Spot form of net blotch (SFNB) (Pyrenophora teres f maculata) is an economically damaging foliar disease of barley in many of the world’s cereal growing areas. The development of SFNB-resistant
cultivars may be accelerated through the use of molecular markers. A screen for SFNB resistance in 96 lines identified four
new sources of resistance, including a feed variety, ‘Galleon’, for which a fully mapped doubled haploid population was available.
Segregation data indicated SFNB resistance was conferred by a single gene in the ‘Galleon’בHaruna Nijo’ cross, positioned
on the long arm of chromosome 7H. This gene is designated Rpt4 and is flanked by the RFLP loci Xpsr117(D) and Xcdo673 at distances of 6.9 cM and 25.9 cM, respectively. The marker Xpsr117(D) was validated using another population segregating for Rpt4, correctly predicting SFNB resistance with more than 90% accuracy.

The pathogenic fungus Cryptococcus neoformans delivers virulence factors such as capsule polysaccharide to the cell surface to cause disease in vertebrate hosts. In this study, we screened for mutants sensitive to the secretion inhibitor brefeldin A to identify secretory pathway components that contribute to virulence. We identified an ortholog of the cell division control protein 50 (Cdc50) family of the noncatalytic subunit of type IV P‐type ATPases (flippases) that establish phospholipid asymmetry in membranes and function in vesicle‐mediated trafficking. We found that a cdc50 mutant in C. neoformans was defective for survival in macrophages, attenuated for virulence in mice and impaired in iron acquisition. The mutant also showed increased sensitivity to drugs associated with phospholipid metabolism (cinnamycin and miltefosine), the antifungal drug fluconazole and curcumin, an iron chelator that accumulates in the endoplasmic reticulum. Cdc50 is expected to function with catalytic subunits of flippases, and we previously documented the involvement of the flippase aminophospholipid translocases (Apt1) in virulence factor delivery. A comparison of phenotypes with mutants defective in genes encoding candidate flippases (designated APT1, APT2, APT3, and APT4) revealed similarities primarily between cdc50 and apt1 suggesting a potential functional interaction. Overall, these results highlight the importance of membrane composition and homeostasis for the ability of C. neoformans to cause disease. 相似文献

Carbon monoxide releasing molecules (CORMs) deliver controlled amounts of CO to biological targets and organs. The reaction of cysteamine with triirondodecacarbonyl yields dicarbonyl bis(aminoethylthiolato)iron(II) that represents an iron-based CORM with biogenic ligands. X-ray diffraction studies at a single crystal show a cis-arrangement of the carbonyl ligands in trans-position to the amino groups with average Fe-C and C-O distances of 176.8 and 114.8 pm. The CO release is mediated by irradiation with visible light (λ > 400 nm). Physiological tests using ion channels sensitive to CO revealed the light- and time-dependent decomposition of CORM-S1 without obvious adverse effects on the cellular level. CORM-S1 is thus suitable for selective CO release and possesses a high potential for therapeutic application. 相似文献

Boron is an essential plant micro-nutrient which can be phytotoxic to plants if present in soils in high concentration. Boron
toxicity has been recognised as an important problem limiting production in the low rainfall areas of southern Australia,
West Asia and North Africa. Genetic variation for boron toxicity tolerance in wheat has been well-characterised. The efficiency
of breeding for boron toxicity tolerance could be greatly enhanced by the development of molecular markers associated with
QTLs for tolerance in wheat. A population of 161 doubled haploids from a cross between the tolerant cultivar Halberd and the
moderately sensitive cultivar Cranbrook was used to identify chromosomal regions involved in boron tolerance. A combined RFLP
and AFLP linkage map of the Cranbrook x Halberd population was used to identify chromosomal regions involved in the boron
tolerance traits measured. Regions on chromosome 7B and 7D were associated with leaf symptom expression. The region on chromosome
7B was also associated with the control of boron uptake and with a reduction in the effect of boron toxicity on root-growth
suppression. RFLP markers at the chromosome 7B and 7D loci were shown to be effective in selecting for improved boron tolerance
in an alternative genetic background. Halberd alleles at the chromosome 7B locus were associated with the concentration of
boron in whole shoots and grain. The concentration of boron in whole shoots and in grain were both related to grain yield
in a field trial conducted on soil containing toxic levels of boron. Implications relating to marker-assisted selection for
boron toxicity tolerance in wheat are discussed.

