The aqueous extract of the venom glands of black widow spiders was fractionated on a column of Sephadex G-200 and then on a column of DEAE-Sephadex A-50 pH 8.2. A protein fraction was obtained that caused a great increase in the frequency of occurrence of miniature end plate potentials at the frog neuromuscular junction, and caused swelling of the nerve terminals and depleted them of their vesicles. The fraction consists of a least four protein components that are similar in their molecular weights (about 130,000) and isoelectric points (ranging from pH 5.2 to 5.5) and are immunologically indistinguishable. It contains no sugar residues and has little or no lipolytic or proteolytic activity. The fraction is toxic to mice and is different from the fractions that act on houseflies, the crayfish stretch receptor and the cockroach heart. It seems pure enough to warrant a detailed study of its site and mode of action.  相似文献

Toll-like receptors (TLRs) have been identified as key sensors of invading microbes by identifying pathogen-associated molecular patterns and activating innate immune responses. Whereas purifying selection has been suggested in mammalian TLR9, evolutionary features of TLR9 in teleosts have not been investigated in detail. We therefore analysed TLR9 DNA sequences of eight teleost species, including zebrafish (Danio rerio), Japanese flounder (Paralichthys olivaceus), pufferfish (Takifugu rubripes), and five seabreams. Eleven sites subjected to positive selection were identified using the codon-substitution models of PAML 3.15. Ten of these 11 sites were found to be associated with leucine-rich repeats (LRRs). Seven of these 10 positively selected sites were associated with the convex surface of the LRR solenoids, leading to variations of the structures of the LRRs possibly by the introduction of flexibility into the LRR solenoids. The positive selection of LRRs in TLR9 may indicate the adaptation of teleosts to different oligodeoxynucleotides present in different bacterial species.  相似文献

Although the quinoline ring is found in a wide variety of biologically active compounds and is frequently condensed with various heterocycles, synthesis and biological evaluation of the indenoquinoline skeleton attracts only very limited attention. We report herein the synthesis and antiproliferative evaluation of certain indeno[1,2-c]quinoline derivatives against the growth of six cancer cell lines including human cervical epithelioid carcinoma (HeLa), oral squamous cell carcinoma (SAS), hepatocellular carcinoma (SKHep), human stomach adenocarcinoma (AGS), prostate cancer (PC-3), and non-small cell lung cancer (A549). The results indicated that 9-methoxy-6-(piperazin-1-yl)-11H-indeno[1,2-c]quinolin-11-one (17b) is more active than its C(6)-amino derivative 17a, C(6)-morpholine and C(6)-piperidine isomers, 17c and 17d, respectively. Treatment of 17b with NH(2)OH afforded its hydroxyimino derivative 20 which is more active than the carbonyl precursor 17b. More potent agents were obtained by further derivatization of 20. Thus, antiproliferative activities decreased in an order of aminoalkoxyimino 22a-d>hydroxyimino 20>alkoxyimino 21, 22e>carbonyl 17b. Both AGS and A549 were resistant to camptothecin with GI(50) values of 23.76 and 2.80 microM, respectively, while GI(50) values for 22a-d were in the range of 5.93-7.11 microM and 0.38-0.87 microM, respectively. Among them, 22b was the most potent with GI(50) values of 0.52, 0.74, 6.76, and 0.64 microM against the growth of HeLa, SKHep, AGS, and A549 cells, respectively. Flowcytometric analysis indicated 22c can induce cell cycle arrest in S phase, and DNA polyploidy (>4n) followed by apoptosis.  相似文献

