本研究试图揭示 (1)社会环境影响雌性果子狸 (Pagumalarvata)的交配日期 ,(2 )哺乳期母兽有雌二醇浓度升高的现象。通过检查是否排出交配栓并结合交配行为的发生与否确定开始交配的日期 ,使用放射免疫方法测定尿中雌二醇浓度。结果表明 (1)一雄两雌组中优势雌兽开始交配的日期显著早于劣势雌兽和单雌组雌兽 ,劣势雌兽和单雌组雌兽之间无差异 ;交配期优势雌兽尿液中雌二醇浓度明显高于劣势雌兽 ,而与单雌组中的雌兽无差异 ,但单雌组中雌兽尿液中雌二醇的浓度明显高于劣势雌兽雌二醇的浓度 ;(2 ) 5头母兽产后第二天雌二醇浓度开始上升 ,在第 5天时达到高峰 ,其中 4头母兽的雌二醇浓度共出现了两个峰值 ,其间隔为 16 75± 4 4 6 (4)天。上述结果说明 :(1)社会影响仅对优势雌性个体的繁殖有促进作用 ;(2 )哺乳期雌二醇水平的升高是季节性多次发情的基础。  相似文献

Lupus-associated anti-DNA Abs display features of Ag selection, yet the triggering Ag in the disease is unknown. We previously demonstrated that the peptide DWEYSVWLSN is bound by a pathogenic anti-DNA Ab, and that immunization of nonautoimmune mice with this peptide induces autoantibodies and renal Ig deposition. To elucidate differences in the induced B cell responses in mice genetically predisposed to autoimmunity, young (NZB x NZW)F(1) mice were immunized with this peptide DNA mimetope. DWEYSVWLSN-immunized mice had significantly increased IgG anti-dsDNA, anti-laminin, and anti-cardiolipin Ab titers compared with controls. In addition, glomerular histopathology in the form of endocapillary disease and crescent formation was markedly more severe in DWEYSVWLSN-immunized mice. Analysis of mAbs from DWEYSVWLSN-immunized mice revealed that anti-peptide Abs were often cross-reactive with DNA. Genetic elements used in the Ab response in immunized mice were homologous to those used in the spontaneous anti-DNA response in (NZB x NZW)F(1) mice, as well as in other, experimentally induced anti-DNA Abs. Our results indicate that peptide immunization can induce a molecular genetic response common to a variety of stimuli that break tolerance to mammalian dsDNA. Based on the similarity between spontaneously arising anti-DNA Abs and several types of induced anti-DNA Abs, we suggest that there may be more than a single Ag that can trigger systemic lupus erythematosus.  相似文献

Reverse gyrase is a hyperthermophilic enzyme that can introduce positive supercoiling in substrate DNA. It is showed in our studies that positive DNA supercoils were induced in both pBR322 vector and an artificially synthesized mini-plasmid DNA by reverse gyrase. The left-handed structures adopted by positively supercoiled DNA molecules could be identified from their right-handed topoisomers through atomic force microscopic examination. Additional structural comparisons revealed that positively supercoiled DNA molecule AFM images exhibited increased contour lengths. Moreover, enzymatic assays showed that the positively supercoiled DNA could not be cleaved by T7 endonuclease. Together, this suggests that the overwound structure of positive supercoils could prevent genomic duplex DNA from randomly forming single-stranded DNA regions and intra-stranded secondary structures.  相似文献

圆偏振光在地球环境中是很少见的, 来自珠宝金龟甲体壁反射圆偏振光是稀有的自然资源之一。铜绿丽金龟Anomala corpulenta Motschulsky与其他珠宝金龟甲一样其体壁能够反射左旋圆偏振光。为了解铜绿丽金龟对圆偏振光的感知能力, 本研究利用室内行为、 田间诱集与视网膜电位(electroretinogram, ERG)等方法研究了铜绿丽金龟对圆偏振光的响应。室内行为研究结果表明, 铜绿丽金龟对左旋圆偏振光与右旋圆偏振光的趋光反应明显低于非偏振光, 避光反应明显大于非偏振光, 且对左旋圆偏振光的趋、 避光反应均大于右旋圆偏振光。田间选择试验结果显示, 铜绿丽金龟对左旋和右旋圆偏振光的选择明显低于非偏振光, 且右旋大于左旋圆偏振光。电生理学研究结果表明, 左旋和右旋圆偏振光与非偏振光一样, 均能引发铜绿丽金龟的ERG反应, 且无明显差异。这些研究结果表明铜绿丽金龟体壁能够反射产生左旋圆偏振光, 且能够感知和区分左旋和右旋圆偏振光, 说明铜绿丽金龟存在偏振视觉, 具有圆偏振光敏感性。  相似文献

