Morphological location of the sex pheromone producing area in the ovipositor of the female corn earworm Helicoverpa zea, was correlated with gas chromatographic analysis of the extracted pheromone. Histological studies showed that the pheromone gland occupied an almost complete ring of specialized columnar cells between the 8th and 9th abdominal segments. Ultrastructure of the pheromone gland cells revealed distinct features such as microvilli, pockets of granular material, intercellular canals with abundant desmosomes. Apparent changes in some of these features are associated with phases of pheromone production and non-production. Examination of the tissue with low temperature scanning electron microscopy showed the presence of excreted droplets at the tips of cuticular hairs in the glandular area during the period of pheromone production.  相似文献

Summary Cortical cells in the apical region of excised radicles from cotton (Gossypium hirsutum L. var. M-8) seeds of differing moisture content (6, 12, or 16%) were examined in thin sections of vapor-fixed tissue and in freeze-etched replicas of unfixed tissue with transmission electron microscopy (TEM). The structure of the plasma membrane, as revealed in chemically-fixed tissue, had the tripartite feature characteristic of unit membranes. Regions of the plasma membrane appearing as discontinuities or breaks were shown by stereo tilting to represent an optical artifact caused by image superposition of curved regions of the plasma membrane contained within the thickness of a thin section. In freeze-etched preparations two complementary-type images of the plasma membrane were consistently observed. The asymmetric distribution of membrane-associated particles (MAPS) in the P and E fracture faces suggests that the structure of plasma membranes in cells of dry seeds resembles that of plasma membranes in germinating seeds. The physiological significance of these findings is that leaching of ions and small molecules during seed imbibition does not appear to involve passage through disorganized membranes in the radicle.  相似文献

A population of aseptate pycnidiospores of the fungus Botryodiplodia theobromae can be induced to germinate or to form septa delimiting two cells; this developmental process is dependent upon nutritional and environmental factors. Transmission electron microscope investigations indicate that during germination of the aseptate spore, a new inner wall layer is synthesized de novo at the site of germ tube emergence. Formation of the septum also involves the de novo synthesis of an inner wall layer which comprises the majority of the septum and completely surrounds the spore. The wall of the germ tube emerging from the septate spore is a direct extension of this inner layer deposited during the formation of the septum. Although the early stages of spore germination may involve localized enzymatic degradation of the internal layers of the spore wall, transmission and scanning electron micrographs of germinating spores show that the outer wall layers are physically fractured by the emerging germ tube. It is suggested that spore germination and septum formation are initially similar processes regarding cell wall genesis but that some mechanism responsive to environmental and nutritional conditions determines the course of development.  相似文献

Summary Morphology and distribution of the relatively less well known organelles of plants have been studied with the electron microscope in tissues fixed in glutaraldehyde and postfixed in osmium tetroxide. An organelle comparable morphologically to the animal microbody and similar to the plant microbody isolated by Mollenhauer et al. (1966) has been encountered in a variety of plant species and tissues, and has been studied particularly in bean and radish roots, oat coleoptiles, and tobacco roots, stems and callus. The organelle has variable shape and is 0.5 to 1.5 in the greatest diameter. It has a single bounding membrane, a granular to fibrillar matrix of variable electron density, and an intimate association with one or two cisternae of rough endoplasmic reticulum (ER). Microbodies are easily the most common and generally distributed of the less well characterized organelles of plant cells. It seems very probable that they contain the enzymes characteristic of animal lysosomes (containing hydrolases) or animal microbodies (containing catalase and certain oxidases). Spherosomes are also possible sites of enzyme activity but are not as common or as widely distributed as microbodies. For this reason it appears likely that the particles designated as plant lysosomes, spherosomes, peroxisomes, etc., in some of the cytochemical and biochemical studies on enzyme localization will prove to be microbodies.Variations in the morphology and ER associations of microbodies in tissues of bean and radish are described and discussed. Crystal-containing bodies (CCBs) are interpreted as a specialized type of microbody characteristic of metabolically less active cells. Stages in the formation of CCBs from microbodies of typical appearance are illustrated for Avena.The general occurrence of microbodies in meristematic and differentiating cells and their close association with the ER suggest that they may play active roles in cellular metabolism. The alterations in their morphology and numbers that are observed in certain differentiating cells suggest further that the enzyme complements and metabolic roles of microbodies might change during cellular differentiation. Cultivation of Spirodela oligorrhiza ( Kurtz ) Hegelm on a sublethal dose of atrazine results in a higher linolenic to linoleic acid ratio in the thylakoid membrane lipids, less starch, more osmiophilic globules, and a reduced stroma lamellar system. Also, the grana become randomly oriented and contain more numerous and elongated lamellae. These alterations in the lipid composition and ultrastructure of the chloroplast resemble those previously observed in triazine-resistant weed biotypes and in chloroplasts developed under low light. Thylakoid membranes from atrazine-adapted plants revealed an additional high-affinity binding constant for [14C]-diuron but the number of diuron binding sites actually decreased by 20 times compared to controls. The 32,000-dalton membrane protein of the chloroplast is synthesized actively, but its breakdown appears decreased compared to control plants. The adaptive reorganization of thylakoid components may be a compensatory mechanism for maintenance of a functional interaction of the proteins and lipids of the photosystem II complex.  相似文献

The life cycle of a bacterial endoparasite of the plant-parasitic nematode Meloidogyne incognita was examined by scanning and transmission electron microscopy. The infective stage begins with the attachment of an endospore to the surface of the nematode. A germ tube then penetrates the cuticle, and mycelil colonies form in the pseudocoelom. Sporulation is initiated when terminal cells of the mycelium enlarge to form sporangia. A septum within each sporangium divides the forespore from the basal or parasporal portion of the cell. The forespore becomes enclosed by several laminar coats. The parasporal cell remains attached to the forespore and forms the parasporal microfibers. After the newly formed spores are released into the soil, these microfibers apparently enable a mature spore to attach to the nematode. These results indicate that the endoparasite is a procaryotic organism having structural features that are more common to members of Actinomycetales and to the bacterium Pasteuria ramosa than to the sporozoans or to the family Bacillaceae, as previous investigatios have concluded.  相似文献
