Six to twenty-eight months after transplanting, the net photosynthetic rate (PN) of Miscanthus leaves was measured at leaf temperatures between 18 to 37 °C. PN of clones from high mountain areas was more adaptable to low temperature, while that of clones from low mountain areas was more adaptable to high temperature. The clones from the lowlands were best adapted to both high and low temperatures. These characteristics lasted at least 28 months after transplanting. Thus Miscanthus had differentiated into different ecotypes to adapt to the thermal environments of different elevations. Comparison of the PN values measured in different seasons and durations after transplanting indicated that PN in Miscanthus could acclimate to environments with various temperature ranges resulting from elevation and seasonal changes. The prevalence of diabetes has increased dramatically over the past three decades, and currently, China has the largest number of diabetics worldwide; this number continues to grow and puts ongoing strains on the medical resources. In this review, we reviewed the diabetes research conducted in China from 1995 to 2015 with the aim of providing new insights regarding the current status and future perspectives for researchers, diabetes health providers, and respective policy-makers. Remarkable progress has been made in diabetes research in China during the past two decades in terms of both the quantity and publication influence. The progress, however, struggles to adequately manage diabetes in China. Here we addressed opportunities to strengthen researches, including new drug development, high quality studies on health economics, and healthcare quality improvement studies. As the expected wave of diabetic complications is upcoming and overwhelming, we therefore recommend that immediate improvements are required to implement the researches regarding their prevention and treatment.  相似文献

We present a two-stage hybrid-resolution approach for rigid-body protein-protein docking. The first stage is carried out at low-resolution (15°) angular sampling. In the second stage, we sample promising regions from the first stage at a higher resolution of 6°. The hybrid-resolution approach produces the same results as a 6° uniform sampling docking run, but uses only 17% of the computational time. We also show that the angular distance can be used successfully in clustering and pruning algorithms, as well as the characterization of energy funnels. Traditionally the root-mean-square-distance is used in these algorithms, but the evaluation is computationally expensive as it depends on both the rotational and translational parameters of the docking solutions. In contrast, the angular distances only depend on the rotational parameters, which are generally fixed for all docking runs. Hence the angular distances can be pre-computed, and do not add computational time to the post-processing of rigid-body docking results.  相似文献

The theory of the second-order reversible reaction, A + B<-->A.B, has been extensively discussed. Apparent first-order behavior is observed when, for example, [B] > [A]. If the reaction exhibits second-order behavior then it is presumed that the concentrations of A and B were initially equal and that they remain equal during the reaction. However, in the case of hydrogen peroxide binding to cytochrome c oxidase, Weng & Baker (1991, Biochemistry 30, 5727-5733) showed that the observed rate was rigorously first order over a broad concentration range of ligand, including the stoichiometric case. It was further shown that kobs increased linearly with [H2O2], precluding the possibility of a rate-limiting, unimolecular pre-step. The current work examines the theoretical rate equation for the bimolecular, reversible reaction when [A] = [B]. Simulations show that this equimolar condition resulted in rigorous exponential binding as kd, the equilibrium dissociation constant for the A.B complex, approached the initial concentration of A (or B). In particular, the second-order simulation was rigorously exponential when [A]o/Kd = 0.5, and showed only minor deviations when the ratio was increased to 25. These results demonstrate that a reversible, bimolecular reaction can appear first order even under second order conditions, without the need for more complicated mechanisms.  相似文献

Okara (soybean residue), a by-product from soymilk and tofu production, has a green, grassy off odour as it contains a large amount of aldehydes. This work investigated the rate-limiting enzyme(s) in the formation of aldehydes in okara and the pathways leading to their bioconversion into fruity, pleasant-smelling esters by the yeast Lindnera saturnus. Lipase and hydroperoxide lyase were shown to be rate-limiting enzymes while endogenous soy lipoxygenase was also crucial for the production of aldehydes in okara. Subsequent fermentation of okara by L. saturnus increased the amount of esters by about 70 times to 165–277 μg/g dried okara. The generation of C7 esters followed our hypothesised pathway, while that for C6 esters was mainly affected by L. saturnus. This study presents a simple and inexpensive one-pot setup for the natural bio-production of esters from okara.

Saporin, a type I ribosome-inactivating protein (RIP), removes adenine residues from the 28S ribosomal RNA as part of a process that leads to inhibition of protein synthesis. However, as shown in this study, neither saporin nor his-tagged saporin (both 0.6-6 pM) exert toxicity on several human cell lines including H-2171, SK-N-SH, HEP-G2, MOLT-3, THP-1, HL-60 and ECV-304. Saporin and his-tagged saporin became highly cytotoxic when they were used in a combined treatment with Soapwort saponins (SA). When combined with SA (2-4 microg/ml) saporin became as cytotoxic as the highly toxic type II RIP rViscumin reflected by an IC50 of 42.5x10(-12) M for saporin and 21.5x10(-12) M for rViscumin. We demonstrated that saporin was internalized via clathrin-mediated endocytosis, followed by the release into the endosomal transport system. Our results indicate that SA triggers this endocytic event rendering the otherwise cell membrane impermeable type I RIP saporin a potent cytotoxin. This effect was not cell line-specific suggesting that saporin exploits a common SA-dependent mechanism to enter cells.  相似文献

Ganoderma lucidum, a medicinal fungus is thought to possess and enhance a variety of human immune functions. An immuno-modulatory protein, Ling Zhi-8 (LZ-8) isolated from G. lucidum exhibited potent mitogenic effects upon human peripheral blood lymphocytes (PBL). However, LZ-8-mediated signal transduction in the regulation of interleukin-2 (IL-2) gene expression within human T cells is largely unknown. Here we cloned the LZ-8 gene of G. lucidum, and expressed the recombinant LZ-8 protein (rLZ-8) by means of a yeast Pichia pastoris protein expression system. We found that rLZ-8 induces IL-2 gene expression via the Src-family protein tyrosine kinase (PTK), via reactive oxygen species (ROS), and differential protein kinase-dependent pathways within human primary T cells and cultured Jurkat T cells. In essence, we have established the nature of the rLZ-8-mediated signal-transduction pathways, such as PTK/protein kinase C (PKC)/ROS, PTK/PLC/PKCalpha/ERK1/2, and PTK/PLC/PKCalpha/p38 pathways in the regulation of IL-2 gene expression within human T cells. Our current results of analyzing rLZ-8-mediated signal transduction in T cells might provide a potential application for rLZ-8 as a pharmacological immune-modulating agent.  相似文献