Because all the cell populations are capable of making switches between different genetic expression states in response to the environmental change, Thattai and van Oudenaarden (Genetics 167, 523–530, 2004) have raised a very interesting question: In a constantly fluctuating environment, which type of cell population (heterogeneous or homogeneous) is fitter in the long term? This problem is very important to development and evolution biology. We thus take an extensive analysis about how the cell population evolves in a periodically switching environment either with symmetrical time-span or asymmetrical time-span. A complete picture of the phase diagrams for both cases is obtained. Furthermore, we find that the systems with time-dependent cellular transitions all collapse to the same set of dynamical equations with the modified parameters. Furthermore, we also explain in detail how the fitness problem bears much resemblance to the phenomenon, stochastic resonance, in physical sciences. Our results could be helpful for the biologists to design artificial evolution experiments and unveil the mystery of development and evolution.  相似文献

Background

Postoperative pain resulting from surgical trauma is a significant challenge for healthcare providers. Opioid analgesics are commonly used to treat postoperative pain; however, these drugs are associated with a number of undesirable side effects.

Objective

This systematic review and meta-analysis evaluated the effectiveness of acupuncture and acupuncture-related techniques in treating postoperative pain.

Data Source

MEDLINE, Cochrane Library, and EMBASE databases were searched until Sep 30, 2014.

Study Eligibility Criteria

Randomized controlled trials of adult subjects (≥ 18 years) who had undergone surgery and who had received acupuncture, electroacupuncture, or acupoint electrical stimulation for managing acute post-operative pain were included.

Results

We found that patients treated with acupuncture or related techniques had less pain and used less opioid analgesics on Day 1 after surgery compared with those treated with control (P < 0.001). Sensitivity analysis using the leave-one-out approach indicated the findings are reliable and are not dependent on any one study. In addition, no publication bias was detected. Subgroup analysis indicated that conventional acupuncture and transcutaneous electric acupoint stimulation (TEAS) were associated with less postoperative pain one day following surgery than control treatment, while electroacupuncture was similar to control (P = 0.116). TEAS was associated with significantly greater reduction in opioid analgesic use on Day 1 post surgery than control (P < 0.001); however conventional acupuncture and electroacupuncture showed no benefit in reducing opioid analgesic use compared with control (P ≥ 0.142).

Conclusion

Our findings indicate that certain modes of acupuncture improved postoperative pain on the first day after surgery and reduced opioid use. Our findings support the use of acupuncture as adjuvant therapy in treating postoperative pain.  相似文献

Neisseria meningitidis expresses a heterogeneous populationof lipooligosaccharide (LOS) inner cores variously substitutedwith 1-3-linked glucose and O-3, O-6, and O-7 linked phosphoethanolamine(PEA), as well as glycine, attached to HepII. Combinations ofthese attachments to the LOS inner core represent immunodominantepitopes that are being exploited as future vaccine candidates.Historically, each LOS immunotype was structurally assessedand prescribed a certain unique inner core epitope. We reportthat a single isolate, strain NMB, possesses the capacity toproduce all of the known neisserial LOS inner core immunotypestructures. Analysis of the inner cores from parental LOS revealedthe presence or absence of 1,3-linked glucose, O-6 and/or O-7linked PEA, in addition to glycine attached at the 7 positionof the HepII inner core. Identification and inactivation oflpt-6 in strain NMB resulted in the loss of both O-6 and O-7linked PEA groups from the LOS inner core, suggesting that Lpt-6of strain NMB may have bifunctional transferase activities orthat the O-6 linked PEA groups once attached to the inner coreundergo nonenzymatic transfer to the O-7 position of HepII.Although O-3 linked PEA was not detected in parental LOS innercores devoid of 1-3-linked glucose residues, LOS glycoformsbearing O-3 PEA groups accumulated in a truncated mutant, NMBlgtK(Hep2Kdo2-lipid A). Because these structures disappeared uponinactivation of the lpt-3 locus, strain NMB expresses a functionalO-3 PEA transferase. The LOS glycoforms expressed by NMBlgtKwere also devoid of glycine attachments, indicating that glycinewas added to the inner core after the completion of the -chainby LgtK. In conclusion, strain NMB has the capability to expressall known inner core structures, but in in vitro culture L2and L4 immunotype structures are predominantly expressed.  相似文献