Fungicide Resistance Phenotypes of Botrytis cinerea Isolates from Commercial Vineyards in South West Germany 总被引:1,自引:0,他引:1
Fungicide resistance frequencies of Botrytis cinerea populations in the German Wine Road region were determined for 4 years. Strains showing specific resistance against carbendazim, iprodione or fenhexamid were found to occur wide‐spread, but at low frequencies. In contrast, cyprodinil resistance increased from 5.4% in 2006 to 21.9% in 2008 and 16% in 2009, and strains resistant to boscalid increased from 2% in 2006 to 26.7% in 2009. Strains with multidrug resistance (MDR) phenotypes were found at high frequencies. One of the three MDR phenotypes, MDR1, with reduced sensitivity to cyprodinil and fludioxonil, was dominating, representing 19% to 35% of the total population. Strains with a combination of cyprodinil resistance and MDR1 were found to be strongly increasing in 2008 and 2009. 相似文献

Photosynthetic efficiency and redox homeostasis are important for plant physiological processes during regular development as well as defence responses. The second‐stage juveniles of Heterodera schachtii induce syncytial feeding sites in host roots. To ascertain whether the development of syncytia alters photosynthesis and the metabolism of reactive oxygen species (ROS), chlorophyll a fluorescence measurements and antioxidant responses were studied in Arabidopsis thaliana shoots on the day of inoculation and at 3, 7 and 15 days post‐inoculation (dpi). Nematode parasitism caused an accumulation of superoxide and hydrogen peroxide molecules in the shoots of infected plants at 3 dpi, probably as a result of the observed down‐regulation of antioxidant enzymes. These changes were accompanied by an increase in RNA and lipid oxidation markers. The activities of antioxidant enzymes were found to be enhanced on infection at 7 and 15 dpi, and the content of anthocyanins was elevated from 3 dpi. The fluorescence parameter Rfd, defining plant vitality and the photosynthetic capacity of leaves, decreased by 11% only at 7 dpi, and non‐photochemical quenching (NPQ), indicating the effectiveness of photoprotection mechanisms, was about 16% lower at 3 and 7 dpi. As a result of infection, the ultrastructure of chloroplasts was changed (large starch grains and plastoglobules), and more numerous and larger peroxisomes were observed in the mesophyll cells of leaves. We postulate that the joint action of antioxidant enzymes/molecules and photochemical mechanisms leading to the maintenance of photosynthetic efficiency promotes the fine‐tuning of the infected plants to oxidative stress induced by parasitic cyst nematodes. 相似文献

A series of new di- and polyamine-caffeine analogues were synthesised and characterised by NMR, FT-IR, and MS spectroscopic methods. To access the stability of the investigated caffeine analogues, molecular dynamic simulations were performed in NAMD 2.9 assuming CHARMM36 force field. To evaluate the antioxidant capacity of new compounds, three different antioxidant assays were used, namely 1,1-diphenyl-2-picryl-hydrazyl free radical (DPPH?) scavenging activity, ferrous ions (Fe2+) chelating activity, and Fe3+→Fe2+reducing ability. In vitro, the ability of new derivatives to protect human erythrocytes against oxidative haemolysis induced by free radical from 2,2′-azobis(2-methylpropionamidine) dihydrochloride (AAPH) was estimated. The cytotoxic activity was tested using MCF-7 breast cancer cells and human erythrocytes. All compounds showed the antioxidant capacity depending mostly on their ferrous ions chelating activity. In the presence of AAPH, some derivatives were able to effectively inhibit the oxidative haemolysis. Two derivatives, namely 8-(methyl(2-(methylamino)ethyl)-amino)caffeine and 8-(methyl(3-(methylamino)propyl)amino)caffeine, showed cytotoxic activity against MCF-7 breast cancer cells but not against human erythrocytes. Therefore, it is concluded that the selected di- and polyamine caffeine analogues, depending on their chemical structure, were able to minimise the oxidative stress and to inhibit the tumour cell growth. The confirmed antioxidant and cytotoxic properties of some caffeine derivatives make them attractive for potential applications in food or pharmaceutical industries. 相似文献
