In order to investigate the effect of large isoform of ribulose-1,5-bisphosphate carboxylase/oxygenase (RuBPCO) activase (RuBPCO-A)
on photosynthesis, cDNA of the enzyme (rca) was transferred to rice cultivars (Oryza sativa f. japonica cv. Nipponbare) under the control of RuBPCO small subunit gene promoter (rbcS) via Agrobacterium tumefaciens-mediated transformation. Transgenic rice plants were identified by polymerase chain reaction (PCR) and Southern and Western
blot analyses. Net photosynthetic rate (P
N) values of the T1 transgenic lines 34 (T34) and 40 (T40) were 45.26 and 46.32 % higher than that of the control plants, respectively. At the
same time, their carboxylation efficiency and RuBPCO initial activity, quantum yield of electron transport in photosystem
2 (ΦPS2), and steady state photochemical fluorescence quenching (qP) increased. In addition, heading time of the transgenic rice was advanced. Thus increasing the amount of large isoform of

This paper reviews five of the eight species of acipenseriforms that occur in China, chiefly those of the Amur and Yangtze rivers. Kaluga Huso dauricus and Amur sturgeon Acipenser schrenckii are endemic to the Amur River. Both species still support fisheries, but stocks are declining due to overfishing. Acipenseriformes of the Yangtze River are primarily threatened by hydroelectric dams that block free passage to spawning and feeding areas. The Chinese paddlefish Psephurus gladius now is rare in the Yangtze River system, and its spawning activities were severely limited by completion of the Gezhouba Dam in 1981. Since 1988, only 3–10 adult paddlefishes per year have been found below the dam. Limited spawning still exists above the dam, but when the new Three Gorges Dam is complete, it will further threaten the paddlefish. Artificial propagation appears to be the only hope for preventing extinction of P. gladius, but it has yet to be successfully bred in captivity. Dabry's sturgeon A. dabryanus is a small, exclusively freshwater sturgeon found only in the Yangtze River system. It is concentrated today in reaches of the main stream above Gezhouba Dam. The fishery has been closed since 1983, but populations continue to decline. Acipenser dabryanus has been cultured since the 1970s, and holds promise for commercial aquaculture; availability of aquacultural methods offers hope for enhancing natural populations. The Chinese sturgeon A. sinensis occurs in the Yangtze and Pearl rivers and seas of east Asia. There is still disagreement about the taxonomy of the Pearl and Yangtze River populations. The Yangtze River population is anadromous. Adults begin spawning at about age 14 years (males) and 21 years (females), and adults spend over 15 months in the river for reproduction. Spawning sites of A. sinensis were found every year since 1982 below the Gezhouba Dam, but it seems that insufficient suitable ground is available for spawning. Since 1983, commercial fishing has been prohibited but more measures need to be taken such as establishing protected areas and characterizing critical spawning, summering and wintering habitats. 相似文献

Potato virus Y (PVY) is an important pathogen of potato (Solanum tuberosum). Although the PBS1–RPS5 immune system is well documented in Arabidopsis thaliana, it has not been reported in potato. In Arabidopsis, the bacterial effector AvrPphB cleaves AtPBS1 to trigger an immune response. Here, we show that the AvrPphB-triggered immune response is mediated by StPBS1, a close homologue of AtPBS1 in potato. However, downstream signalling of StPBS1 was mediated by unknown resistance (R) proteins other than potato orthologues of AtRPS5 and HvPBR1, which is important for HvPBS1 signalling in barley. Immune signalling of StPBS1 is mediated by the AvrPphB C-terminal cleavage domain and an STKPQ motif, in contrast to AtPBS1-mediated immunity in which both AvrPphB cleavage fragments and an SEMPH motif are essential. The cleavage sequence of AvrPphB in StPBS1 was replaced with that of the PVY NIa-Pro protease to obtain StPBS1NIa. StPBS1NIa overexpression potato displayed stronger immunity to PVY infection than did the StPBS1 transgenic lines. StPBS1NIa was cleaved at the expected target site by NIa-Pro protease from PVY. Thus, we characterized the function of StPBS1 in potato immunity and provide a biotechnology control method for PVY via transformation of decoy-engineered StPBS1NIa. 相似文献

黑叶猴是栖息于喀斯特石山地区的珍稀濒危灵长类动物,由于非法捕杀和栖息地丧失,种群数量急剧减少。2017年10-12月,我们采用样方法调查恩城国家级自然保护区及其周边地区黑叶猴的种群分布、数量及保护现状,分析其致危因素,为制定有效管理行动计划提供理论依据。本次调查共记录到黑叶猴夜宿地89处,其中有43处夜宿地下方有新鲜排泄物。结合本次调查和访问,确定黑叶猴的种群数量为18群100~105只,包括1只全白化黑叶猴成年雌性个体;其中11群分布在保护区内,7群分布在保护区外。猴群由3~12只个体组成。栖息地破碎化和退化是威胁恩城保护区黑叶猴种群生存的主要因素。 相似文献

