Research on copperII 1,10-phenanththroline (phen) derivatives continues to attract interest in the context of structure and biological properties. In this paper, two metal complexes [Cu2(phen)2(μ-Cl)2]Cl2 (1), [Zn(phen)2(H2O)Cl]Cl·4H2O (2) were synthesized and characterized. The crystal structures of 1 and 2 were determined by X-ray diffraction. In order to investigate the biological properties of the prepared complexes, spectroscopic and biological studies were performed. Results of X-ray diffraction showed that 1 and 2 form two types of crystal structures in a given system: dinuclear and mono-nuclear complex. The preliminary study on the DNA cleavage activity has shown that 1 under study behaved as the chemical nucleases. The DNA binding interaction of 1 & 2 with CT-DNA has been investigated by UV–Visible and fluorescence emission spectrometry and the apparent binding constant (K app) values are 5.1 × 104 and 1.2 × 104 M?1, respectively. In addition, fluorescence spectrometry of bovine serum albumin (BSA) with 1 & 2 showed that the quenching mechanism might be a static quenching procedure with one binding sites for BSA. In addition, the cytotoxicity of 1 in vitro on tumor cells lines (MCF-7, HepG2 and HT29) was examined by MTT and showed better antitumor effect on the tested cells.  相似文献

人表皮生长因子(hEGF), 一种由53个氨基酸残基组成的单链多肽, 具有广阔的应用前景。本文主要探讨家蚕表达人表皮生长因子gp67信号肽融合蛋白的生物活性。采用家蚕杆状病毒表达系统来表达该信号肽融合蛋白。构建了重组质粒pBacPAKS-hEGF, 将该重组质粒与线性化病毒Bm-BacPAK6 DNA共转染家蚕细胞, 筛选获得重组病毒vBacPAK-SEGF, 用vBacPAK-SEGF感染家蚕BmN细胞和五龄蚕, Western blot检测表明在家蚕细胞、五岭幼虫的血淋巴和蛹中均有约12 kD的目的蛋白表达。ELISA检测发现在家蚕细胞中的表达量为23 mg/ 106细胞, 五龄幼虫中的表达量可达到82 mg/mL血淋巴。利用小鼠成纤维细胞Balb/c3T3分析家蚕表达的hEGF信号肽融合蛋白的生物活性, 结果表明表达产物能显著促进Balb/c3T3细胞的增值。另外, 研究还发现hEGF信号肽融合蛋白可使新生ICR小鼠体重增 加, 睁眼和萌齿时间提前。本研究为进一步开发利用家蚕表达的hEGF提供理论基础。  相似文献

目的 预测与筛选结直肠癌组织特异性基因,作为靶向治疗的候选靶点.方法 利用自主开发的Python语言程序分析人类正常组织与结直肠癌组织mRNA表达的组织特异性,结合人类胚胎干细胞富集基因集以及文献挖掘结果,筛选可能的与结直肠癌发生或发展相关的基因作为候选靶点,并对其进行通路分析及基因富集分析.结果 获得了结直肠癌组织特异的且与肿瘤生物学通路密切相关的4个基因,作为进一步研究的候选靶点.结论 应用生物信息学方法从芯片数据进行挖掘,可以为结直肠癌的靶向治疗提供候选靶点,并为后续的药物设计奠定基础.  相似文献

A rapid and sensitive microarray assay for the detection of HCV-1b was developed in our laboratory and a cDNA fragment library for HCV-1b cDNA microarray probes was constructed. The full-length cDNAs of HCV-1b were digested with restriction endonuclease Sau3A I and the fragments were cloned with the pMD18-T vectors. Positive clones were isolated and identified by sequencing. The cDNA microarray was prepared by spotting the gene fragment on the surface of an amido-modified glass slide using the robotics system and samples were fluorescent labeled by the restriction display PCR (RD-PCR) technique, In the present study, modified protocols were used for probe selection and hybridization temperature. The detection of a microarray was validated by the hybridization and the sequence analysis. A total of 22 different specific gene fragments of HCV-1b ranging from 250 to 750 bp were isolated and sequenced, and these fragments were further used as probes in the microarray preparation. The diagnostic validity of the microarray method was evaluated after the washing and scanning process. The results of hybridization and sequence data analysis showed a significant specificity and sensitivity in the detection of HCV-1b RNA. The method of preparing microarray probes by construction of cDNA fragments library was effective, rapid, and simple; the optimized microarray was sensitive in the clinical detection of HCV-1b. The RD-PCR technique for the sample labeling was useful for significantly increasing the sensitivity of the assay. The cDNA microarray assay can be widely used in the clinical diagnosis of HCV-1b.  相似文献

沃尔巴克氏体(Wolbachia)作为节肢动物的胞内共生菌,可以引起宿主产生雌性化、孤雌生殖、杀雄和胞质不相容性(cytoplasmic incompatibility, CI) 4种生殖表型。其中CI是最常见的现象,表现为受感染的雄性昆虫与未感染或感染不兼容Wolbachia的雌性昆虫交配时引起胚胎死亡;而雌性感染同种Wolbachia时胚胎能够正常发育。CI是由被称为CI因子(cifA和cifB)的Wolbachia基因对调控的。其中,CifB作为毒剂在雄性中表达诱导产生CI,而CifA作为解毒剂在雌性中表达拯救CI。本文综述了CI因子结构、功能和作用机制的研究,以期为未来利用Wolbachia和CI进行蚊媒疾病和农业虫害的防控奠定基础。  相似文献

Tumor immune cell infiltration was significantly correlated with the progression and the effect of immunotherapy in cancers including esophageal carcinoma (ESCA). However, no biomarkers were identified which were associated with immune infiltration in ESCA. In the present study, a total of 128 common differentially expressed genes (DEGs) were identified between esophageal squamous cell carcinomas (ESCC) and esophageal adenocarcinomas (EAC). The results of gene ontology (GO) enrichment and Reactome pathway analysis displayed that the up-regulated DEGs were mainly involved in the regulation of extracellular matrix (ECM), while the down-regulated DEGs were mainly involved in the regulation of cornification and keratinocyte differentiation. The most significant module of up-regulated DEGs was selected by Molecular Complex Detection (MCODE). Top ten similar genes of COL1A2 were explored, then validation and the prognostic analysis of these genes displayed that COL1A2, COL1A1, COL3A1, ZNF469 and Periostin (POSTN) had the prognostic value which were up-regulated in ESCA. The expressions of COL1A2 and its four similar genes were mainly correlated with infiltrating levels of macrophages and dendritic cells (DCs) and showed strong correlations with diverse immune marker sets in ESCA. To summarize, COL1A2 and its four similar genes were identified as the potential biomarkers associated with immune infiltration in ESCA. These genes might be applied to immunotherapy for ESCA.  相似文献
