation. Of the existing forest fragments, 34.7% are projected to increase their AGB, while 2.6% are projected to have their AGB reduced by 2100. The regions likely to lose most AGB—up to 40% compared to the baseline—are found between latitudes 13° and 20° south. Overall, although climate change effects on AGB vary latitudinally for the 2071–2100 period under the RCP 4.5 scenario, our model indicates that AGB stocks can potentially increase across a large fraction of the AF. The patterns found here are recommended to be taken into consideration during the planning of restoration efforts, as part of climate change mitigation strategies in the AF and elsewhere in Brazil.  相似文献

Gastrointestinal nematode parasitism is a major burden to small ruminant production globally, compounded by increasing anthelmintic resistance. Previous studies have identified essential oils (EOs) from the Lippia genus with antiprotozoal and anthelmintic effects. Lippia dominguensis Moldenke (Ld), an endemic specie from the Dominican Republic, has similar popular uses, however, is chemically and pharmacologically yet uncharacterized. Here, we investigated the in vitro anthelmintic activity of LdEO and its ultrastructural effects on eggs and adult nematodes of Haemonchus contortus multidrug-resistant isolated. The GC/MS analysis showed linalool (33.85 %), 1,8-cineole (30.88 %), and δ-terpineol (10.61 %) as the main EO constituents. The LdEO showed an IC50=0.523 mg/mL in the egg hatch test, and the motility in the adult worm motility test was 95.8 % at 1 mg/mL. The confocal scanning laser microscopy of eggs indicated permeabilization or disruption of egg cell membranes as the possible mechanism of action of LdEO. The scanning electron microscopy of adult worms showed wrinkling, undulations, and cuticular disruptions. The LdEO displayed significant in vitro anthelmintic activity on eggs and adult worms of H. contortus. Additionally, the LdEO showed low oral toxicity in mice at 2,000 mg/kg. Thus, additional in vivo studies are justified to determine its anthelmintic efficacy in small ruminants.  相似文献

Summary The influence of temperature on blood viscosity and consequently on the potential for oxygen transport by blood was determined using a controlled flow, variable pressure tube viscometer, and blood from adult bullfrogs. Blood viscosity was determined as a function of hematocrit and temperature, and oxygen capacity was determined as a function of hematocrit. These data were used to describe 1) the potential for oxygen transport in the tube viscometer, and 2) the relation between the optimal hematocrit, the hematocrit which provided the greates oxygen transport, and temperature. The optimal hematocrit increased at a rate of 0.237% per °C increase in temperature. This value is close to the rate of change inin vivo hematocrit of 0.246 and 0.240% per °C increase in body temperature (Tb) observed in winter bullforgs acclimated to 5 and 20°C, respectively. During the summer the hematocrit ratio showed no consistent relation to Tb. These results suggest that in bullfrogs the cardiovascular adjustments to change in Tb involve the optimal hematocrit in winter, but not in summer.  相似文献

Kilham rat virus (KRV) is adsorbed into the rat nephroma cell within 1 hr after infection. There follows a latent period of about 12 hr during which less than 1% of the input infectious virus can be accounted for. New infectious virions can be detected at about 12 hr and the maximal yield of virus is attained by 23 hr after infection. The increase in final virus yield is about 200-fold over that found in the latent period. During this 23-hr period of virus growth, the rate of protein synthesis remains 75 to 100% of that in the uninfected cell. Ribonucleic acid (RNA) synthesis during this period is maintained at 100 to 150% of that found in the control cells. The addition of the inhibitor of deoxyribonucleic acid (DNA) synthesis, 5-fluoro-deoxyuridine (FUDR), up to 8 hr after infection completely suppresses virus production. After 8 hr, viral DNA production has started and FUDR inhibition progressively decreases until by 23 hr the addition of the inhibitor no longer causes a reduced virus yield. Viral DNA synthesis once initiated is required for the remainder of the 23-hr virus cycle. Viral DNA synthesis probably begins about 4 hr before the production of infectious virions. In the KRV-infected cells, DNA synthesis decreased sharply for 6 to 7 hr after infection in comparison to the uninfected cell. At 7 to 8 hr after infection, DNA synthesis in the infected cell increased and was maintained at a higher level than in the control cells for the rest of the virus growth period.  相似文献

High yields (2.3 × 105 to 1.3 × 106 protoplasts/g.f.wt.) of isolated protoplasts were obtained from cotyledons of Cirus sinensis (L.) Osb. 'Valencia'. Osmotic potential of the medium and enzyme concentrations were important in obtaining high viability of preparations as indicated by FDA fluorescence. Adding malt extract to a Murashige-Tucker basal medium increased plating efficiencies somewhat, but not the rate or duration of cell division. However, modifying the NAA and kinetin concentration optimized plating efficiencies (up to 20%) of protoplasts and also the rate or duration of cell division. The highest plating efficiency and number of cells per colony were obtained on a defined medium containing NAA (15 μ M ). and kinetin (4.6 μ M ). Coincidence of percentage protoplast viability after 13 days (assessed by FDA fluorescence) with plating efficiency after 21 days indicates that FDA fluorescence is an accurate indicator of citrus protoplast viability.  相似文献