利用脯氨酸类似物羟脯氨酸(Hyp)的生长抑制作用,筛选出抗Hyp的甘蔗(Saccha-rum sinensis Roxb)细胞变异系R932。抗性系体内游离脯氨酸含量超常积累(×3.2)。而且其体内脯氨酸合成途径中的关键酶γ-谷氨酸激酶对脯氨酸的反馈抑制较不敏感。R932抗PEG和低温能力较供体加强。实验结果表明γ-谷氨酸激酶特性的变化可能导致细胞内脯氨酸的过量积累,脯氨酸的过量积累有利于植物细胞对抗恶劣环境。 相似文献

通过野外观察并采用杂交指数(OCI)测定、花粉/胚珠比(P/O)检测、人工控制授粉等方法,对长萼兰花蕉(Orchidantha chinensis var.longisepala(D.Fang) T.L.Wu)种群的繁育系统进行了研究,采用常规石蜡切片与扫描电子显微镜(SEM)观察了柱头与"V"形黏盘的结构与形态。结果表明,长萼兰花蕉单花花期一般为18 d,依其花部形态的变化可分为蕾期、花萼未反转期、花萼反转期、唇瓣枯萎期、花萼枯萎期5个时期;根据杂交指数值为4、P/O值为253.89 ±21.09、人工异花授粉结实率分别为45%(2014年)和75%(2015年),显示出长萼兰花蕉的繁育系统属于异交,且需要传粉者。石蜡切片观察到长萼兰花蕉黏盘区与柱头可授区之间是光滑的表皮细胞,结合人工授粉实验与分泌物含糖量测定结果表明,长萼兰花蕉的"V"形黏盘不具有可授性,其作用可能是分泌黏液附着在传粉者背部使其便于携带花粉。长萼兰花蕉整个花期环境湿冷、多雨且开花同步性较低,这些因素很可能造成其有效传粉媒介缺乏,影响了传粉成功;另一方面,长萼兰花蕉有性繁殖受到限制,其主要通过根状茎进行无性繁殖后代,所以分布范围比较狭窄。 相似文献

Brassica rapa L., also called NIUMA, is used empirically in Tibetan medicine for its antioxidant, anti‐inflammatory and antiradiation activities. This study explored the hepatoprotective effects of B. rapa polysaccharides (BRPs) on acute liver injury induced by carbon tetrachloride (CCl4) in mice and the underlying mechanisms. Mice were treated with CCl4 after the oral administration of BRPs (55, 110 and 220 mg/kg) or bifendate (100 mg/kg) for 7 days. Blood and liver samples of mice were collected for analysis after 24 h. The ALP, ALT and AST levels and the biological activities of SOD, MDA and GSH?Px were measured. Histopathological changes in the liver were determined through hematoxylin and eosin staining. Moreover, TNF‐α, IL‐1β and IL‐6 expression levels were detected by commercial reagent kits. Finally, Western blot analysis was used to check the relative expression levels of caspase‐3, p‐JAK2 and p‐STAT3. The BRP pre‐treatment significantly decreased the enzymatic activities of ALT, ALP and AST in the serum, markedly increased the activities of SOD and GSH?Px in the liver and reduced the MDA concentration in the liver. BRPs alleviated hepatocyte injury and markedly inhibited the expression of TNF‐α, IL‐1β and IL‐6, also downregulating the CCl4‐induced hepatic tissue expression of caspase‐3. Furthermore, BRPs inhibited the JAK2/STAT3 signaling pathway in a dose‐dependent manner in the liver. This study demonstrated that BRPs exert hepatoprotective effect against the CCl4‐induced liver injury via modulating the apoptotic and inflammatory responses and downregulating the JAK2/STAT3 signaling pathway. Photosystem I (PSI) is one of the two photosystems in photosynthesis, and performs a series of electron transfer reactions leading to the reduction of ferredoxin. In higher plants, PSI is surrounded by four light-harvesting complex I (LHCI) subunits, which harvest and transfer energy efficiently to the PSI core. The crystal structure of PSI-LHCI supercomplex has been analyzed up to 2.6 Å resolution, providing much information on the arrangement of proteins and cofactors in this complicated supercomplex. Here we have optimized crystallization conditions, and analyzed the crystal structure of PSI-LHCI at 2.4 Å resolution. Our structure showed some shift of the LHCI, especially the Lhca4 subunit, away from the PSI core, suggesting the indirect connection and inefficiency of energy transfer from this Lhca subunit to the PSI core. We identified five new lipids in the structure, most of them are located in the gap region between the Lhca subunits and the PSI core. These lipid molecules may play important roles in binding of the Lhca subunits to the core, as well as in the assembly of the supercomplex. The present results thus provide novel information for the elucidation of the mechanisms for the light-energy harvesting, transfer and assembly of this supercomplex. 相似文献