将拟南芥BAK1基因采用Gateway方法连接到植物表达载体,通过侵花粉管进行转化,从基因和蛋白表达水平检测转化是否成功。以不同BAK1表达水平植株作为试验材料,分析BAK1在芜菁缩叶病毒(Turnip crinkle virus,TCV)-拟南芥(Col-0)亲和互作系统中对植株防御的影响。结果显示,在接种TCV后,BAK1缺陷型植株对TCV较为感病,衰老相关基因表达水平增加,表明BAK1能够增强宿主对病毒的防御作用。  相似文献

目的:探讨丙泊酚靶控输注麻醉维持对老年胃癌根治术患者血流动力学以及术后苏醒质量的影响.方法:ASAI~Ⅱ级行胃癌根治术的60例患者随机分为A组和B组.两组使用靶控输注麻醉诱导,起始血浆靶浓度为1μg·ml-1 1%丙泊酚泵注,A组采用靶控输注丙泊酚维持麻醉,B组采用七氟烷吸入维持麻醉.记录麻醉前(T0)、切皮后5min(T1)、手术开始30 rmin(T2)、手术开始60min(T3)、停药时(T4)、出室时(T5)各时间点的血流动力学指标的变化.记录患者术后睁眼时间、拔管时间、定向力恢复时间.结果:两组血流动力学指标(SBP、DBP、HR)在T1~T3时点迅速下降,与T0比较均有显著性差异(P<0.05),T4时血流动力学指标(SBP、DBP、HR)开始上升,至T5时基本达到基线水平,与T0比较差异无统计学意义(P>0.05).血流动力学指标(SBP、DBP、HR)在组间比较差异均无统计学意义(P>0.05).两组术后睁眼时间比较差异无统计学意义(P>0.05),A组术后拔管时间、定向力恢复时间均低于B组,相比较有显著性差异(P<0.05).结论:丙泊酚靶控输注麻醉维持对老年胃癌根治术患者血流动力学影响小,患者术后苏醒质量高,是老年胃癌根治术患者理想的麻醉方法.  相似文献

miRNAs have recently been shown to play a significant role in human aging. However, data demonstrating the effects of aging‐related miRNAs in human mesenchymal stem cells (hMSCs) are limited. We observed that hMSC differentiation decreased with aging. We also identified that miR‐10a expression was significantly decreased with age by comparing the miRNA expression of hMSCs derived from young and aged individuals. Therefore, we hypothesized that the downregulation of miR‐10a may be associated with the decreased differentiation capability of hMSCs from aged individuals. Lentiviral constructs were used to up‐ or downregulate miR‐10a in young and old hMSCs. Upregulation of miR‐10a resulted in increased differentiation to adipogenic, osteogenic, and chondrogenic lineages and in reduced cell senescence. Conversely, downregulation of miR‐10a resulted in decreased cell differentiation and increased cell senescence. A chimeric luciferase reporter system was generated, tagged with the full‐length 3′‐UTR region of KLF4 harboring the seed‐matched sequence with or without four nucleotide mutations. These constructs were cotransfected with the miR‐10a mimic into cells. The luciferase activity was significantly repressed by the miR‐10a mimic, proving the direct binding of miR‐10a to the 3′‐UTR of KLF4. Direct suppression of KLF4 in aged hMSCs increased cell differentiation and decreased cell senescence. In conclusion, miR‐10a restores the differentiation capability of aged hMSCs through repression of KLF4. Binary NiFe layer double hydroxide (LDH) serves as a benchmark non‐noble metal electrocatalyst for the oxygen evolution reaction, however, it still needs a relatively high overpotential to achieve the threshold current density. Herein the catalyst's electronic structure is tuned by doping vanadium ions into the NiFe LDHs laminate forming ternary NiFeV LDHs to reduce the onset potential, achieving unprecedentedly efficient electrocatalysis for water oxidation. Only 1.42 V (vs reversible hydrogen electrode (RHE), ≈195 mV overpotential) is required to achieve catalytic current density of 20 mA cm?2 with a small Tafel slope of 42 mV dec?1 in 1 m KOH solution, which manifests the best of NiFe‐based catalysts reported till now. Electrochemical analysis and density functional theory +U simulation indicate that the high catalytic activity of NiFeV LDHs mainly attributes to the vanadium doping which can modify the electronic structure and narrow the bandgap thereby bring enhanced conductivity, facile electron transfer, and abundant active sites.  相似文献
